Production and characterization of a monoclonal antibody against an Ascaris suum allergenic component BJMBR

Ascaris suum allergenic components (PIII) separated by gel filtration chromatography of an adult worm extract were used to immunize BALB/c mice. Popliteal lymph node cells taken from the immunized animals were fused with SP2/O myeloma cells using polyethylene glycol (MW 1450) as fusogen. The hybridomas were cultured in HAT-containing medium and cloned at limiting dilutions. Supernatants from the growing hybrids were screened by ELISA using plates coated with PIII or the A. suum crude extract. Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by intense polyclonal production of autoantibodies and circulating immune complexes. Some reports have associated SLE with a Th2 immune response and allergy. In the present study 21 female patients with SLE were investigated for total IgE and IgE antibodies to dust house aeroallergens by an automated enzyme-linked fluorescent assay, and were also evaluated for antinuclear IgE autoantibodies by a modified indirect immunofluorescence test using HEp-2 cells as antigen substrate. A model of chronic IgE-mediated food allergy in ovalbumin-sensitized mice BJMBR

Food allergy is most frequently the result of IgE-mediated hypersensitivity reactions. Here, we describe a chronic model in which some of the intestinal and systemic consequences of continuous egg white solution ingestion by ovalbumin-sensitized eight-week-old BALB/c mice, 6 animals per group, of both sexes, were investigated. There was a 20% loss of body weight that began one week after antigen exposure and persisted throughout the experiment (3 weeks). The sensitization procedure induced the production of anti-ovalbumin IgG1 and IgE, which were enhanced by oral antigen exposure (129% for IgG1 and 164% for IgE, compared to sensitization values). Immunoglobulin E (IgE) and mast cells are believed to play important roles in allergic inflammation. However, their contributions to the pathogenesis of human asthma have not been clearly established. Significant progress has been made recently in our understanding of airway inflammation and airway hyperresponsiveness through studies of murine models of asthma and genetically engineered mice. Some of the studies have provided significant insights into the role of IgE and mast cells in the allergic airway response. In these models mice are immunized systemically with soluble protein antigens and then receive an antigen challenge through the airways. The pharmacological substances adenosine deaminase (ADA), histamine, and IgE are endogenously present in animals. They are implicated in allergy and asthma and detectable in blood serum. This research reports the presence of ADA, histamine, and IgE at varying levels in almost all major organs of mice. This research further reports that intramuscular injection of sub-lethal dose of Naja kaouthia venom disrupted homeostasis and lowered the levels of ADA, histamine, and IgE in the organs of mice. Pharmacological substances such as adenosine deaminase (ADA), collagen, histamine, IgE, myoglobin, and nerve growth factor (NGF) are endogenously present in animals. Research from this laboratory reported decreased levels of ADA, histamine, IgE, and NGF in organs of mice injected with sub-lethal doses of cobra venom. The goal of this research is to observe the levels of ADA, collagen, histamine, IgE, myoglobin, and NGF in certain organs of mice injected with venom from the bee Apis mellifera. Adenosine deaminase (ADA), histamine, and IgE are endogenously present in animals. Research from this laboratory reported decreased levels of these substances in organs of mice as a consequence of sub-lethal injection of Naja kaouthia venom. This research reports that decreased ADA, histamine, and IgE levels were prevented by specific treatment and prolonged recovery periods. Adult Balb/c mice injected IM with sub-lethal venom dose were divided into five groups.
