Lectins are carbohydrate-binding proteins that have been isolated from various sources and presented a wide spectrum of biological activities. The effects of four lectins, namely, Phaseolus vulgaris phytohemagglutinin, PHA, wheat germ agglutinin, WGA, Artocarpus integrifolia seed lectins, jacalin and artocarpin, on in vitro fibroblasts proliferation were investigated. The lectins did not influence the initial cell adhesion to the plate. PHA and WGA at 10-20 µg/mL concentrations significantly decreased fibroblasts proliferation. At these concentrations, they caused morphological alterations on cells and over 80 µg/mL, promoted cell death. Neither jacalin nor artocarpin significantly affected cell proliferation. |

The presence of carbohydrate-binding proteins, namely lectins, ß-galactosidases and amylases, was determined in aqueous extracts of plants collected in Uruguay. Twenty-six extracts were prepared from 15 Uruguayan plants belonging to 12 Phanerogam families. Among them, 18 extracts caused hemagglutination (HAG) that was inhibited by mono- and disaccharides in 13 cases, indicating the presence of lectins. The other 8 extracts did not cause any HAG with the four systems used to detect HAG activity (rabbit and mouse red cells, trypsin-treated rabbit and mouse red cells). For the extracts prepared from Solanum commersonii, HAG activity and HAG inhibition were similar for those prepared from tubers, leaves and fruits, with the chitocompounds being responsible for... |

Lectins are carbohydrate-binding proteins of non-imune origin. This group of proteins is distributed widely in nature and they have been found in viruses, microorganisms, plants and animals. Lectins of plants have been isolated and characterized according to their chemical, physical-chemical, structural and biological properties. Among their biological activities, we can stress its fungicidal action. It has been previously described the effect of the lectins Dviol, DRL, ConBr and LSL obtained from the seeds of leguminous plants on the growth of yeasts isolated from vaginal secretions. In the present work the experiments were carried out in microtiter plates and the results interpreted by both methods: visual observations and a microplate reader at 530nm.... |

Two lectins were isolated from Canavalia ensiformis and Dioclea grandiflora seeds. Gel filtration produced a fraction corresponding to Con A or D. grandiflora lectin while erythroagglutination assays revealed a distinct fraction presenting a lectin that agglutinates human red blood cells (RBCs) but not rabbit RBCs. Hydrophobic interaction chromatography showed that the latter fraction yielded a protein that readily agglutinates human erythrocytes; the lectin was also purified by affinity chromatography on Lac-Sepharose showing similar properties to that of the Phenyl-Sepharose-purified lectin. Despite minor differences (carbohydrate content or A1%1cm), the two lectins showed similar molecular properties in that they consisted of two non-covalently linked... |

Chenopodium quinoa seeds have high protein content. The nutritional value of quinoa is superior compared with traditional cereals. Its essential amino acid composition is considered next to the ideal, and its quality matches that of milk proteins. In this study, the seed storage proteins from Chenopodium quinoa were extracted, fractionated, partially purified, and characterized. The structural characterization was performed by Tricine-SDS-PAGE and two-dimensional electrophoresis, and it confirmed the presence of proteins of molecular weight of 30 and 7kDa, probably corresponding to lectins and trypsin inhibitors, respectively. The functional characterization of these proteins evidenced their activity as antinutritional factors due to their in vitro... |

This work studied the effect of the lectin phytohaemagglutinin (PHA) and bacterial lipopolysaccharide (LPS) on the activity of nonspecific cytotoxic cells (NCCs) and macrophages of peripheral blood in the rainbow trout. NCCs are cells of the innate immunity, homologues to Natural Killer (NK) cells of mammals, capable of eliminating cells infected with intracellular pathogens and also tumoral cells. Their lytic activity is mediated by the perforin/granzyme system and the Fas receptor-Fas ligand (FasR-FasL) system. Activity of NCCs was evaluated by their capacity to lyse HL-60 tumor cells. On the other hand, macrophages eliminate microorganisms by several mechanisms, including the production of reactive oxygen species (ROS) and nitric oxide (NO). Results... |

The stomach of three species of non-human primates was investigated by lectin histochemistry to clarify the staining affinity and distribution patterns of their sugar residues. All gastric regions, with little differences between the deep and superficial parts of the same region, were rich in. in N-acetylglucosamine and/or neuraminic acid. Although, the superficial regions of the gastric mucosa were scanty in N-acetylgalactosamine, a- D-glucose and a -D-mannose, the deep parts of the gastric mucosa were rich in these sugars. In conclusion, there is a difference among the mucosubstances of surface and foveolar mucous cells, mucous neck cells, and gastric gland cells. This indicates heterogeneous composition of gastric mucus, or mucus molecules with... |

Lectins are proteins with binding sites that recognize a specific sequence of sugar moieties in complex glycoconjugates. In the present study the tomato lectin Lycopersicon esculentum (LEL) (a selective microglial and endothelial marker) has been reported to recognize specific residues of N-acetylglucosamine (GlyNAc) and poly-N-acetyllactosamine. In the pineal gland the biotinylated LEL was used to investigate the appearance of these sugar residues in the structures of the rats during their development and adult life. Our results showed that the binding of LEL occurred exclusively in the material adherents on surface of the endothelia of the vessels in the peripheral and central regions of the gland. An exception can be cited to rats in first postnatal... |

Secreted arginases from Evernia prunastri and Xanthoria parietina thalli, exhibiting lectin activity, have been purified to homogeneity. Analyses of both amino acid and glycoside composition are reported. Purified proteins have been subjected to tryptic digestion and some amino acid sequences have been analyzed. An undecapeptide from Evernia arginase, which shows some degree of homology with the domain to which Mn²+ binds, a heptapeptide and an undecapeptide from Xanthoria arginase have been analyzed, and homologies with some arginases from other fungi and plants have been found. |

ABSTRACT The benefits of various foods, such as fruits and vegetables, have been the focus of several studies aimed at improving welfare, health, and reducing the incidence of diseases. Therefore, the present paper objectified to investigate the presence of molecules of biotechnological and pharmacological interest in peel, seed, and pulp of Annona x atemoya Mabb. Aqueous extracts of the fruit parts were obtained with different buffers and assessed as to their protein and phenolic compounds content. The three parts of the fruit presented different proportions of these compounds when subject to different extraction conditions, with the highest concentrations of proteins being found in the seed and phenolic compounds in the peel of the studied fruit.... |
