Prolonged exposure of human embryonic stem cells to heat shock induces necrotic cell death Biocell
Heng,Boon Chin; Vinoth,Kumar Jayaseelan; Lu,Kai; Deng,Xuliang; Ge,Zigang; Bay,Boon Huat; Cao,Tong.
We investigated the effects of prolonged heat shock treatment on human embryonic stem cell (hESC) viability. The hESC viability steadily declined with longer exposure to heat shock treatment (43ºC). After 4 h of exposure to heat shock at 43ºC, only 56.2 ± 1.5% of cells were viable. Viability subsequently declined to 37.0 ± 3.3% and 3.5 ± 0.7% after 8 h and 16 h, respectively of heat shock treatment at 43ºC. Transmission electron micrographs showed that the morphology of the dead/dying cells after heat shock treatment was characteristic of cellular necrosis with an uncondensed chromatin and a non-intact plasma membrane. This was further confirmed by flow cytometry analysis which showed that the DNA of the dead/ dying cells was still mostly intact, unlike...

Effect of exogenous nitric oxide on sperm motility in vitro Biol. Res.
Wang,Jiangtao; He,Qingliu; Yan,Xingyu; Cai,Youmei; Chen,Junyi.
BACKGROUND: Nitric oxide (NO) has been shown to be important in sperm function, and the concentration of NO appears to determine these effects. Studies have demonstrated both positive and negative effects of NO on sperm function, but have not been able to provide a clear link between NO concentration and the extent of exposure to NO. To study the relationship between nitric oxide and sperm capacitationin vitro, and to provide a theoretical basis for the use of NO-related preparations in improving sperm motility for in vitro fertilization, we investigated the effects of NO concentration and time duration at these concentrations on in vitro sperm capacitation in both normal and abnormal sperm groups. We manipulated NO concentrations and the time duration of...

Does the change on gastrointestinal tract microbiome affects host? BJID
Beirão,Elisa M.; Padovan,Ana Carolina B.; Furtado,Juvêncio J.D.; Colombo,Arnaldo L.; Medeiros,Eduardo A.S..
During the past decade, studies on the composition of human microbiota and its relation to the host became one of the most explored subjects of the medical literature. The development of high-throughput molecular technologies allowed a deeper characterization of human microbiota and a better understanding of its relationship with health and disease. Changes in human habits including wide use of antimicrobials can result in dysregulation of host–microbiome homeostasis, with multiple consequences. The purpose of this review is to highlight the most important evidence in the literature of host–microbiome interactions and illustrate how these intriguing relations may lead to new treatment and prevention strategies.

Fatal adenoviral necrotizing bronchiolitis case in a post-cardiac surgery intensive care unit BJID
Castelli,Jussara Bianchi; Siciliano,Rinaldo F; Vieira,Ricardo D; Aiello,Vera D; Strabelli,Tânia MV.
We report a case of a 67 year-old-male patient admitted to the intensive care unit in the post-coronary bypass surgery period who presented cardiogenic shock, acute renal failure and three episodes of sepsis, the latter with pulmonary distress at the 30th post-operative day. The patient expired within five days in spite of treatment with vancomycin, imipenem, colistimethate and amphotericin B. At autopsy severe adenovirus pneumonia was found. Viral pulmonary infections following cardiovascular surgery are uncommon. We highlight the importance of etiological diagnosis to a correct treatment approach.

The use of reverse transcription-PCR for the diagnosis of X-linked chronic granulomatous disease BJMBR
Agudelo-Flórez,P.; López,J.A.; Redher,J.; Carneiro-Sampaio,M.M.S.; Costa-Carvalho,B.T.; Grumach,A.S.; Condino-Neto,A..
Chronic granulomatous disease (CGD) is an inherited disorder of the innate immune system characterized by a defective oxidative burst of phagocytes and subsequent impairment of their microbicidal activity. Mutations in one of the NADPH-oxidase components affect gene expression or function of this system, leading to the phenotype of CGD. Defects in gp91-phox lead to X-linked CGD, responsible for approximately 70% of CGD cases. Investigation of the highly heterogeneous genotype of CGD patients includes mutation analysis, Northern blot or Western blot assays according to the particular case. The aim of the present study was to use reverse transcription (RT)-PCR for the analysis of molecular defects responsible for X-linked CGD in eight Brazilian patients and...

Up-regulated expression of CD147 gene in malignant bone tumor and the possible induction mechanism during osteoclast formation BJMBR
Qiao,Suchi; Liu,Chang; Xu,Weijie; AZhaTi,WuBuLi; Li,Cheng; Wang,Zhiwei.
It is increasingly evident that the microenvironment of bone can influence cancer phenotype in many ways that favor growth in bone. CD147, a transmembrane protein of the immunoglobulin (Ig) superfamily, was identified independently in different species and has many designations across different species. However, expression levels of CD147 mRNA in bone cancer have not been described. In this study, we have used real-time fluorescence quantification (RT-PCR) to demonstrate CD147 expression in malignant bone cancer and benign bone tumor tissues. The results suggested that the expression of CD147 gene was significantly up-regulated in malignant bone cancer. Moreover, we found that over-expressed RANKL progressively enhanced osteoclast formation up to 48 h,...

Evaluation of behavioral states among morning and evening active healthy individuals BJMBR
Hidalgo,M.P.; Camozzato,A.; Cardoso,L.; Preussler,C.; Nunes,C.E.; Tavares,R.; Posser,M.S.; Chaves,M.L.F..
The Horne-Östberg questionnaire partly covers some factors that may be important determinants of peak time and characterize patterns of behavior. We conducted a study for the evaluation of self-reported behavioral states (hunger sensation, availability for study, physical exercise, solving daily problems, and time preferences) as expressions of underlying cyclic activity. Three hundred and eighteen community subjects without history of medical, psychiatric, or sleep disorders were evaluated in a cross-sectional design. A self-report about daily highest level of activity was used to categorize individuals into morning, evening, and indifferently active. Time-related behavioral states were evaluated with 23 visual analog questions. The responses to most...

CLIMATE CHANGE AND RURAL WORKERS THERMAL COMFORT: HISTORICAL AND FUTURE IMPACTS REA
Amaro,André L. N.; Yanagi Junior,Tadayuki; Yanagi,Sílvia de N. M.; Ferraz,Gabriel A. E S.; Campos,Alessandro T..
Abstract The aim of the present research was to propose a bioclimatic mapping to classify the thermal comfort and discomfort of rural workers within the state of Minas Gerais, considering historical and future scenarios. Monthly historical series (1976-2014) of minimum, mean and maximum temperature-humidity index (THI), determined through the values of air temperature (minimum, mean and maximum) and relative humidity from 48 weather stations located in the state of Minas Gerais were used to analyze the trends through Mann-Kendall and linear regression assays. The bioclimatic mapping of human comfort, obtained via geostatistical analysis, was developed as a function of the minimum, medium and maximum THI for the historical period (1976-2014) and future...

True polyploid meiosis in the human male Genet. Mol. Biol.
Pearson,Peter L.; Madan,Kamlesh.
Abstract Polyploidy does not usually occur in germinal cells of mammals and other higher vertebrates. We describe a unique example of mosaic autotetraploidy in the meiosis of a human male. Although the original observations were made in the late 1960s, we did not publish them at that time, because we expected to detect further examples that could be described together. However, this did not occur and we have now decided to make the observations available to demonstrate that polyploidy in mammalian male meiosis can arise at a higher frequency than expected by random polyploidization of individual meiotic cells, by either DNA duplication or cell fusion prior to synapsis. This is the first description of a population of primary spermatocytes exhibiting...

Diagnostic moléculaire de l'origine des contaminations fécales dans l'environnement littoral - Développement de marqueurs Bacteroidales spécifiques de l'hôte ArchiMer
Mieszkin, Sophie.
Human and animal faecal pollution affects environmental water in inland and coastal areas, with negative implications for recreational uses, public safety and shellfish sanitary status due to the presence of enteric pathogens. Starting from 2011, the revised Bathing Water European Directive (2006/7/CE) requires the establishment of bathing water profiles with an inventory and study of the pollution sources likely to affect water quality. The faecal microbiological indicators used in these regulations, Escherichia coli and enteroccoci, cannot distinguish between human and animal faecal contamination. Thus, alternative methods, that focus on target microorganisms such as bacteria belonging to the Bacteroidales order were developed or are being developed to...

Comparative assessment of human and farm animal faecal microbiota using real-time quantitative PCR ArchiMer
Furet, Jean-pierre; Firmesse, Olivier; Gourmelon, Michele; Bridonneau, Chantal; Tap, Julien; Mondot, Stanislas; Dore, Joel; Corthier, Gerard.
Pollution of the environment by human and animal faecal pollution affects the safety of shellfish, drinking water and recreational beaches. To pinpoint the origin of contaminations, it is essential to define the differences between human microbiota and that of farm animals. A strategy based on real-time quantitative PCR (qPCR) assays was therefore developed and applied to compare the composition of intestinal microbiota of these two groups. Primers were designed to quantify the 16S rRNA gene from dominant and subdominant bacterial groups. TaqMan((R)) probes were defined for the qPCR technique used for dominant microbiota. Human faecal microbiota was compared with that of farm animals using faecal samples collected from rabbits, goats, horses, pigs, sheep...

Molecular cloning of flounder Xp18, a newly identified highly conserved protein mainly expressed in the ovary ArchiMer
Cachot, Jerome; Bultelle, Florence; Drouot, Laurent; Galgani, Francois; Frebourg, Thierry; Leboulenger, François; Danger, Jean-michel.
Screening of a flounder ovary cDNA library with a rainbow trout p53 probe led to the isolation of a p53-unrelated cDNA encoding an unknown 161 amino acid protein. In view of its apparent molecular weight and yet unknown function, the deduced protein was named Xp18. Corresponding orthologous cDNAs or expressed sequence tags have been identified in several species including human, rodents, bovine, chicken and zebrafish and a related cDNA has also been isolated in the fruit fly. Deduced amino acid sequences appeared to be extremely well conserved throughout vertebrate evolution. Structure predictions suggested that Xp 18 may correspond to an integral protein comprising four transmembrane domains. The charged C-termini of all known vertebrate Xp18-like...

Regression and Simulation Models for Human and Baboon Brain Parameters International Journal of Morphology
Pokhariyal,Ganesh; Hassanali,Jameela.
The variations in morphometric parameter of mammalian brains may be influenced by process of functional complexity, evolution and adaptation. Comparative analysis of linear measurements of cerebrum in the human and baboon has shown morphometric differences. In the present study linear measurements from human and baboon cerebrum (n=10 each) were used to predict various values for human and baboon brain and body parameters through multiple regression models. The average brain weights were found to be 2.08% and 0.84% of the body weights for humans and baboons respectively. The elasticity of regression models revealed that unit percentage increase in Occipital-Frontal (OF) distance would increase the human brain weight by 66.19%, while the baboon brain weight...

Comparative Scanning Electron Microscopy of Platelets and Fibrin Networks of Human and Differents Animals International Journal of Morphology
Pretorius,E; Vieira,W. A; Oberholzer,H. M; Auer,R. E. J.
The purpose of the present study was to compare the platelet and fibrin network ultrastructure of humans to eight different animal species in order to determine the differences between human and animal platelet and fibrin morphology, and to determine whether the animals studied differ in their platelet and fibrin morphology, and whether these differences can be observed by scanning electron microscopy. Platelets and fibrin networks play an important role both in the coagulation process as well as physiologically in allergic processes and immunological mechanisms. The thickness of human fibrin networks were compared to mouse (Mus musculus), equine (Equus caballus), vervet monkey (Chlorocebus aethiops previously Cercopithecus aethiops), oryx (Oryx gazella),...
