ABSTRACT The economic exploitation of macaw palm [Acrocomia aculeate(Jacq.) Lodd. ex Mart.] is currently in transition, from extractivism to agricultural cultivation, thus requiring studies on the fertilization of the crop. This study evaluated the response of three genotypes of macaw palm to increasing rates of nitrogen and potassium, grown in the field until the 2nd year and to establish reference contents of mineral nutrients in the leaf. ABSTRACT Knowing the maturation of the Acrocomia aculeata (macaw palm) fruit is decisive for harvest to be performed when the fruit has a higher content and quality of oil. There are different approaches to monitor maturation and they can be classified as destructive and non-destructive. Variables obtained by optical instruments have been used to determine maturation stage. The present study evaluated the relationship between the biological activity measured by the optical technique of biospeckle laser and firmness, measured in the Acrocomia aculeata fruit pulp between the 41st and 61st week after flowering. Macaw palm (Acrocomia aculeata (Jacq.) Lodd. ex Mart.) is a tropical species with multiple uses, including oil supply for biodiesel production. However, structural and physiological studies of the seed are still scarce, as well as in vitro propagation techniques. The aim of this study was to characterize the anatomy, histochemistry and ultrastructure of the seed and the in vitro somatic embryo of A. aculeata, gaining insight into relationships between the post-harvest seed behavior and the somatic embryo conversion to plant. Somatic embryogenesis in Acrocomia aculeata Jacq. (Lodd.) ex Mart using the thin cell layer technique Acta Botanica

Considering the necessity of Acrocomia aculeata propagation for large-scale production, the aim of this study was to establish a somatic embryogenesis protocol using the thin cell layer (TCL) technique. Aerial parts of in vitro plants were transversally cut at the base into eight TCLs and placed in a culture medium for callus induction. The induction medium was composed of Y3 salts and Morel´s vitamins and supplemented with 150, 300 or 600 μM picloram. After 12 weeks the calli were transferred to a medium supplemented with BAP or 2-iP (12.5 or 25 μM). After 18 weeks, the somatic embryo clusters were transferred to a conversion medium (plant growth regulator-free medium).
