Increased transient genetic transformation in immature embryos of Brazilian BR 451 maize co-cultivated with Agrobacterium tumefaciens 1
Didoné, Dielli Aparecida; Silva, Marília Rodrigues de; Ceccon, Cassia Canzi; Teixeira, Tiago; Suzin, Marilei; Grando, Magali Ferrari.
Genetic engineering has amplified the possibilities of crop breeding and supported sustainability ideals. Agrobacterium tumefaciens is the preferred transformation system, since it produces transgenic plants with more stable transgene expression and inheritance. The agrobacteria and plant tissue must be co-cultivated in conditions that allow gene transfer. This study aimed to evaluate how co-cultivation time and temperature affect the transformation of immature maize embryos of the Hi-II hybrid (model genotype) and the Brazilian BR 451 variety with A. tumefaciens EHA101:pTF102. The pTF102 plasmid carries a uidA reporter gene that enables transient transformation to be quickly verified by GUS histochemical assays. Increasing the co-cultivation period from...

Engineering biodiversity as a model for the species conservation 42
RECH,ELIBIO L..
Abstract: Early humans have domesticated plant and animal species based on ancient empirical concepts (Darwin 1868, 1876). In 1886, Mendel established a new paradigm of hereditary laws (Mendel 1866, 1870, 1950) based on genotypic and phenotypic traits of cross-compatible species, establishing a complex breeding technology that is currently utilized for the development of most food and livestock-derived products. Recently, studies on deciphering the double-helical structure (Watson and Crick 1953a, b) and how to restrict DNA (Arber 2012) have established the foundation of recombinant DNA technology. A new era is paving the way for genetic manipulation of important traits among all the kingdom’s organisms, allowing for the development of innovative and...

Cyanobacterial defense mechanisms against foreign DNA transfer and their impact on genetic engineering 48
Stucken,Karina; Koch,Robin; Dagan,Tal.
Cyanobacteria display a large diversity of cellular forms ranging from unicellular to complex multicellular filaments or aggregates. Species in the group present a wide range of metabolic characteristics including the fixation of atmospheric nitrogen, resistance to extreme environments, production of hydrogen, secondary metabolites and exopolysaccharides. These characteristics led to the growing interest in cyanobacteria across the fields of ecology, evolution, cell biology and biotechnology. The number of available cyanobacterial genome sequences has increased considerably in recent years, with more than 140 fully sequenced genomes to date. Genetic engineering of cyanobacteria is widely applied to the model unicellular strains Synechocystis sp. PCC 6803...

Genetic transformation of coffee 61
Ribas,Alessandra Ferreira; Pereira,Luiz Filipe Protasio; Vieira,Luiz Gonzaga E..
In the last 15 years, considerable advances were made in coffee genetic transformation. Different research groups in the world have been able to transform coffee with genes for insect resistance, decaffeinated coffee, herbicide resistance and control of fruit maturation. Although the majority of the research is still limited to laboratory and greenhouse studies, initial field tests with transformed coffee are beginning to appear in the literature. In this review we provide an update on the state of coffee genetic transformation, presenting technical aspects related to tissue culture systems, strategies for selection and transformation with particle bombardment, as well as the use of Agrobacterium tumefaciens. We also discuss the potential applications of...

Knowledge about and acceptance of genetically modified organisms among pre-service teachers: a comparative study of Turkey and Slovenia 69
Sorgo,Andrej; Ambrozic-Dolinsek,Jana; Usak,Muhammet; Özel,Murat.
Genetically modified organisms cannot be regarded as merely a topic for academic debate, since these have serious implications as a research field and for production based on genetic engineering. Public debates rarely base their arguments on elements rooted in scientific arguments and knowledge but are heavily loaded with emotions, opinions and informal reasoning. This study aimed to investigate the knowledge and acceptance of genetically modified organisms among prospective teachers in Slovenia and Turkey. Knowledge of genetic modification was measured with a two-tier instrument. The level of acceptance of genetic modification was measured with a 17-item instrument. Findings revealed that knowledge of genetics and biotechnology barely influenced the...

Path analysis suggests phytoene accumulation is the key step limiting the carotenoid pathway in white carrot roots 74
Santos,Carlos Antonio Fernandes; Senalik,Douglas; Simon,Philipp Wallace.
Two F2 carrot (Daucus carota L.) populations (orange rooted Brasilia x very dark orange rooted High Carotene Mass - HCM cross and the dark orange rooted cultivated variety B493 x white rooted wild carrot Queen Anne's Lace - QAL cross) with very unrelated genetic backgrounds were used to investigate intrinsic factors limiting carotenoid accumulation in carrots by applying phenotypic correlation and path analysis to study the relationships between major root carotenes, root color and several other morphological traits. Most of the correlations between traits were close and agreed in sign between the two populations. Root weight had a moderate to highly significant positive correlation with leaf length, root length and top and middle root diameter. Although...

Transgene inheritances and genetic similarities of near isogenic lines of genetically modified common beans 86
Pinheiro,Patrícia Valle; Faria,Josias Corrêa de; Nogueira,Elsa Oliveira Paranaguá e Lago; Aragão,Francisco José Lima.
The objective of the present work was to determine the inheritance and stability of transgenes of a transgenic bean line expressing the genes rep-trap-ren from Bean golden mosaic virus and the bar gene. Crosses were done between the transgenic line and four commercial bean cultivars, followed by four backcrosses to the commercial cultivars. Progenies from each cross were evaluated for the presence of the transgenes by brushing the leaves with glufosinate ammonium and by polymerase chain reaction using specific oligonucleotides. Advanced generations were rub-inoculated with an isolate of Bean common mosaic necrosis virus (BCMNV). The transgenes were inherited consistently in a Mendelian pattern in the four crosses studied. The analyzed lines recovered close...

Screening of Brazilian soybean genotypes with high potential for somatic embryogenesis and plant regeneration 86
Droste,Annette; Silva,Anelise Machado da; Souza,Iliane Freitas de; Wiebke-Strohm,Beatriz; Bücker-Neto,Lauro; Bencke,Marta; Sauner,Márcia Viviane; Bodanese-Zanettini,Maria Helena.
The aim of this work was to identify Brazilian soybean (Glycine max) genotypes with potential to respond to in vitro culture stimuli for primary somatic embryo induction, secondary embryo proliferation and plant regeneration. Differences among eight tested cultivars were observed at each stage. Two cultivars, IAS-5 and BRSMG 68 Vencedora, were selected for the evaluation of the capacity for embryo differentiation and plant regeneration. These cultivars had high embryo induction frequencies, repetitive embryogenic proliferation, and low precocious embryo germination in the initial experiment. The effect of abscisic acid (ABA) and charcoal addition on plant regeneration was investigated. The addition of ABA to proliferation medium and of ABA and activated...

Comparative analysis of the national biosafety regulatory systems in East Africa 31
Jaffe, Gregory.
This paper analyzes the current and proposed biosafety systems in Kenya, Tanzania, and Uganda using a set of components and characteristics common to functional and protective biosafety regulatory systems. It also assesses how those systems take into account the major international legal obligations that relate to biosafety, such the Cartagena Biosafety Protocol. The paper identifies certain areas in each country’s biosafety regulatory systems where further development and clarification would improve the biosafety system, making it more functional and protective. Those areas include: (1) the addition of procedures to ensure the food safety of genetically engineered organisms; (2) the inclusion of the standard and criteria for making an approval decision;...
