Cloning, mapping and molecular characterization of porcine progesterone receptor membrane component 2 (PGRMC2) gene Genet. Mol. Biol.
Chen,Congying; Sargent,Carole; Quilter,Claire; Yang,Zhuqing; Ren,Jun; Affara,Nabeel; Brenig,Bertram; Huang,Lusheng.
Progesterone plays an important role in sow reproduction by stimulating classic genomic pathways via nuclear receptors and non-genomic pathways via membrane receptors such a progesterone receptor membrane component 2 (PGRMC2). In this work, we used radiation hybrid mapping to assign PGRMC2 to pig chromosome 8 and observed that this receptor has two transcripts in pigs. The full-length cDNA of the large transcript is 1858 bp long and contains a 669-bp open reading frame (ORF) encoding a protein of 223 amino acids. The shorter transcript encodes a protein of 170 amino acids. The porcine PGRMC2 gene consists of three exons 446 bp, 156 bp and 1259 bp in length. The promoter sequence is GC-rich and lacks a typical TATA box. Several putative cis-regulatory DNA...

Microsatellite variability analysis in farmed catfish (Ictalurus punctatus) from Tamaulipas, Mexico Genet. Mol. Biol.
Perales-Flores,Laura E.; Sifuentes-Rincón,Ana María; León,Francisco J. García de.
Analysis of cultured catfish from six farms in Tamaulipas, Mexico was achieved using a combination of microsatellite PCR analysis and semiautomatic fluoresce-based detection, in order to provide a first assessment of the genetic variability on cultured catfish in Mexico. Five microsatellites showed extensive polymorphism with allele numbers ranging from 10 and 20. Overall observed heterozygosity at each locus ranged between 0.76 and 0.91 and the average polymorphic information content (PIC) for the five loci was 0.811, indicating that these loci can be used for studies of paternity identification, linkage and population genetics. On the basis of the F ST values (F ST = 0.03829; p = 0.00000) it appears that there was a small amount of genetic...

Genetic similarity between coriander genotypes using ISSR markers Horticultura Brasileira
Melo,Roberto de A; Resende,Luciane V; Menezes,Dimas; Beck,Ana Paula A; Costa,José Carlos da; Coutinho,Alisson E; Nascimento,Ana Verônica S do.
With the development of new cultivars, a precise genetic characterization is essential for improvement programs or for cultivar registration and protection. Molecular markers have been complementing the traditional morphological and agronomic characterization techniques because they are virtually unlimited, cover the whole genome and are not environmentally influenced. Genetic characterization constitutes the basis for studies involving estimates of genetic similarity. Therefore, the objective of the present study was to evaluate the genetic similarity between ten coriander genotypes (nine cultivars and one line) using ISSR markers. The cultivars used were: Americano, Asteca, Palmeira, Português, Santo, Supéria, Tabocas, Tapacurá, Verdão and the...

Genetic divergence in basil cultivars and hybrids Horticultura Brasileira
Alves,Rodrigo P; Silva,Ana Veruska C; Almeida-Pereira,Camila S; Costa,Tatiana S; Alvares-Carvalho,Sheila V; Arrigoni-Blank,Maria de Fatima; Blank,Arie F.
ABSTRACT Basil is an aromatic herb that stands out for its economic importance. It is consumed in natura and used to obtain essential oil. The cultivation of this species in several regions of the world has allowed variations by natural crosses and euploidy, leading to the wide genetic variability found nowadays. Considering the importance of this species, we aimed to analyze the genetic diversity of 27 basil genotypes using ISSR molecular markers. Fourteen primers were employed for DNA amplification, resulting in 86% polymorphism. Based on the Jaccard’s dissimilarity index, the highest index (0.80) was observed between the individuals BAS001 and BAS012, while the lowest index (0.18) was detected between the genotypes BAS014 and BAS015. The genetic...

Microbial communities of recirculating aquaculture facilities: interaction between heterotrophic and autotrophic bacteria and the system itself ArchiMer
Michaud, Luigi.
Recirculating Aquaculture Systems (RAS) may be considered an alternative to pond aquaculture technology (by consuming lower amounts of water producing similar yields of crop) or can be integrated in a sort of "production chain" with net pen systems. The interest in RAS is due to their intrinsic advantages like the reduction of land and water requirements, the high degree of environmental control allowing year-round growth at optimum rates and the possibility to produce in close proximity to markets. Moreover, the high level of control improves risk management linked with bacterial and parasitic infections and predator pests. Finally, waste production (both particulate and soluble) can be controlled and minimized contributing to make these systems...

Rescue of a non-viable accession and rapd analysis of recovered plants of Arachis retusa PAB
Gagliardi,Rachel Fatima; Pacheco,Georgia Pereira; Oliveira,Carlos Alberto; Carneiro,Leonardo Alves; Valls,José Francisco Montenegro; Vieira,Maria Lucia Carneiro; Mansur,Elisabeth.
In vitro regeneration of Arachis retusa was examined for the purpose of germplasm renewal and conservation. Random amplified polymorphic DNA (RAPD) fingerprinting was used to evaluate the genetic stability of plants derived from embryo axes and apical segments. Ten arbitrary decamer primers were screened and five of them were selected. Ninety genomic regions were evaluated, with an average of 18 loci per clone. All amplified segments were monomorphic. The results indicate that recovered plants are genetically stable at the assessed genomic regions and that both regeneration processes are suitable for in vitro germplasm preservation of Arachis species.

Development of an SSR-based identification key for Tunisian local almonds Scientia Agricola
Gouta,Hassouna; Ksia,Elhem; Buhner-Zaharieva,Tatiana; Mliki,Ahmed; Gogorcena,Yolanda.
Ten simple sequence repeat (SSR) loci were used to study polymorphism in 54 almond genotypes. All genotypes used in this study originated from almond-growing areas in Tunisia with different climatic conditions ranging from the sub-humid to the arid and are preserved in the national collection at Sidi Bouzid. Using ten SSR, 130 alleles and 250 genotypes were revealed. In order to develop an identification key for each accession, the data were analysed separately for each microsatellite marker. The most polymorphic microsatellite (CPDCT042) was used as a first marker. Two microsatellite loci (CPDCT042 and CPDCT025) were sufficient to discriminate among all accessions studied. Neighbour-joining clustering and principal coordinate analysis were performed to...

Genetic divergence among and within Colletotrichum lindemuthianum races assessed by RAPD Trop. Plant Pathol.
Talamini,Viviane; Souza,Elaine A.; Pozza,Edson A.; Silva,Gilvan F.; Ishikawa,Francine H.; Camargo Júnior,Osnil A..
Genetic divergence within and among races of Colletotrichum lindemuthianum was determined using RAPD markers. In addition to the different races of the fungus three isolates of the sexual stage of Colletotrichum lindemuthianum (Glomerella cingulata f.sp. phaseoli) were included in this study. The band patterns generated using 11 primers produced 133 polymorphic bands. The polymorphic bands were used to determine genetic divergence among and within the pathogen races. The isolates analyzed were divided into six groups with 0.75 relative similarity. Group VI, formed by three isolates of the sexual phase of Colletotrichum lindemuthianum, was the most divergent. Races previously determined using differential cultivars did not correlate with the results...

Molecular and pathogenic variability of Drechslera isolates from oats Trop. Plant Pathol.
MEHTA,Y. R..
Severe epidemics of leaf blotch and black leaf spot of oat (Avena sativa) caused by Drechslera avenae and Drechslera sp., respectively, are frequently observed in the State of Paraná, Brazil. Although some morphological differences between the isolates causing two different symptoms were noticed, the genetic relationship between them was not clear. Twenty-four isolates of D. avenae and Drechslera sp, collected between 1996-98, were assessed for the genetic variability by molecular and pathogenic analyses. The amplification products using primer pair ITS4/ITS5 showed a fragment length of approximately 600 bp for all the isolates except for one black spot isolate, where the fragment length was approximately 550 bp. Restriction enzymes Hinf I and Taq I, that...
