ABSTRACT Cryopreservation is a technique used to preserve genetic material at ultra-low temperatures in cryogenic germplasm banks. In order to enable the cryopreservation of a seed, it is necessary to study its appropriate freezing temperature and its water content, because seeds reach maximum physiological quality with high water contents, and they tolerate drying when they are orthodox seeds, but they may not tolerate cryopreservation. Jacaranda decurrens (Bignoniaceae) is an endemic species of the Cerrado with validated antitumoral activity. The genetic diversity of six populations of J. decurrens located in the State of São Paulo was determined in this study by using molecular markers for randomly amplified polymorphic DNA (RAPD) and amplified fragment length polymorphism (AFLP). Following optimization of the amplification reaction, 10 selected primers generated 78 reproducible RAPD fragments that were mostly (69.2%) polymorphic. A core collection of the common bean (Phaseolus vulgaris L.), representing genetic diversity in the entire Mexican holding, is kept at the INIFAP (Instituto Nacional de Investigaciones Forestales, Agricolas y Pecuarias, Mexico) Germplasm Bank. Visando o melhoramento genetico da sacaca, foi instalado um Banco Ativo de Germoplasma (BAG) na Embrapa Amazonia Ocidental. Estao sendo feitas coletas e multiplicacao vegetativa de acesso de sacaca de diversos locais da Amazonia. O BAG-Sacaca visa nao somente a conservacao do germoplasma, mas o estudo da variabilidade genetica entre acesso, para selecao de genotipos superiores para producao de biomassa (folhas), e com maior potencial de producao de oleos essenciais. Resumo: Esta publicação tem por objetivo descrever a história, a composição da Coleção de trabalho de Microrganismos Patogênicos a Caprinos e Ovinos, e de que forma ela tem conservado, caracterizado, documentado e utilizado seus isolados. A coleção foi criada em 2012 e atualmente é composta por 141 cepas bacterianas e seis cepas virais, pertencentes às espécies Corynebacterium pseudotuberculosis, Staphylococcus aureus e ao gênero Lentivirus. The objective of this work was to evaluate in vitro storage of Piper aduncum and P. hispidinervum under slow-growth conditions. Shoots were stored at low temperatures (10, 20 and 25°C), and the culture medium was supplemented with osmotic agents (sucrose and mannitol - at 1, 2 and 3%) and abiscisic acid - ABA (0, 0.5, 1.0, 2.0 and 3.0 mg L-1). After six-months of storage, shoots were evaluated for survival and regrowth. Low temperature at 20ºC was effective for the in vitro conservation of P. aduncum and P. hispidinervum shoots. In vitro cultures maintained at 20ºC on MS medium showed 100% survival with slow-growth shoots. The objective of this work was to evaluate ricin concentration in castor bean seeds (Ricinus communis) of 20 accessions from the Banco de Germoplasma de Mamoneira of the Embrapa Algodão, Campina Grande, PB, Brazil, using the Enzyme Linked Immunosorbent Assay. Significant differences were observed among accessions. BRA 3271 had the highest ricin concentration in seeds (32.18 ng µg-1), and BRS Paraguaçu had the lowest (3.53 ng µg-1). There is the possibility of selecting genotypes with different ricin concentrations, which can be used according on the interest of the breeding programs. |

The objective of this work was to estimate the genetic parameters and variability among accessions (half-sib families) of physic nut (Jatropha curcas) during the early stages of development. For this study, 110 accessions in the first year of development of the physic nut germplasm bank, maintained at Embrapa Cerrados, DF, Brazil, were evaluated in situ. The objective of this work was to identify sources of resistance to dry root rot induced by Fusarium sp. in cassava accessions. A macroconidial suspension (20 µL) of 11 Fusarium sp. isolates was inoculated in cassava roots, from 353 acessions plus seven commercial varieties. Ten days after inoculation, the total area colonized by the pathogen on the root pulp was evaluated by digital image analysis. Cluster analysis revealed the presence of five groups regarding resistance. The root lesion areas ranged from 18.28 to 1,096.07 mm² for the accessions BGM 1518 and BGM 556, respectively. The genotypes BGM 1042, BGM 1552, BGM 1586, BGM 1598, and BGM 1692 present the best agronomical traits. |

The purpose of this study was to characterize cultivated genotypes of three jabuticaba species (Plinia cauliflora, P. trunciflora, and P. jaboticaba). Phenology and fruit growth, as well as leaf, flower and fruit traits were evaluated. Variability in all traits was observed among genotypes of the three jabuticaba species. The trait peduncle size is indicated for differentiation of the three species under study. The leaf and fruit sizes of the genotypes P. trunciflora 3, P. trunciflora 4, P. trunciflora 5 and P. jaboticaba 1 differ from those described in the literature for these species, indicating the formation of ecotypes. Jabuticaba fruit skin contains high anthocyanin and flavonoid concentrations, with potential use in food and pharmaceutical industries. |

Breeding programs for Brachiaria spp. use both intraspecies and interspecies crosses between sexual and apomictic plants in order to obtain new cultivars with the desired characteristics. As there are different ploidy levels both within and between species of this genus, it becomes necessary to evaluate the genotypes used in breeding programs, as a guide to breeders when adopting crossing strategies. Zeyheria montana, an endemic species of the Bignoniaceae family from the Brazilian Cerrado's known for its anti-cancer properties, is widely used as imuno stimulant in the popular medicine and its therapeutic activity must be validated by scientific data. Cassava (Manihot esculenta Crantz) belongs to the Euphorbiaceae family, and is widely cultivated in Brazil. The objective of this study was to evaluate the isoenzymatic variability of 200 cassava accessions from the germplasm bank of Embrapa Amazonia Oriental. Seven groups were formed according to their origin: 1-Amazonas, 2-Amapa, 3-Bahia, 4-Para, 5-Rondonia, 6-Various, for accessions with a maximum of three individuals per place of origin, and 7 - Accessions of indefinite origin.
