Quantification of Vibrio penaeicida, the etiological agent of Syndrome 93 in New Caledonian shrimp, by real-time PCR using SYBR Green I chemistry ArchiMer
Goarant, Cyrille; Merien, F.
Shrimp farming is a small but growing industry in New Caledonia. Since 1993, "Syndrome 93" has been affecting New Caledonian shrimp farming industry every cold season, causing severe epizootic mortalities in grow-out ponds and significant losses. Highly pathogenic strains of Vibrio penaeicida are considered the etiological agent of the disease in Litopenaeus stylirostris. On one hand, studies demonstrated that healthy shrimp may carry V penaeicida for weeks with a high overall prevalence, regardless of any seasonal pattern or temperature conditions. On the other hand, larvae are free of V penaeicida and are also resistant to experimental infection. V penaeicida is frequently detected in incoming water pumped from the bays, which was shown, by a molecular...

Sequence polymorphism-based identification and quantification of Vibrio nigripulchritudo at the species and subspecies level targeting an emerging pathogen for cultured shrimp in New Caledonia ArchiMer
Goarant, Cyrille; Reynaud, Yann; Ansquer, Dominique; De Decker, Sophie; Merien, Fabrice.
In a previous study, we demonstrated the existence of an emerging cluster of Vibrio nigripulchritudo that proved to be associated with shrimp mortality events in New Caledonia. Using sequence polymorphisms evidenced in this previous MultiLocus Sequence Typing study, we developed two new quantitative PCR assays permitting the detection and quantification of V. nigripulchritudo at the genospecies level using SYBR Green I chemistry and at the emerging cluster level using Fluorescence Resonance Energy Transfer technology with hybridization probes. The use of this molecular diagnostic tool evidenced the colonization of the shrimp pond ecosystem by the pathogenic cluster at least at the onset of the disease. This new tool will allow better investigation of the...

What role for genomics in fisheries management and aquaculture? ArchiMer
Wenne, Roman; Boudry, Pierre; Hemmer-hansen, Jakob; Lubieniecki, Krzysztof P.; Was, Anna; Kause, Antti.
The development and application of genomics has been facilitated in a number of fields by the availability of new methodologies and tools, such as high throughput DNA sequencing and complementary DNA (cDNA) microarrays. Genomic tools are already used in research on commercially important fish and shellfish species. Thousands of expressed sequence tags (EST) are now available for some of these species, and the sequencing of complete genomes of tilapia, cod, salmonids, flatfishes, sea bass and Pacific oyster has been proposed. Microarray technology through simultaneous analysis of the expression of thousands of genes allows the identification of candidate genes involved in the function of multiple physiological, morphological and behavioural traits of...

Advancement and diversification of the aquaculture industry is reliant on the development of captive breeding and rearing protocols for novel fish species. Orbicular batfish (Platax orbicularis), a major emerging species in Polynesian aquaculture, live in brackish and marine waters around coral reefs, and are highly prized by Pacific Island communities for their high-quality meat. The present study describes the larval growth of P. orbicularis from hatching until 16 days after hatching (DAH) using meristics and thyroid hormone (T3) quantification. Our study highlighted that metamorphosis of P. orbicularis is critical to their production in aquaculture, as found for other species. Levels of the thyroid hormone T3 in P. orbicularis reached a peak at 12 DAH...

Molecular epidemiology of Vibrio nigripulchritudo, a pathogen of cultured penaeid shrimp (Litopenaeus stylirostris) in New Caledonia ArchiMer
Goarant, Cyrille; Reynaud, Yann; Ansquer, Dominique; De Decker, Sophie; Saulnier, Denis; Leroux, Frederique.
A collection of 57 isolates of Vibrio nigripulchritudo from either diseased or healthy shrimp and from shrimp farms environment was studied in order to gain a better understanding of the epidemiology of this pathogen, notably isolated from two distinct shrimp disease complexes. Molecular typing using two different techniques, arbitrarily primed PCR (AP-PCR) and multi-locus sequence typing (MLST), studied together with experimental pathology data allowed a relevant epidemiological insight into this possibly emerging pathogen. Additionally, results obtained with the two molecular typing techniques were congruent and allowed discriminating the strains associated with the "Summer Syndrome" from strains isolated from other contexts, especially the other shrimp...

Fisheries Science Collaborations: The Critical Role of the Community OceanDocs
Wilson, D.C..
The present paper is based on a collation of information, through both a literature review and an internet search, on North American programs that involve some kind of scientific collaboration between fishers and fisheries scientists. It identifies four basic models of such collaboration and offers examples of collaborative activities that seem to fit into each of them. The first model defers to the expertise of the scientist for all major decisions and creates a strong programmatic distinction between what is science, and shall be done by scientists, and what are other, related activities that can be carried out by others. Various kinds of tagging programs, fisher advisory boards, at-sea research collaboration and many other activities fit this model. The...

Studies on Tanzanian hypneaceae: seasonal variation in content and quality of Kappa-Carrageenan from Hypnea musciformis (Gigartinales : Rhodophyta) OceanDocs
Buriyo, A.S.; Mtolera, M.S.P..
Seasonal effects on yield and quality of kappa-carrageenan from the red alga Hypnea musciformis were investigated in samples collected from natural populations in Oyster Bay, Dar es Salaam during June 1996–May 1997. The mean annual carrageenan yield, gel strength (after treatment with 0.1 M KCl) gelling and melting temperature (± standard deviation) were 25.24 ± 4.44 % dry weight, 171.72 ± 41.42 g/cm2, 54.66 ± 3.12 ºC and 68.62 ± 0.60 ºC, respectively. Carrageenan yield and quality (gel strength) during the SE and NE monsoon seasons were not significantly different (t = 0.55, p > 0.05) and (t = 1.91, p > 0.05), respectively. The reported carrageenan yield and gel strength values were, respectively, about 50% and 40% those of carrageenan from...

Water Quality in the Integrated Mariculture Pond Systems (IMPS) at Makoba Bay, Zanzibar, Tanzania OceanDocs
Mmochi, A.J.; Mwandya, A.W..
The Makoba integrated mariculture pond system project (IMPS) has been on-going since 1998. Except for a period from mid 1999 to 2001, various water quality parameters, namely temperature, salinity, pH, dissolved inorganic nutrients and dissolved oxygen concentration and saturation, have been monitored continuously. In 2002, measurement of sediment oxygen demand (SOD) and consumption of oxygen by sediments was initiated. Dissolved oxygen concentration varied from the highest monthly average of 7.16 mg/l in October 1998, to 2.2 mg/l in March 2000 (r = –0.69). Nutrient concentrations remained moreor- less constant, except in the finfish ponds, where slight variations were recorded. The lowest concentration of ammonium-nitrogen (3.02 mg-at N/l) was recorded in...

Tosta,Gitonilson Antonio Moreira; Albinati,Ricardo Castelo Branco; Carvalho,Gleidson Giordano Pinto de; Carvalho,Jaciane Vergne de; Souza Filho,José Jerônimo de; Vasconcelos,Fabíola Rios; Kuhnen,Vanessa Villanova; Sanches,Eduardo Gomes.
Abstract: The objective of this work was to evaluate different feeding rates on the productive performance of cobia (Rachycentron canadum) grown in brackish waters. For the experiment, 1,600 juveniles were distributed in 16 enclosures installed in an estuarine pond. Four different feeding rates were evaluated: 1, 3, 5, and 7% live weight (LW) per day. The following performance parameters were assessed: survival, final biomass, specific growth rate, daily weight gain, apparent feed conversion, protein efficiency rate, lipid efficiency rate, and hepatosomatic index. Survival was greater than 80% at all feeding rates. The most efficient feed conversion rate was obtained with the feeding rate of 5% LW per day, which also resulted in better values for the...

Performance of common snook subjected to different feeding frequencies and rates using automatic feeders PAB
Herrera,Leandro Amaral; Santos,Ana Paula dos; Sousa,Otávio Mesquita de; Kuhnen,Vanessa Villanova; Sanches,Eduardo Gomes.
Abstract: The objective of this work was to evaluate the growth performance of common snook (Centropomus undecimalis) subjected to different feeding rates and frequencies using automatic feeders. Two experiments of 60 days each were carried out. In the first one, 20 fishes per tank were distributed in three feeding rates of: 5, 10, and 15% body weight per day. In the second experiment, 20 fishes per tank were distributed in nine tanks, in three feeding frequency: once a day, three times a day, and six times a day. Water parameters were maintained at the recommended level for the species. Final weight, body weight gain, specific growth rate, and feed conversion rate did not differ significantly between the 10 and 15% feeding rates. However, both rates...

Maintenance of Octopus vulgaris Type II paralarvae in an estuarine area R. Bras. Zootec.
Bastos,Penélope; Vieira,Graziela Cleuza.
ABSTRACT We assessed the survival of paralarvae kept in a floating wooden box attached to an oyster extensive cultivation system with no extra food supply. A total of 7700 newly hatched paralarvae were maintained in a 10.5-L floating box (7 cm height × 30 cm width × 50 cm length) covered with a 180-μm mesh net for 14 days with no extra food supply. Skin damages and tentacle deformities were observed in 43% of the paralarvae at 14 days after hatching (DAH). The survival rate was 64.7% at 7 DAH and 42.8% at 14 DAH. The floating box is a promising structure for culturing O. vulgaris paralarvae in an extensive system.
