Estimation of genetic distance among genotypes of caraway (Carum carvi L.) using RAPD-PCR Agronomy
Seidler-Łożykowska, Katarzyna; Institute of Natural Fibres and Medicinal Plants; Kuczyńska, Anetta; Polish Academy of Sciences; Mikołajczyk, Katarzyna; Plant Breeding and Acclimatization Institute; Nowakowska, Joanna; Plant Breeding and Acclimatization Institute; Bocianowski, Jan; Poznań University of Life Sciences.
In order to estimate genetic diversity among starting materials and breeding strains of caraway, a collection of 17 accessions from botanical gardens in Europe, two cultivars ‘Rekord’ and ‘Kończewicki’, and four own breeding strains were analyzed by RAPD-PCR. The representative samples, each of five individual plants of accession, cultivar or strain, were taken from young rosette leaves. Forty of Genset Oligos RAPD primers were used for analysis and eight of them produced clear and reproducible banding patterns. In total, 62 banding patterns were obtained revealing 23 polymorphic bands, whereas the number of polymorphic bands ranged from two to four for one primer. The GS12 and GS43 primers generated two polymorphic bands, while each of the GS8, GS21,...

Genetic variability and evidence of founder effect in Hemiodus orthonops (Characiformes: Hemiodontidae) from the upper Paraná River basin, Brazil Biological Sciences
Marques, Liégie Alher; Renesto, Erasmo.
Hemiodus orthonops is a small fish of the Hemiodontidae family, order Characiformes, with a maximum of 25 cm standard length. Until recently, H. orthonops was an endemic species from the Paraná-Paraguay basin and it was absent from the upper Paraná River basin. Since 2008, it has started to be collected in the upper Paraná River, representing up to 10% of catches. Two population samples of H. orthonops from two localities of the upper Parana River basin (Porto Camargo and Porto Figueira) were analyzed using the allozymes electrophoresis technique. Twenty-one enzymatic loci were detected. The population sample from Porto Camargo displayed a genetic variability (He = 0.1061) higher than that from Porto Figueira (He = 0.0580) and homozygote excess in both of...

Genetic diversity, population structure and AFLP markers associated with maize reaction to southern rust Bragantia
Giordani,Willian; Scapim,Carlos Alberto; Ruas,Paulo Maurício; Ruas,Claudete de Fátima; Contreras-Soto,Rodrigo; Coan,Marlon; Fonseca,Inês Cristina de Batista; Gonçalves,Leandro Simões Azeredo.
ABSTRACT Maize is one of the species with greater genetic diversity among cereals and possibly the most diverse crop species known. Accessing this variability is essential for maize breeding, allowing breeders to achieve progress of yield increasing, overcome environmental challenges or deal with pests and diseases. Among the maize diseases, southern rust is one of the most important, causing significant losses in yield and presenting severe epidemics worldwide. In the present study, the AFLP technique was applied to analyze population structure and genetic diversity among 145 tropical maize inbred lines, and to test for preliminary evidence of association between AFLP markers and the reaction to southern rust. Disease severity was evaluated in two crop...

Genetic divergence among processing tomato hybrids and formation of new segregating populations Ciência e Agrotecnologia
Figueiredo,Alex Sandro Torre; Resende,Juliano Tadeu Vilela de; Schwarz,Kélin; Marodin,Josué Clock; Galvão,Alexandre Gonçalves; Resende,Nathalia Campos Vilela.
ABSTRACT Tomato is the most important vegetable species and has a strong bottleneck effect in its domestication and evolution. In exploring the existing genetic variability in commercial germplasm, germplasm has been proven to be an excellent alternative to obtain inbred lines in order to provide superior new hybrids in the future. In this sense, the objective of this study was to estimate the genetic distance among commercial processing tomato hybrids via agronomical and quality postharvest fruit traits with the aim of suggesting promising crosses for the formation of base populations for tomato breeding. Ten hybrids of processing tomato were evaluated in a complete randomized block design with three replicates. In total, eleven agronomic and postharvest...

Genetic diversity in Egeria densa and E. najas in Jupiá Reservoir, Brazil Ciencia e Investigación Agraria
Mori,Edson S; Martins,Dagoberto; Velini,Edivaldo D; Marino,Celso L; Gouvêa,Cantídio F; Leite,Suzi M.M; Camacho,Edwin; Guries,Raymond P.
The aquatic plant species Egeria densa (Planch.) Casp and E. najas Planchon occur naturally in the Paraná River Basin of southeastern Brazil. Hydroelectric dam construction in the river basin has created several lakes, changing the ecology of the river and altering the population ecology of Egeria. Large, dense populations of Egeria now grow in Jupiá Lake and its tributaries, occasionally blocking hydroelectric turbines. This study is part of a larger project examining environmental changes associated with large dam projects; the research objective of this study was to assess patterns of genetic diversity in Egeria populations growing in Jupiá and Três Irmãos Lakes and their Paraná River tributaries using genetic markers. Forty-two plants of E. najas and...

Carrasco,Basilio; Díaz,Carole; Moya,Mario; Gebauer,Marlene; García-González,Rolando.
The genetic characterization of 29 elite Japanese plum cultivars (Prunus salicina) and 4 Prunus cultivars was carried out by analyzing 97 Simple Sequence Repeat (SSR) alleles and 232 binary Inter Simple Sequence Repeat (ISSR) loci. A high level of genetic variability was found for these two molecular markers among the Japanese plum cultivars compared to other Prunus species. On average, the variability found by analyzing the SSR alleles were Na = 12.1, Ne = 5.2, Ho = 0.9, He = 0.8 and F= -0.127, whereas ISSR yielded values of h = 0.15 and I = 0.27. The genetic relationship among cultivars was estimated with Principal Coordinate Analysis (PCA) and a Bayesian clustering approach using the software program Structure. This program identified two subgroups...

Oliveira,Joelliton Domingos de; Igarashi,Maria Luiza Silveira de Paiva; Machado,Théa Mírian Medeiros; Miretti,Marcos Mateo; Ferro,Jesus Aparecido; Contel,Eucleia Primo Betioli.
The genetic relationships and structure of fourteen goat (Capra hircus) populations were estimated based on genotyping data from 14 goat populations (n = 410 goats) at 13 microsatellite loci. We used analysis of molecular variance (AMOVA), principal component analysis (PCA) and F statistics (F IS, F IT and F ST) to evaluate the genetic diversity (Ho, He and ad) of the goats. Genetic distances between the 14 goat populations were calculated from allelic frequency data for the 13 microsatellite markers. Moderate differentiation was observed for the populations of the undefined breeds (including the Anglo-Nubian-M breed), the naturalized Brazilian breeds (Moxotó, Canindé), the exotic purebred breeds (Alpine, Saanen, Toggenbourg and Anglo-Nubian) and the...

Short-tandem repeat analysis in seven Chinese regional populations Genet. Mol. Biol.
Song,Xing-bo; Zhou,Yi; Ying,Bin-wu; Wang,Lan-lan; Li,Yi-song; Liu,Jian-feng; Bai,Xiao-gang; Zhang,Lei; Lu,Xiao-jun; Wang,Jun; Ye,Yuan-xin.
In the present study, we investigated the application of 13 short tandem repeat (STR) loci (D13S317, D7S820, TH01, D16S539, CSFIPO, VWA, D8S1179, TPOX, FGA, D3S1358, D21S11, D18S51 and D5S818) routinely used in forensic analysis, for delineating population relationships among seven human populations representing the two major geographic groups, namely the southern and northern Chinese. The resulting single topology revealed pronounced geographic and population partitioning, consistent with the differences in geographic location, languages and eating habits. These findings suggest that forensic STR loci might be particularly powerful tools in providing the necessary fine resolution for reconstructing recent human evolutionary history.

Skewed RAPD markers in linkage maps of Citrus Genet. Mol. Biol.
Oliveira,Roberto Pedroso de; Aguilar-Vildoso,Carlos Ivan; Cristofani,Mariângela; Machado,Marcos Antônio.
The objective of this work was to analyze the effects of RAPD markers with skewed segregation on genetic linkage maps. Segregation data for 123 Citrus sinensis (L.) Osbeck cv. Pêra markers and 53 C. reticulata Blanco cv. Cravo markers in F1 progeny composed of 94 hybrids were used. Genetic linkage maps of the two varieties were constructed with non-skewed markers (p < 0.05 and p < 0.01) using the program MAPMAKER 3.0 and a pseudo-testcross strategy. The maps were compared to those constructed with all markers. Alterations in the genetic distances were observed based on the location of the skewed markers within the linkage groups. Generally, the skewed markers were located at the end of the linkage groups, sometimes forming entire linkage groups,...

Predicting performance of soybean populations using genetic distances estimated with RAPD markers Genet. Mol. Biol.
Barroso,Paulo Augusto Vianna; Geraldi,Isaias Olívio; Vieira,Maria Lúcia Carneiro; Pulcinelli,Carlos Eduardo; Vencovsky,Roland; Dias,Carlos Tadeu dos Santos.
In order to verify whether genetic distance (GD) is associated with population mean (PM), genetic variance (GV) and the proportion of superior progenies generated by each cross in advanced generations of selfing (PS), the genetic distances between eight soybean lines (five adapted and three non-adapted) were estimated using 213 polymorphic RAPD markers. The genetic distances were partitioned according to Griffing's Model I Method 4 for diallel analysis, i.e., GDij = GD+ GGDi+ GGDj + SGDij. Phenotypic data were recorded for seed yield and plant height for 25 out of 28 populations of a diallel set derived from the eight soybean lines and evaluated from F2:8 to F2:11 generations. No significant correlation for seed yield was detected between GD and GV, while...

Genetic diversity between herds of Alpine and Saanen dairy goats and the naturalized Brazilian Moxotó breed Genet. Mol. Biol.
Araújo,Adriana Mello de; Guimarães,Simone Eliza Facioni; Machado,Thea Mírian Medeiros; Lopes,Paulo Sávio; Pereira,Carmen Silva; Silva,Francisco Luiz Ribeiro da; Rodrigues,Marcelo Teixeira; Columbiano,Virgínia de Souza; Fonseca,Cleusa Graça da.
Brazilian naturalized goat breeds are adapted to the semiarid conditions prevalent in the Northeast region of the country (which has the largest Brazilian goat heard) and represent an as yet uninvestigated source of genetic diversity. Currently, imported goat breeds are crossed with Brazilian naturalized goat breeds, endangering the genetic potential of the naturalized breeds. We used 11 microsatellite markers to determine the genetic diversity among imported (non-naturalized) dairy Alpine and Saanen goats and naturalized Brazilian Moxotó goats. We genotyped 292 goats from three herds (one private, one from the University of Minas Gerais and the Moxotó conservation herd from Embrapa Caprinos) and found that the general heterozygosity was 0.6952 for Alpine,...

Genetics and morphology of the genus Tritetrabdella (Hirudinea, Haemadipsidae) from the mountainous rain forests of Sabah, Borneo, reveal a new species with two new subspecies Naturalis
Kappes, H..
Blood-feeding terrestrial leeches of the family Haemadipsidae are a notorious part of the invertebrate diversity in Asian and Australian rain forests. All hitherto published records of terrestrial leeches of Borneo belong to the genus Haemadipsa. Here, a second, poorly known haemadipsid genus is reported from Mount Kinabalu and Crocker Range National Park. The individuals were barcoded and compared to sequences available in GenBank. The results show that the genus Tritetrabdella has representatives in the Indochinese and the Sundaic bioregions. All six specimens from Borneo are from a single new Tritetrabdella lineage, T. kinabaluensis spec. nov. Within the Bornean lineage, two groups differing 4-5% in the COI barcoding sequence were identified. Because 1)...

Genetic variability of the Amazon River prawn Macrobrachium amazonicum (Decapoda, Caridea, Palaemonidae) Naturalis
Vergamini, F.G.; Pileggi, L.G.; Mantelatto, F.L..
The freshwater prawn Macrobrachium amazonicum is widely distributed in South America, and occupies habitats with a wide range of salinities. Several investigations have revealed the existence of wide intraspecific variability among different populations, although the understanding of this variability is still fragmentary and incomplete. We compared and characterized inland and coastal populations of M. amazonicum from Brazil, using molecular data (16S and COI mtDNA) to describe the degree of variability, structure, and relationships among them. Genetic divergence rates among populations showed variability at the intraspecific level. All the analyses evidenced significant genetic divergence among populations, structuring them in three groups: I- inland...
