HLA-Bw4-B*57 and Cw*18 alleles are associated with plasma viral load modulation in HIV-1 infected individuals in Salvador, Brazil BJID
Silva,Edinete Melo da; Acosta,Angelina Xavier; Santos,Eduardo José Melo; Netto,Eduardo Martins; Lemaire,Denise Carneiro; Oliveira,Adriano Silva; Barbosa,Carolina Matos; Bendicho,Maria Teresita; Galvão-Castro,Bernardo; Brites,Carlos.
Host genetic factors play an important role in mediating resistance to HIV-1 infection and may modify the course of infection. HLA-B alleles (Bw4 epitope; B*27 and B*57) as well as killer cell immunoglobulin-like receptors have been associated with slow progression of HIV-1 infection. OBJECTIVE: To evaluate the association between serological epitopes HLA-Bw4 and HLA-Bw6 and prognostic markers in AIDS. METHODS: 147 HIV-infected individuals in Bahia, Northeast Brazil, were genotyped for HLA class I locus. HLA class I genotyping was performed by hybridization with sequence-specific oligonucleotide probes following amplification of the corresponding HLA-A, HLA-B and HLA-C genes. Statistical analysis was performed using Fisher's exact and ANOVA tests for...

Association of Alport's syndrome with HLA-DR2 antigen in a group of unrelated patients BJMBR
Donadi,E.A.; Voltarelli,J.C.; Paula-Santos,C.M.; Kimachi,T.; Ferraz,A.S..
A few family studies have evaluated HLA antigens in Alport's syndrome; however, there are no large population studies. In the present report, we studied 40 unrelated white patients with Alport's syndrome seen at the Unit of Renal Transplantation, Faculty of Medicine of Ribeirão Preto, São Paulo, Brazil. HLA-A, -B, -DR and -DQ antigens were typed using a complement-dependent microlymphocytotoxicity assay. A control white population (N = 403) from the same geographical area was also typed for HLA antigens. Although the frequencies of HLA-A and -B antigens of patients were not statistically different from controls, the frequency of HLA-DR2 antigen observed in patients (65%) was significantly increased in relation to controls (26%; P<0.001). The...

MHC-restricted antigen presentation and recognition: constraints on gene, recombinant and peptide vaccines in humans BJMBR
Cunha-Neto,E..
The target of any immunization is to activate and expand lymphocyte clones with the desired recognition specificity and the necessary effector functions. In gene, recombinant and peptide vaccines, the immunogen is a single protein or a small assembly of epitopes from antigenic proteins. Since most immune responses against protein and peptide antigens are T-cell dependent, the molecular target of such vaccines is to generate at least 50-100 complexes between MHC molecule and the antigenic peptide per antigen-presenting cell, sensitizing a T cell population of appropriate clonal size and effector characteristics. Thus, the immunobiology of antigen recognition by T cells must be taken into account when designing new generation peptide- or gene-based vaccines....

Idiopathic focal segmental glomerulosclerosis and HLA antigens BJMBR
Gerbase-DeLima,M.; Pereira-Santos,A.; Sesso,R.; Temin,J.; Aragão,E.S.; Ajzen,H..
The objective of the present study was to investigate a possible association between HLA class II antigens and idiopathic focal segmental glomerulosclerosis (FSGS). HLA-A, -B, -DR and -DQ antigens were determined in 19 Brazilian patients (16 white subjects and three subjects of Japanese origin) with biopsy-proven FSGS. Comparison of the HLA antigen frequencies between white patients and white local controls showed a significant increase in HLA-DR4 frequency among FSGS patients (37.7 vs 17.2%, P<0.05). In addition, the three patients of Japanese extraction, not included in the statistical analysis, also presented HLA-DR4. In conclusion, our data confirm the association of FSGS with HLA-DR4 previously reported by others, thus providing further...

MHC Class II haplotypes of Colombian Amerindian tribes Genet. Mol. Biol.
Yunis,Juan J.; Yunis,Edmond J.; Yunis,Emilio.
We analyzed 1041 individuals belonging to 17 Amerindian tribes of Colombia, Chimila, Bari and Tunebo (Chibcha linguistic family), Embera, Waunana (Choco linguistic family), Puinave and Nukak (Maku-Puinave linguistic families), Cubeo, Guanano, Tucano, Desano and Piratapuyo (Tukano linguistic family), Guahibo and Guayabero (Guayabero Linguistic Family), Curripaco and Piapoco (Arawak linguistic family) and Yucpa (Karib linguistic family). for MHC class II haplotypes (HLA-DRB1, DQA1, DQB1). Approximately 90% of the MHC class II haplotypes found among these tribes are haplotypes frequently encountered in other Amerindian tribes. Nonetheless, striking differences were observed among Chibcha and non-Chibcha speaking tribes. The DRB1*04:04, DRB1*04:11, DRB1*09:01...

Polymorphism of exon 2-3 of bovine major histocompatibility complex class I BoLa-A gene Genet. Mol. Biol.
Bhushan,Bharat; Patra,Biswa Nath; Das,Pranab Jyoti; Dutt,Triveni; Kumar,Pushpendra; Sharma,Arjava; Umang,; Dandapat,Satyabarta; Ahlawat,Shiv Pal Singh.
The exon 2-3 region of bovine major histocompatibility complex (MHC) class I BoLa-A gene was investigated for polymorphisms in three breeds of cattle originated in the Indian subcontinent namely Sahiwal, Tharparkar, Hariana, as well as crossbred (Bos taurus x Bos indicus) cattle and Jersey, the exotic breed (Bos taurus). The PCR amplified fragment of 714 bp showed distinct DdeI-, TaqI- and HinfI- RFLP patterns, thus confirming a higher degree of polymorphism in this region. To our knowledge this is the first report of HinfI restriction patterns for BoLa-A exon 2-3. The sequencing results revealed a number of nucleotide substitutions in this region, which resulted in amino acid changes. The present investigation confirmed that MHC class I BoLa-A exon 2-3 is...

Molecular characterization of MHC-DRB cDNA in water buffalo (Bubalus bubalis) Genet. Mol. Biol.
Naskar,Soumen; Deb,Sitangsu M.; Niranjan,Saket K.; Kumar,Subodh; Sharma,Deepak; Sakaram,Durgam; Sharma,Arjava.
In the present study, water buffalo MHC (Bubu)-DRB cDNA was cloned and characterized. The 1022 base long-amplified cDNA product encompassed a single open reading frame of 801 bases that coded for 266 amino acids. The Bubu-DRB sequence showed maximum homology with the BoLA-DRB3*0101 allele of cattle. A total of seven amino acid residues were found to be unique for the Bubu-DRB sequence. The majority of amino acid substitutions was observed in the β1 domain. Residues associated with important functions were mostly conserved. Water buffalo DRB was phylogenetically closer to goat DRB*A.

Report of new alleles at BG loci in Camperos chickens JBAG
Iglesias,G. M; Huguet,M. J; Goto,R. M.; Miquel,M. C; Miller,M. M.
Camperos are meat-type chickens developed in Argentina that are under study for their response to disease. Nine new alleles were found in Campero chickens (from INTA EEA Pergamino) at the highly polymorphic BG loci within the Major Histocompatibility Complex (MHC), and were later reported to the GenBank (Accession numbers: DQ17443, DQ174444, DQ176443-DQ6449). All of them vary from previously reported BG alleles obtained from white Leghorn chickens and the Red Jungle Fowl from the Chicken Genome Project. Animals were selected from line-crossing among Plymouth Rock Red, Rhode Island Red and Cornish Red. Sequences reported add further evidence about the extreme diversity of these highly polymorphic, rapidly-evolving MHC genes.
