Micropropagation protocol for Syngonanthus elegans (Bong.) Ruhland: an ornamental species Agronomy

The research aimed to establish a protocol of micropropagation of Syngonanthus elegans. Seed germination in WPM media containing 0%, 25%, 50%, 75% and 100% salt concentrations was evaluated. Establishment of plantlets in WPM and MS media, with 50% or 100% salt concentrations, was studied. The addition of 0, 5, 10, 15, 20, 25 and 30 g L-1of sucrose in WPM media was evaluated. The effects of TDZ (0.0; 0.5; 1.0; 2.0 and 4.0 mg L-1) and ANA (0.0; 0.5 and 1.0 mg L-1) in all possible combinations were investigated. Organogenesis and plant regeneration of Arachis villosa Benth. (Leguminosae) through leaf culture Biocell

With the aim of developing an efficient plant regeneration protocol, leaflet explants of three accessions of Arachis villosa Benth. (S2866, S2867 and L97) were cultured on basic Murashige and Skoog medium supplemented with different combinations of plant growth regulators: α-naphthalenacetic acid, indole-3-butyric acid, 6-benzylaminopurine, kinetin and thidiazuron. The accession L97 was the only one able to differentiate buds through indirect organogenesis. The most suitable combination for bud regeneration was the basic medium added with 13.62 μM thidiazuron and 4.44 μM 6-benzylaminopurine. Growth curve and development of the internal calli structure of Eucalyptus camaldulensis Dehn BABT

The objective of this work was to elucidate the growth curve of Eucalyptus camaldulensis Dehn. calli analyzing their anatomical modifications. A sigmoid aspect of the growth curve of the calli fresh matter was observed, with five different phases (lag, exponential, linear, deceleration and decline). In the lag phase, the highest growth percentage 87%, was observed, which reduced during the evaluation period to 17% in the linear phase. As for the anatomical analyses, cellular multiplications was observed during the lag and exponential phases and increase in cell size during the linear phase, promoting the calli volume growth and the establishment of the globular conformation.

Protoplast production from napier grass and pearl millet triploid hybrids Ciência e Agrotecnologia

The objective of this work was to obtain protoplasts from napier grass and pearl millet triploid hybrids as a basis for future studies on chromosomal duplication. Explants were taken from mesophyll of in vitro- and in vivo-cultured plants or from calli of two triploid hybrids (H1 and H2), which were treated with enzymatic solutions containing different concentrations of cellulase R-10 (0.5, 1.0, 1.5 and 2.0%) with an additional 0.2% macerozyme and 0.1% driselase or 1.0% pectolyase Y-23 and 0.5% hemicellulase. Enzymatic digestion was monitored once every hour for five hours. Protoplasts were obtained from in vitro and in vivo leaflets of both triploid hybrids, and in vitro leaflets were the best explant sources. The quantity of produced protoplasts varied...

Micropropagation of Syngonanthus elegantulus Ciência e Agrotecnologia

The species Syngonanthus elegantulus Ruhland is an important native plant used as cut flowers, but extractive exploration has caused the endangered status of this plant. Therefore, the objective of this study was to establish a protocol of the micropropagation of S. elegantulus. To this end, seed germination was tested in media containing different concentrations of WPM salts. For plantlet establishment, MS and WPM media were evaluated with 50 and 100% of the salt concentrations. Different the sucrose levels in the WPM medium were also evaluated. ABSTRACT: Cochlospermum regium roots are used in popular medicine and its extract has diverse phytochemical molecules some with antimicrobial activity, consequently exposing this specie to genetic erosion risks. Thus, the objective of this study was to develop an in vitro multiplication protocol using chemical sterilization of culture medium. The success in micropropagation of Dendrobium phalaenopsis Deang Suree is high, but when transplanted into the greenhouse, their survival is minimal. To increase survival in production in the present study it was evaluated the effect of intermediate acclimatization for 30 days in a grow room utilizing the following luminosity conditions: 1- white fluorescent light (B) (18.9µmol m-2 s-1); 2- white fluorescent light + red fluorescent light (GRO-LUX(r)) (BV) (14.85µmol m-2 s-1); 3- red fluorescent light (GRO-LUX(r)) (V) (9.45µmol m-2 s-1) and the control plants were accommodated directly in a greenhouse (162.0µmol m-2 s-1). Here, we present the current state of knowledge on the diversity of Chilean bryophytes (mosses, liverworts, and hornworts), the secondary metabolites present in these plants, and the biological activity of extracts from populations of Chilean species. Our goal is to establish the potential of these plants as a source of useful molecules for pharmaceutical and agricultural industries, thus promoting their conservation. Hemic proliferative disease in the bay mussel, Mytilus edulis, has been studied following mortalities in Washington State (USA). The disease has been reproduced in the labortory. The establishment of this experimental model led to study tumoral characteristics of the atypic hemocytes : ultrastrure, in vitro cultivation, transplantation. The infectious etiology of this tumor has been verified. It has been hypothesized to result from a viral infection since retrovirus-like particules have been observed following induction by bromodeoxyuridine. In Vitro Cultivation of Blood Stream Trypomastigotes of Trypanosoma vivax without Feeder Cell Layers OAK

Bloodstream trypomastigotes (BSFs) of 2 clones (IL 1392 and IL 3671) of Trypanosoma vivax were cultured without feeder layers in 3 systems. System I: metacyclic-producing cultures of T. vivax IL 1392 were initiated with BSFs derived from infected mice at 27 OC in the presence of feeder layers using TVM-1 medium. Metacyclics harvested were then transferred to flasks containing feeder cells and maintained at 34 OC using TVM-22 medium. Under such conditions, the metacyclics transformed to BSFs. Bloodstream trypomastigotes which were then transferred to new flasks, continued to grow in HMI-162 medium without feeder cells.
