Influence of the pH of glutaraldehyde and the use of dextran aldehyde on the preparation of cross-linked enzyme aggregates (CLEAs) of lipase from Burkholderia cepacia Electron. J. Biotechnol.

The preparation of cross-linked enzyme aggregates (CLEAs) of lipase has been a challenge due the low amount of lysine residues that lipases have on their surface. The aim of the present study was to assess the potential of valorisation of a solid industrial derivative of tallow, composed of saturated free-fatty acids ("stearin"), by fermentations carried out by the yeast Yarrowia lipolytica ACA-DC 50109 in order to produce microbial lipid, biomass and extra-cellular lipase. High quantities of biomass were produced (biomass yield of around 1.1 ± 0.1 g of total biomass produced per g of fat consumed) when the organism was grown in shake flasks, regardless of the concentration of extra-cellular nitrogen present. The purpose of this research was to study the effect of the methyl branching of a high log P alkane solvent and the water activity in the organic medium on the initial rate and the enantioselectivity of ibuprofen esterification catalyzed by Candida rugosa lipase. Resolution of ibuprofen is important because S-(+)-ibuprofen has the desired pharmacological activity, whereas the R-(-)-enantiomer causes much of the side effects. The Candida rugosa lipase-catalyzed reaction in isooctane at 40ºC and 0.73 water activity gave the best results, both in terms of the initial reaction rate and the enantioselectivity of the reaction. Lipases are placed only after proteases and carbohydrases in world enzyme market and share about 5% of enzyme market. They occur in plants, animals and microorganisms and are accordingly classified as plant, animal and microbial lipases. Wherever they exist, they function to catalyze hydrolysis of triglycerides to glycerol and fatty acid. A thermostable lipase was partially purified from the culture supernatant of a thermophilic Bacillus sp. The enzyme is optimally active at 60ºC and pH 8.0. The enzyme showed enhancement in activity in presence of benzene or hexane (30% v/v each). The activity (assayed by determining the release of pNP from pNP laurate) was stimulated up to 60% of these solvents in enzyme reaction mixture. The esterification of phenylpropanoid and hydrophenylpropanoid acids, catalyzed by candida antarctica lipase B (CAL-B), with several alcohols has demonstrated that the substitution pattern on the aromatic ring has a very significant influence on the reactivity of the carboxyl group due, mainly, to electronic effects, when compared to the unsaturated acids with the hydrogenated acids. It is also clear that in the saturated acids there still remain some unclear effects related to the aromatic substituents.

Thermostable lipase from a newly isolated Staphylococcus xylosus strain; process optimization and characterization using RSM and ANN Electron. J. Biotechnol.

Normal feed forward back-propagation artificial neural network (ANN) and cubic backward elimination response surface methodology (RSM) were used to build a predictive model of the combined effects and optimization of culture parameters for the lipase production of a newly isolated Staphylococcus xylosus. The results demonstrated a high predictive accuracy of artificial neural network compared to response surface methodology. The optimum operating condition obtained from the ANN model was found to be at 30ºC incubation temperature, pH 7.5, 60 hrs incubation period, 1.8% inoculum size and 60 rpm agitation. Ribosome-inactivating proteins (RIPs) are enzymes that inhibit protein synthesis after depurination of a specific adenine in rRNA. The RIP family members are classified as type I RIPs that contain an RNA-N-glycosidase domain and type II RIPs that contain a lectin domain (B chain) in addition to the glycosidase domain (A chain). In this work, we identified 30 new plant RIPs and characterized 18 Ricinus communis RIPs. Influence of dietary live yeast on European sea bass (Dicentrarchus labrax) larval development ArchiMer

Recent results have demonstrated that live yeast in diets improved gut maturation in sea bass larvae. In this study, we tested the absence and two levels of live yeast in sea bass larvae diets. Specimens were fed from time of mouth opening to 37 days after hatching a diet of 0%, 1.1%, or 5.7% wet weight of live yeast (Debaryomyces hansenii CBS 8339). Yeast incorporation improved survival 10%, and reduced malformed larvae. In groups fed 1.1% yeast, only 1% of larvae were malformed, compared to 14% in the control group. Final mean weight in groups fed 1.1% yeast was twice that of other groups. 乳脂肪およびその分別物のリパーゼ分解 OAK

微生物起源の市販リパーゼを用いて、天然のバターフレーバーを強化する目的で乳脂肪を分解した。 1)市販酵素であるパラターゼAとリパーゼFを比較すると、 リパーゼFの方が分解率が高く、フレーバーも良好であった。 また、酵素添加量を増やすことにより短時間で分解率を上げることができた。 2)リパーゼFでバターオイルを分解した壌合、水相量の違い(10%と20%)、水道水の使用および水酸化物添加による加水分解率への影響はほとんどなかった。 3)リパーゼ分解により遊離した脂肪酸としては11種が検出され、そのうち主なものはパルミチン酸、ステアリン酸およびオレイン酸であった。24hと48hの分解時間で比較すると、遊離した脂肪酸量には違いがあったが、その組成はほとんど変わらなかった。またショートニング希釈およびマドレーヌでの実用試験においても、分解時間によるフレーバーの違いは単に強度的なものだけであった。 4)乳脂肪から分別された低融点画分を同様にリパーゼで分解すると、分解率はバターオイルの場合と比べて多少低かったが、遊離した短・中鎖の脂肪酸組成には差がなかった。官能試験では、刺激臭が弱くなり風味的にマイルドで、またマドレーヌでの実用試験にいでもフレーバー改善効果が認められた。しかし、これはバターフレーバー成分が分別の過程で低融点性画分に多く移行したことによると判断された。

O objetivo deste trabalho foi avaliar o efeito da adição na ração das enzimas digestivas exógenas amilase, lipase e protease sobre o desempenho de tambaqui, em três experimentos independentes. Em cada experimento foi avaliado o efeito da inclusão de uma enzima na ração em diferentes porcentagens: 0,0, 0,05, 0,1 e 0,2%, cada uma com quatro repetições, com 28 dias de duração. Foram realizadas biometrias do peso inicial, para homogeneização da amostra, e do peso final a fim de verificar o efeito dos tratamentos sobre o desempenho dos peixes. Assessment of lipolytic activity of isolated microorganisms from the savannah of the Tocantins Biological Sciences

Current study assesses the biodiversity and selects lipase-producer microorganisms with industrial interest, from the savannah of the state of Tocantins, Brazil. Seventeen pequi microorganisms (Caryocar brasiliense) were isolated in the decomposition stage and 35 microorganisms were retrieved from the soil fraction under the collected pequi. Yarrowia lypolitica strain was used as positive control in all assays. The 52 strains were subjected to tests in a solid medium with Tween 20 for checking halos formed by crystals, indicating lipase production by inoculated strains. Reatividade in vitro de lipase submetida a diferentes tratamentos tecnológicos Arq. Bras. Med. Vet. Zootec.

Analisou-se a atividade enzimática in vitro de uma lipase em diferentes condições de temperatura (simulando o processo de peletização) e pH (acidez gástrica) e na presença de íons minerais e metálicos. Para avaliar a estabilidade térmica, a enzima foi incubada a 80(0)C por quatro tempos (0, 5, 10 e 15 minutos). O efeito do pH foi estudado submetendo-se a enzima a quatro valores de pH (7,3, 2,0, 3,2 e 5,0). Para avaliar o efeito da lipase na presença de íons minerais e metálicos, incubou-se a enzima em diferentes soluções contendo cálcio, cobre, cobalto, manganês e ferro. Frente às diferentes condições, a lipase conservou atividade parcial, mas não pôde ser considerada uma enzima ideal em função do efeito inibidor promovido pelo pH ácido e pelas soluções...
