Genetic analysis of reciprocal differences in the inheritance of in vitro characters in pearl millet Genet. Mol. Biol.
Satyavathi,Valluri V; Manga,V.; Rao,Muktinutalapati V. Subba; Chittibabu,Malladi.
Abstract Reciprocal differences persist in nature because of the unequal contribution of cytoplasmic determinants from male and female gametes to the zygote. The inheritance of genetic differences is an important factor that influences various traits, including somatic embryogenesis and regeneration in vitro. In this report, we estimate the cytoplasmic and maternal effects in pearl millet and their adequacy in describing the observed reciprocal differences based on an in depth study of the parents, F2s and reciprocal backcross progenies needed for fitting genetical models. Our study revealed that of the two characters examined, embryogenic callus quantity and regeneration frequency, the former showed a greater proportion of cytoplasmic nuclear interaction...

In vitro plant regeneration via indirect organogenesis from different explants of Lathyrus sativus L. and Lathyrus cicera L. Phyton
Li,RS; Tao,YJ; Liu,FJ; Hu,X; Xu,QL; Li,KY.
The grass pea (Lathyrus sativus L.) and flatpod peavine (Lathyrus cicera L.) are the most economically important and widely cultivated Lathyrus species. However, their utilization is limited due to the presence of their endogenous toxin β-N-oxalyl-L-α,β-diaminopropionic acid (β-ODAP). Thus, a β-ODAP free variety should be developed through some plant breeding technique like either mutational breeding or genetic-manipulation. In this circumstance, the plant regeneration of Lathyrus species becomes a bottleneck. In the present study, an efficient system for in vitro regeneration of L. sativus with high β-ODAP levels, and L. cicera with low β-ODAP levels, was developed from different explants...

The Role of Plant Growth Regulators in the Development of in vitro Flowering, Histology and Ultrastructural Studies in Impatiens balsamina cv. Dwarf Bush Planta Daninha
MOHAMED,N.; TAHA,R.M.; RAZAK,U.N.A.A.; ELIAS,H..
ABSTRACT: An efficient protocol for in vitro flowering was successfully established for Impatiens balsamina cv Dwarf Bush, an important medicinal plant, through tissue culture techniques. Shoot, stem and petiole explants obtained from 4 week-old aseptic seedlings cultured on MS medium supplemented with different concentrations of plant growth regulator (PGR) were used for in vitro flower induction. Gibberellic acid (GA3), benzylaminopurine (BAP) and kinetin (Kin) treatment singly applied in MS media (pH 5.8), could all stimulate flowering at 23-26 oC with photoperiod of 16 hours light and 8 hours dark. It was observed that shoot explants were more responsive than stem explants in floral formation. Regeneration was achieved via direct organogenesis. For...

Efficient Regeneration of Eucalyptus urophylla x Eucalyptus grandis from Stem Segment BABT
Ouyang,Lejun; Huang,Zhenchi; Zhao,Longyan; Sha,Yuee; Zeng,Fuhua; Lu,Xiangyang.
The aim of the present study was to establish an efficient regeneration system for the hybrid E. urophylla x E. grandis by means of organogenesis. Stem segments from seedlings were used as explants and cultured in a modified Murashige and Skoog medium (MS), supplemented with 13.2 µM N-phenyl-N'-[6-(2-chlorobenzothiazol)-yl] urea (PBU) and 0.285 µM indole-3-acetic acid (IAA). PBU was a useful growth regulator. After cultivating for 5 d, 96% explants formed callus. After 30 d, the calli obtained were transferred to MS medium containing different combinations of 6-benzyladenine (BA) and naphthalene acetic acid (NAA). Compared with other growth regulator combinations, PBU stimulated more vigorous calli and restrained browning. In addition, a large percentage...

Adventitious shoots from nodule cluster cultures of Vriesea reitzii: an endemic and endangered bromeliad from atlantic forest Ciência Rural
Rech Filho,Arlindo; Vesco,Lírio Luiz Dal; Guerra,Miguel Pedro.
The Atlantic Forest is a biome with megadiversity and a number of bromeliads take part of it. This is the case of Vriesea reitzii, an endemic bromeliad threatened with extinction. Tissue culture techniques are valuable tools for the mass propagation of bromeliads, thus reducing pressure in the natural habitat. The aim of the present work was to establish an in vitro protocol based on the induction and proliferation of nodule cluster cultures of this species. Plantlets maintained in MS liquid culture medium plus NAA (2µM) and BAP (4µM) had the basal regions of leaves excised and then inoculated in gelled with agar (7g L-1) MS culture medium plus with Dicamba (2.5; 5; 10; 20 e 30µM) and Kin (2µM) or free of plant growth regulators. Nodule cluster cultures...

Genetic transformation of the Brazilian BR 451 maize variety by the Agrobacterium tumefaciens method Ciência Rural
Silva,Marilia Rodrigues de; Didoné,Dielli Aparecida; Ceccon,Cássia Canzi; Almeida,Vinícius de Oliveira; Grando,Magali Ferrari.
ABSTRACT: The asexually gene introduction by genetic engineering has brought enormous possibilities to innovate plant breeding. However, principally because of the low in vitro response, genetic transformation has been restricted to only certain genotypes of agronomically significant species. With the objective of establishing a protocol for genetically transforming the Brazilian BR 451 maize variety through Agrobacterium tumefaciens, it was studied the capacity of plant regeneration in vitro from embryogenic calli cultivated in three regeneration media, each having different growth regulators. It was also evaluated the temperature stress effect on the transformation of the immature embryos with A. tumefaciens EHA 101 containing the plasmid pTF102 with...
