EFFECTS OF THREE FRACTIONS OBTAINED FROM Naja haje VENOM ON HEMOLYSIS AND LIPID METABOLISM IN RABBITS J. Venom. Anim. Toxins
EL-AAL,A. A.; EZZAT,A. R..
Three fractions (F1, F2 and F3) were obtained from the venom of the Egyptian cobra Naja haje by gel filtration. F1 and F2 had indirect hemolytic effects on rabbit erythrocytes. The HU50 (the amount of the fraction causing 50% hemolysis ) values of F1 and F2 were 12.04 ± 3.89 and 36.57 ± 2.20 µg, respectively. The effects of the three cobra venom fractions on lipid metabolism were studied after 1, 2, 3 and 4 hours. Serum total lipids and cholesterol were significantly decreased at almost all tested times, but variable changes were observed in the serum triglycerides. Serum inorganic phosphorus levels were significantly increased with F1 and F2 administration more than that with the F3 fraction. These results suggest that F1 and F2 possess phospholipase...

On some toxinological aspects of the starfish Stellaster equestris (Retzius, 1805) J. Venom. Anim. Toxins incl. Trop. Dis.
Kanagarajan,U; Bragadeeswaran,S; Venkateshvaran,K.
Whole-body extracts in methanol were obtained from the starfish Stellaster equestris. The crude toxin was fractionated stepwise using diethylaminoethyl (DEAE) cellulose column chromatography. The crude toxin was lethal to male albino mice at a dose of 1.00 mL (containing 531.0 µg/mL protein) when injected intraperitoneally (IP) but the toxicity was abolished in all cases except one upon fractionation. The crude toxin and all the adsorbed fractions exhibited potent hemolytic activity on chicken, goat and human blood. However, group B human erythrocytes were resistant to lysis by all fractions and group O by most of the fractions. Paw edema in mice was caused by the crude toxin and all fractions. Pheniramine maleate and piroxicam blocked the toxicity when...

Hemolitic action of Naja naja atra cardiotoxin on erythrocytes from different animals J. Venom. Anim. Toxins incl. Trop. Dis.
Troiano,J. C.; Gould,E. G.; Gould,I..
A comparative study on the sensitivity of erythrocytes from different vertebrate species (avian, mammalian and reptilian) to the hemolytic action caused by cardiotoxin isolated from Naja naja atra venom was carried out. Cardiotoxin was able to induce direct hemolysis in washed erythrocytes from several animals, except for llama. The EC50 values from hemolysis of the most sensitive (cat) and the most resistant (snake) animal varied approximately tenfold. According to the cell behavior, it was possible to characterize four types of behavior: The first was observed in cat, horse and human cells; the second in rat, rabbit and dog erythrocytes; and the third only in llama erythrocytes, which were resistant to cardiotoxin concentrations up to 300 µg/ml. Finally,...

Wei,Shuangshuang; Chi,Tingting; Meng,Aiyun; Chen,Congwei; An,Tianchen; Wang,Manchuriga; Zhang,Yingxia.
Background The hemolytic activity of skin secretions obtained by stimulating the frog Kaloula pulchra hainana with diethyl ether was tested using human, cattle, rabbit, and chicken erythrocytes. The skin secretions had a significant concentration-dependent hemolytic effect on erythrocytes. The hemolytic activity of the skin secretions was studied in the presence of osmotic protectants (polyethylene glycols and carbohydrates), cations (Mg2+, Ca2+, Ba2+, Cu2+, and K+), or antioxidants (ascorbic acid, reduced glutathione, and cysteine). Results Depending on their molecular mass, osmotic protectants effectively inhibited hemolysis. The inhibition of skin hemolysis was observed after treatment with polyethylene glycols (1000, 3400, and 6000 Da). Among divalent...

Cytolytic and systemic toxic effects induced by the aqueous extract of the fire coral Millepora alcicornis collected in the Mexican Caribbean and detection of two types of cytolisins J. Venom. Anim. Toxins incl. Trop. Dis.
Hernández-Matehuala,Rosalina; Rojas-Molina,Alejandra; Vuelvas-Solórzano,Alma Angelica; Garcia-Arredondo,Alejandro; Ibarra Alvarado,Cesar; Olguín-López,Norma; Aguilar,Manuel.
AbstractBackground Millepora alcicornis is a branching hydrocoral common throughout the Caribbean Sea. Like other members of this genus, this species is capable of inducing skin eruptions and blisters with severe pain after contact. In the present study, we investigated the toxicity of theM. alcicornis aqueous extract on several animal models. Considering that some cnidarian hemolysins have been associated to local tissue damage, since they also induce lysis of other cell types, we also made a partial characterization of the hemolytic activity of M. alcicornis aqueous extract. This information is important for understanding the defense mechanisms of the “fire corals”.Methods The effects of pH, temperature, and some divalent cations on the hemolytic...

Acute pancreatitis and acute renal failure following multiple hornet stings J. Venom. Anim. Toxins incl. Trop. Dis.
Sharma,N.; Balamurugeshan,P. K.; Sharma,A..
Hymenoptera is a class of insects that sting in order to subdue their prey. Humans coming into accidental contact with these insects results in stings that may cause from mild local reaction like weal formation around the sting site to severe systemic reactions such as intravascular hemolysis, acute renal failure, pulmonary edema, cerebral edema, and rarely pancreatitis. We report here the clinical course of a patient who developed concurrent acute pancreatitis and pigment-induced acute renal failure after multiple hornet stings.

Hemolytic activity of venom from crown-of-thorns starfish Acanthaster planci spines J. Venom. Anim. Toxins incl. Trop. Dis.
Lee,Chi-Chiu; Tsai,Wann-Sheng; Hsieh,Hernyi Justin; Hwang,Deng-Fwu.
Background : The crown-of-thorns starfish Acanthaster planci is a venomous species from Taiwan whose venom provokes strong hemolytic activity. To understand the hemolytic properties of A. planci venom, samples were collected from A. planci spines in the Penghu Islands, dialyzed with distilled water, and lyophilized into A. planci spine venom (ASV) powder. Results : Both crude venom and ASV cause 50% hemolysis at a concentration of 20 μg/mL. The highest hemolytic activity of ASV was measured at pH 7.0-7.4; ASV-dependent hemolysis was sharply reduced when the pH was lower than 3 or greater than 8. There was almost no hemolytic activity when the Cu2+ concentration was increased to 10 mM. Furthermore, incubation at 100°C for 30 to 60 minutes sharply decreased...

Bioactivities of extracts from the marine sponge Halichondria panicea J. Venom. Anim. Toxins incl. Trop. Dis.
Purushottama,GB; Venkateshvaran,K; Pani Prasad,K; Nalini,P.
In the present study, we screened the biological activity of extracts from the marine sponge Halichondria panicea collected in the Arabian Sea. Crude toxin was obtained by methanol, chloroform-methanol (2:1) and aqueous extraction. Subsequently, the protein concentration of each crude extract was determined. The impact of both sponge methanolic and aqueous extracts was found to increase activities of Na+-K+ ATP-ase and Mg++ ATP-ase. In the case of chloroform-methanol extract, higher concentrations increased acetylcholine esterase (AchE) activity. The methanolic and chloroform-methanol extracts exhibited hemolytic activity on chicken and human erythrocytes, whereas the aqueous extract failed to do so. Methanol and aqueous extracts produced an...

Hemagglutinating/Hemolytic activities in extracts of marine invertebrates from the Brazilian coast and isolation of two lectins from the marine sponge Cliona varians and the sea cucumber Holothuria grisea Anais da ABC (AABC)
Moura,Raniere M.; Melo,Arthur A.; Carneiro,Rômulo F.; Rodrigues,Cícera R.f.; Delatorre,Plínio; Nascimento,Kyria S.; Saker-Sampaio,Silvana; Nagano,Celso S.; Cavada,Benildo S.; Sampaio,Alexandre H..
Twenty species of marine invertebrates from the Brazilian coast were screened for hemagglutinating/hemolytic activity. In at least twelve tested species, hemagglutinating activity was different for different blood types, suggesting the presence of lectins. Extracts from four species showed hemolytic activity. Two new lectins were purified from the marine sponge Cliona varians (CvL-2) and sea cucumber Holothuria grisea (HGL). CvL-2 was able to agglutinate rabbit erythrocytes and was inhibited by galactosides. The hemagglutinating activity was optimal in pH neutral and temperatures below 70 °C. CvL-2 is a trimeric protein with subunits of 175 kDa. On the other hand, HGL showed both hemagglutinating and hemolytic activity in human and rabbit erythrocytes, but...

Mentha pulegium crude extracts induce thiol oxidation and potentiate hemolysis when associated to t-butyl hydroperoxide in human’s erythrocytes Anais da ABC (AABC)
BIANCHINI,MATHEUS C.; GALVÃO,DENNYURA O.; TAMBORENA,TATIANA; ALVES,CLAUDIA O.; PUNTEL,ROBSON L..
ABSTRACT Mentha pulegium (Lamiaceae) tea has been used as a traditional medicine; however, the modulatory effect of M. pulegium extracts on damage to human erythrocytes associated to t-butyl hydroperoxide (t-BHP) exposure remains to be investigated. Accordingly, we perform this study in order to test the hypothesis that aqueous and ethanolic extracts of M. pulegium could modulate the hemolysis associated to t-BHP exposure, non-protein thiol (NPSH) oxidation and lipid peroxidation (measured as thiobarbituric acid reactive substances - TBARS) in human erythrocytes. Samples were co-incubated with t-BHP (4 mmol/L) and/or aqueous or ethanolic extracts (10-1000 mg/mL) during 120 min to further analysis. We found that both extracts, when associated to t-BHP,...

Solubilization of human erythrocyte membranes by ASB detergents BJMBR
Domingues,C.C.; Malheiros,S.V.P.; de Paula,E..
Understanding the membrane solubilization process and finding effective solubilizing agents are crucial challenges in biochemical research. Here we report results on the interaction of the novel linear alkylamido propyl dimethyl amino propanosulfonate detergents, ASB-14 and ASB-16, with human erythrocyte membranes. An estimation of the critical micelle concentration of these zwitterionic detergents (ASB-14 = 100 µM and ASB-16 = 10 µM) was obtained using electron paramagnetic resonance. The amount of proteins and cholesterol solubilized from erythrocytes by these detergents was then determined. The hemolytic activities of the ASB detergents were assayed and the detergent/lipid molar ratios for the onset of hemolysis (Re sat) and total lysis (Re sol) were...

An experimental model of hemolysis-induced acute pancreatitis BJMBR
Saruc,M.; Yuceyar,H.; Turkel,N.; Ozutemiz,O.; Tuzcuoglu,I.; Yuce,G.; Huseyinov,A..
The literature indicates that acute pancreatitis is a complication of massive hemolysis with a prevalence of about 20%. We describe an experimental model of hemolysis-induced acute pancreatitis. Hemolytic anemia was induced in rats by a single ip injection of 60 mg/kg of 20 mg/ml acetylphenylhydrazine (APH) in 20% (v/v) ethanol on the first experimental day (day 0). One hundred and fifty Wistar albino rats weighing 180-200 g were divided into three groups of 50 animals each: groups 1, 2 and 3 were injected ip with APH, 20% ethanol, and physiological saline, respectively. Ten rats from each group were sacrificed on study days 1, 2, 3, 4 and 5. Serum amylase, lipase levels and pancreatic tissue tumor necrosis factor-alpha (TNF-alpha) and platelet-activating...

Effects of different general anesthetics on serum hemolysis and hepatic and muscular glycogenolysis in rats BJMBR
Machado,E.F.A.; Normand,A.C.R.; Nunes,L.A.S.; Brenzikofer,R.; Macedo,D.V..
Anesthetics can affect the structure and biological function of tissues and systems differentially. The aim of the present study was to compare three injectable anesthetics generally used in experiments with animals in terms of the degree of hemolysis and glycogenolysis occurring after profound anesthesia. Twenty-four male Wistar rats (330-440 g) were divided into three groups (N = 8): chloral hydrate (CH), ketamine + xylazine (KX), Zoletil 50® (zolazepam and tiletamine) + xylazine (ZTX). After deep anesthesia, total blood was collected. The liver and white (WG) and red gastrocnemius (RG) muscles were also immediately removed. The degree of serum hemolysis was quantified on the basis of hemoglobin concentration (g/L). Hepatic and muscular glycogen...

Antimicrobial susceptibility, hemolysis, and hemagglutination among Aeromonas spp. isolated from water of a bovine abattoir BJM
Bizani,Delmar; Brandelli,Adriano.
The presence of Aeromonas spp. in food has been demonstrated. They are often introduced from water, which is the natural habitat of many species and is thought to be the main source of contamination. The occurrence of Aeromonas spp. was investigated in 70 water samples of a bovine abattoir. Aeromonas spp. were present in 21.4% of water samples. A. hydrophila was isolated from 11.4% of supplying water samples and from 25.7% of the water drained from carcasses, whereas A. sobria was sole isolated from 5.7% of supplying water samples. Although greater number of positive samples were from water drained from carcasses, only A. hydrophila isolates were present. This suggests that the supplying water is the source of contamination. The antibiotic susceptibility...
