A survey of the Anopheles Mosquitoes (Diptera: Culicidae) along the Acre And Purus Rivers, Amazon Basin, Brazil Acta Amazonica
ZIMMERMAN,Robert H; PAULINO,Francisco Xavier; ARRUDA,Mercia de.
From January 19 to February 25, 1997 an entomological survey of the seringais and larger towns along the Acre and Purus rivers was made as part of the project “Revisitando a Amazônia de Carlos Chagas; da Borracha a Biodiversidad”. Eleven anopheline species and 1285 specimens were collected landing on human baits. The four most abundant species were Anopheles albitarsis s.l. (n=778), A. darlingi (n=359), A. rangeli (n=69) and A. oswaldoi (n=60). A total of 252 larvae were collected of which 10 anopheline species were identified. The most abundant species collected were A. albitarsis s.l. (n=88), A. deaneorum (n=45) and A. triannulatus (n=40). The low numbers of Anopheles collected and the absence of the principal malaria vector A. darlingi at the seringais...

ABSTRACT Malaria is a disease of global tropical distribution, being endemic in more than 90 countries and responsible for about 212 million cases worldwide in 2016. To date, the strategies used to eradicate this disease have been ineffective, without specific preventive measures such as vaccines. Currently, the existing therapeutic arsenal is limited and has become ineffective against the expansion of artemisinin-resistant Plasmodium, demonstrating the need for studies that would allow the development of new compounds against this disease. In this context, we studied the volatile oil obtained from rhizomes of Cyperus articulatus (VOCA), a plant species commonly found in the Amazon region and popularly used as a therapeutic alternative for the treatment of...

ABSTRACT The middle Rio Negro region is an interesting place to seek novel antimalarial compounds because of the traditional knowledge of the Amazon population in conjunction with the high biodiversity of the region. The objective of this work was to study the use of antimalarial plants in Barcelos, state of Amazonas, Brazil. Fifty-two local specialists from eight different communities were interviewed over one year. The identified plants were collected with the assistance of local specialists, classified to species level and deposited in herbarium. In total, 55 antimalarial plants were identified in use in the region, of which 16 had not been previously reported in other published studies. Many factors could be involved with the use of antimalarial plants...

EXPORT AND TRAFFICKING OF Plasmodium PROTEINS WITHIN THE HOST ERYTHROCYTE Acta biol.Colomb.
WISER,MARK F.
The export and trafficking of parasite proteins within the infected erythrocyte is a complex process and not well understood. Export of proteins from the parasite was previously speculated to involve a specialized compartment originally designated as the secondary endoplasmic reticulum of the Apicomplexa, or sERA. The properties of this Plasmodium export compartment are reviewed in regards to more recent observations about the trafficking of Plasmodium proteins within the host erythrocyte. In addition, a calcium ATPase unique to the Apicomplexa and with homology to the sarcoplasmicendoplasmic reticulum calcium ATPase is discussed in the context of this unique export compartment. It is also speculated that the Plasmodium export element, PEXEL, may play a...

Current Antimalarial Therapies and Advances in the Development of Semi-Synthetic Artemisinin Derivatives Anais da ABC (AABC)
PINHEIRO,LUIZ C.S.; FEITOSA,LÍVIA M.; SILVEIRA,FLÁVIA F. DA; BOECHAT,NUBIA.
ABSTRACT According to the World Health Organization, malaria remains one of the biggest public health problems in the world. The development of resistance is a current concern, mainly because the number of safe drugs for this disease is limited. Artemisinin-based combination therapy is recommended by the World Health Organization to prevent or delay the onset of resistance. Thus, the need to obtain new drugs makes artemisinin the most widely used scaffold to obtain synthetic compounds. This review describes the drugs based on artemisinin and its derivatives, including hybrid derivatives and dimers, trimers and tetramers that contain an endoperoxide bridge. This class of compounds is of extreme importance for the discovery of new drugs to treat malaria.

Signal transduction in Plasmodium-Red Blood Cells interactions and in cytoadherence Anais da ABC (AABC)
Cruz,Laura N.; Wu,Yang; Craig,Alister G.; Garcia,Célia R.S..
Malaria is responsible for more than 1.5 million deaths each year, especially among children (Snow et al. 2005). Despite of the severity of malaria situation and great effort to the development of new drug targets (Yuan et al. 2011) there is still a relative low investment toward antimalarial drugs. Briefly there are targets classes of antimalarial drugs currently being tested including: kinases, proteases, ion channel of GPCR, nuclear receptor, among others (Gamo et al. 2010). Here we review malaria signal transduction pathways in Red Blood Cells (RBC) as well as infected RBCs and endothelial cells interactions, namely cytoadherence. The last process is thought to play an important role in the pathogenesis of severe malaria. The molecules displayed on the...

Spotlight on Plasmodium falciparum evolutionary system in the southeastern Atlantic forest Biota Neotropica
Laporta,Gabriel Zorello.
Abstract Malaria elimination is now set to occur in Brazil until 2030. While this achievement is feasible, as it is for other endemic regions worldwide, it is important to recognize resistance of parasites and vectors against anti-malarial interventions. Resistance against drugs and insecticides can lead to discontinuities of malaria transmission, known as residual malaria transmission. Herein, we described a novel phenomenon that is occurring in a residual malaria transmission scenario in the southeastern Atlantic forest. This novel phenomenon does not belong to what is known and therefore we decided to explain it based on an evolutionary perspective. Although it shall not be viewed as a threat to public health, the phenomenon has important aspects that...

Plasmodium vivax malaria presenting with severe thrombocytopenia BJID
Makkar,Ravinder Pal Singh; Monga,Surabhi Mukhopadhyay Amitabh; Gupta,Ajay Kr..
Plasmodium falciparum and Plasmodium vivax malaria are endemic infections in India and are commonly associated with mild hematological abnormalities. Severe thrombocytopenia is common in isolated falciparum and mixed falciparum/vivax malaria, but is very rare in isolated P.vivax infection. We hereby report a case of severe thrombocytopenia (platelet count of 8x10(9)/L) in a case of vivax malaria. This is only the second case ever reported in the literature of such profound thrombocytopenia in a case of isolated P.vivax malaria.

Immune effector mechanisms of the nitric oxide pathway in malaria: cytotoxicity versus cytoprotection BJID
Nahrevanian,Hossein.
Nitric oxide (NO) is thought to be an important mediator and critical signaling molecule for malaria immunopathology; it is also a target for therapy and for vaccine. Inducible nitric oxide synthase (iNOS) is synthesized by a number of cell types under inflammatory conditions. The most relevant known triggers for its expression are endotoxins and cytokines. To date, there have been conflicting reports concerning the clinical significance of NO in malaria. Some researchers have proposed that NO contributes to the development of severe and complicated malaria, while others have argued that NO has a protective role. Infection with parasites resistant to the microbicidal action of NO may result in high levels of NO being generated, which could then damage the...

TLR2 and TLR4 mediated host immune responses in major infectious diseases: a review BJID
Mukherjee,Suprabhat; Karmakar,Subhajit; Babu,Santi Prasad Sinha.
Abstract During the course of evolution, multicellular organisms have been orchestrated with an efficient and versatile immune system to counteract diverse group of pathogenic organisms. Pathogen recognition is considered as the most critical step behind eliciting adequate immune response during an infection. Hitherto Toll-like receptors (TLRs), especially the surface ones viz. TLR2 and TLR4 have gained immense importance due to their extreme ability of identifying distinct molecular patterns from invading pathogens. These pattern recognition receptors (PRRs) not only act as innate sensor but also shape and bridge innate and adaptive immune responses. In addition, they also play a pivotal role in regulating the balance between Th1 and Th2 type of response...

Tarazona,Alonso Soto; Zerpa,Lely Solari; Mendoza Requena,Daniel; Llanos-Cuentas,Alejandro; Magill,Allan.
OBJECTIVE: To determine the sensitivity and specificity of the rapid diagnostic test OptiMAL® for diagnosis of Plasmodium vivax malaria. MATERIAL AND METHODS: We included all the patients who sought medical attention in the San Martin Pangoa Hospital, Junin, an area endemic for vivax malaria in Peru, between October and December 1998, who had fever during the previous 72 hours and who were older than 12 months. The gold standard for diagnosis was thick blood film microscopy. We determined the parasitemia rate for each of the positive slides. We calculated sensitivity, specificity, positive predictive value and negative predictive value of the test. RESULTS: We included 72 patients; 39 of them were positive for P. vivax by microscopic examination. The...

Acute respiratory distress syndrome due to vivax malaria: case report and literature review BJID
Lomar,André V.; Vidal,José E.; Lomar,Frederico P.; Barbas,Carmen Valente; Matos,Gustavo Janot de; Boulos,Marcos.
Severe pulmonary involvement in malaria has been frequently reported in cases of Plasmodium falciparum infection, but rarely in vivax malaria. Among the 11 previous cases of vivax-related severe respiratory involvement described in the literature, all except one developed it after the beginning of anti-malarial treatment; these appear to correspond to an exacerbation of the inflammatory response. We report the case of a 43-year-old Brazilian woman living in a malaria-endemic area, who presented acute respiratory distress syndrome (ARDS) caused by P. vivax before starting anti-malarial treatment. The diagnosis was made based on microscopic methods. A negative rapid immunochromatographic assay, based on the detection of Histidine Rich Protein-2 (HRP-2) of P....

Serum ascorbic acid concentration in patients with acute Falciparum malaria infection: possible significance BJID
Hassan,Garba Ibrahim; Gregory,Ubom; Maryam,Haruna.
Falciparum malaria infection is associated with significant destruction of erythrocytes. This leads to the release of toxic metabolic products, including oxidant compounds. We measured the serum concentration of the antioxidant, ascorbic acid, in 129 patients presenting with acute falciparum malaria infection and in 65 healthy individuals. None of the study subjects administered any form of ascorbic acid supplementation within one week prior to participation in this study. The mean serum ascorbate concentration in infected adult males (n = 49, age range 18-50 years) was found to be 2.02 ± 0.20 mg/dL, and it was 2.03 ± 0.24 mg/dL in infected adult females (n = 56, age range 18-50 years). These values were significantly greater than the serum ascorbate levels...

Genetic characterization and evolutionary inference of TNF-α through computational analysis BJID
Awasthi,Gauri; Singh,Suchita; Dash,A.P.; Das,Aparup.
TNF-α is an important human cytokine that imparts dualism in malaria pathogenicity. At high dosages, TNF-α is believed to provoke pathogenicity in cerebral malaria; while at lower dosages TNF-α is protective against severe human malaria. In order to understand the human TNF-α gene and to ascertain evolutionary aspects of its dualistic nature for malaria pathogenicity, we characterized this gene in detail in six different mammalian taxa. The avian taxon, Gallus gallus was included in our study, as TNF-α is not present in birds; therefore, a tandemly placed duplicate of TNF-α (LT-α or TNF-β) was included. A comparative study was made of nucleotide length variations, intron and exon sizes and number variations, differential compositions of coding to...
