Isolation of a UDP-glucose: Flavonoid 5-O-glucosyltransferase gene and expression analysis of anthocyanin biosynthetic genes in herbaceous peony (Paeonia lactiflora Pall.) Electron. J. Biotechnol.

Herbaceous peony (Paeonia lactiflora Pall.) is an excellent material for studying the formation of flower colour because of its abundant colour. The full-length cDNA of a UDP-glucose: Flavonoid 5-O-glucosyltransferase gene (UF5GT) containing 1629 bp nucleotides was obtained from P. lactiflora. The expression patterns of nine related anthocyanin biosynthetic genes (PlPSY, PlCHS, PlCHI, PlF3H, PlF3'H, PlDFR, PlANS, PlUF3GT and PlUF5GT) in diurnal variation petals showed that their expression peaks were basically at 15:00 and the expression patterns were consistent with the trend of sampling conditions except individual gene. And the highest expression levels were in PlCHS, PlDFR and PlUF3GT, which could be the candidates to regulate P. lactiflora flower...

Application of response surface methodology for glucosyltransferase production and conversion of sucrose into isomaltulose using free Erwinia sp. cells Electron. J. Biotechnol.

Isomaltulose is a structural isomer of sucrose commercially used in food industries. Glucosyltransferase produced by Erwinia sp. D12 catalyses an intramolecular transglucosylation of sucrose giving isomaltulose. The Experimental design and response surface methodology were applied for the optimization of the nutrient concentration in the culture medium for the enzyme production in shaken flasks at 200 rpm and 30ºC. The three variables involved in this study were sugar cane molasses, bacteriological peptone and yeast extract Prodex Lac SD®. The statistical analysis of the results showed that, in the range studied, all the factors had a significant effect (p < 0.05) on glucosyltransferase production and the highest enzyme activity was observed in culture...

Glucosyltransferase produced by strain Erwinia sp. is an intracellular enzyme that catalyzes the formation of isomaltulose from sucrose. Isomaltulose is a non-cariogenic reducing dissacharide commercially used in foods. Response surface methodology and 2³-factorial central composite design were employed to optimize a fermentation medium for the production of glucosyltransferase by Erwinia sp. in shaken flasks at 200 rpm and 30ºC. Glucosyltransferase production by Klebsiella sp. K18 and conversion of sucrose to palatinose using immobilized cells BJM

The strain Klebsiella sp. K18 produces the enzyme glucosyltransferase and catalyses the conversion of sucrose to palatinose, an alternative sugar that presents low cariogenicity. Response Surface Methodology was successfully employed to determine the optimal concentration of culture medium components. Maximum glucosyltransferase production (21.78 U mL-1) was achieved using the optimized medium composed by sugar cane molasses (80 g L-1), bacteriological peptone (7 g L-1) and yeast extract (20 g L-1), after 8 hours of fermentation at 28°C.
