In vitro plant regeneration system for common bean (Phaseolus vulgaris): effect of N6-benzylaminopurine and adenine sulphate Electron. J. Biotechnol.

A method for regeneration of the commercially important common bean (Phaseolus vulgaris ) using N6-benzylaminopurine(BAP) and adenine sulphate (AS) was established. Embryogenic axes of the Costa Rican common bean cultivars Bribrí, Brunca, Guaymí, Huetar and Telire were cultured on Murashige and Skoog medium supplemented with 100 mgl-1 myo-inositol, 1 mgl-1 thiamine, 30 gl-1 sucrose, BAP (0, 5 and 10 mgl-1), AS (0, 20 and 40 mgl-1) and 8 gl-1 agar. Regardless of the concentration of BAP and AS in the induction medium, the number of shoots and leaves differed significantly among the common bean cultivars evaluated. The aim of this review is to critically analyze the role of silver nitrate (AgNO3) in modulating plant growth and development. In recent years, basic studies on ethylene regulation opened new vistas for applied research in the area of micro-propagation, somatic embryogenesis, in vitro flowering, growth promotion, fruit ripening, and sex expression. Silver nitrate has proved to be a very potent inhibitor of ethylene action and is widely used in plant tissue culture. Few properties of silver nitrate such as easy availability, solubility in water, specificity and stability make it very useful for various applications in exploiting plant growth regulation and morphogenesis in vivo and in vitro. Intake of high levels of vitamin A and polyunsaturated fatty acids during different developmental periods modifies the expression of morphogenesis genes in European sea bass (Dicentrarchus labrax) ArchiMer

The effect of the feeding period on larval development was investigated in European sea bass larvae by considering the expression level of some genes involved in morphogenesis. Larvae were fed a control diet except during three different periods (period A: from 8 to 13 d post-hatching (dph); period B: from 13 to 18 dph; period C: from 18 to 23 dph) with two compound diets containing high levels of vitamin A or PUFA. European sea bass morphogenesis was affected by these two dietary nutrients during the early stages of development. A study of the sexual physiology of penaeid prawns essentially based upon Penaeus japonicus is performed in both descriptive and experimental ways, using the controlled rearings conducted at the Centre Oceanologique de Bretagne (CNEXO-COB), Brest. Sexual morphogenesis is described. This preliminary study enables definition of simple criteria for determination of the stage of genital development of the prawns during their growth. The neuroendocrine control of the genital system development is studied, using eyestalks ablations and male eyestalks extracts injection in prawns belonging to the same sex. The presence of two (or at least one) eyestalks is required for inducing a complete, monoecdysial metamorphosis in Palaemonetes varians . This induction is performed, at 20 degree C, in about 20 hours situated on either side of the third larval moult when metamorphosis occurs at the fourth one. The mechanism of induction is discussed.

Problemes de morphogenese cuticulaire chez les crustaces ArchiMer

The crustacean integument is an excellent model for the study of cuticular morphogenesis, at different organization levels. At the supermolecular level, subunits of chitin and protein assemble and show the organization of a stabilized liquid crystal. At higher levels corresponding to cells and cells populations, several controls are involved in the fine architecture of the epicuticle, in the mineral distribution and in characters which concern the whole animal, such as isometric or allometric growth.

Heat shocks at 40 degree were delivered to Artemia at the end of embryonic development and at the beginning of postembryonic development and their effects on subsequent development examined. Heat shocks delivered during preemergence development have no effect on subsequent development. Heat shocks delivered to newly hatched nauplii induce anomalies on the morphogenesis of thoracic and genital appendages. The induction of morphological anomalies by thermal conditions prevailing during early larval life makes it easier to integrate the morphological effects previously induced by nutritional conditions imposed to older feeding stages.

Mise en place de l'epicuticule chez le crabe Carcinus maenas ArchiMer

The epicuticle of the crab Carcinus maenas consists in three distinct layers: the fibrous surface coat, the cuticulin layer and the inner epicuticle. Short segments of the cuticulin layer are first deposited at the top of dense plaques of the ectodermal cell membrane. Their assembling gives rise to a five-layered continuous structure of about 37 nm thick. Afterwards, the inner epicuticle is added inside close to the cuticulin layer but dense plaques are not concerned with the phenomenon. The deposition of the fibrous surface coat outside to the cuticulin layer takes place only during late postmolt stages (D2-D3). In mineralized cuticles, this typical epicuticle structure persists during all intermolt long.

The epithelial cells of the ovary secondary follicles have been studied in the prawn Macrobrachium rosenbergii using electron microscope and horseradish peroxidase. A network of circomvolutioned and anastomosed tubules, measuring about 0.15 mu m in diameter, communicates with the extracellular space. This tubular network is a temporary structure which extensively increases the permeability of the follicular epithelium.

In the Present study, the complete cDNA fragment encoding a precursor of endothelin-1, the preproendothelin-1 (PPET1), was cloned by RACE-PCR from European sea bass. A new analysis within the framework of developmental genetics provides both raw data and theoretical support to the “old” morphology and suggests a new, more predictive, approach to the concept of homology. We distinguish between “positional homologues” and “structural homologues” as independent components of the more general concept of homology. We discuss some general patterns seen in the anatomy of animals and in their morphogenesis. Slack et al. Observations by Scanning Electron Microscopy of Surface Events during Encystment and Excystment in Gastrostyla steinii (Cliophora, Hypotrichida) OAK

The surface of the protozoan Gastrostyla steinii undergoes profound morphological changes during the encystment and excystment processes. Resorption of the various ciliary structures occurs progressively during differentiation of the free living cell into the resting cyst. In ciliar disassembly, layers of the cyst wall are deposited on the surface of the encysting organism. At excystation, the hatching trophozoite emerges from the protective wall through a cleft after the formation of the ciliary organelles.

The ontogeny of the first stages of development of hake (Merluccius hubbsi)considering yolksac larvae, preflexion larvae, larvae in flexion, postflexion larvae, transformation larvae and posttransition juveniles, is described. Fish larvae and posttransition juveniles were collected in the north Patagonian area, between 42° and 46°S, from surroundings of the coast to the 100 m isobath (Argentine Sea, SW Atlantic). Se presentan datos con respecto a la forma de vida y del ambiente de 13 mesogastropodos superiores. Los huevos de Strombus gigas, S. pugilis, Polinices lacteus, P. hepaticus, Natica livida, N. canrena, Morum oniscus, Cypraecassis testiculus, Charonia variegata, Cymatium nicobaricum, C. pileare, Bursa cubaniana y Tonna galea se describen en detalle y se caracterizan las larvas recién nacidas. En la mayoría de las especies mencionadas se pudo determinar el tiempo de desarrollo desde el huevo hasta su nacimiento. Los huevos aquí descritos son comparados con lo que se conoce de la literatura, según sus caracteres morfológicos se distinguen seis tipos de cápsulas y masas de huevos.

Somatic embryogenesis and plant regeneration in elite clones of Theobroma cacao PAB

The objective of this work was to evaluated a procedure for somatic embryogenesis and regeneration of cacao (Theobroma cacao L.) elite clones. Petal explants from cacao clones TSH 565 and TSH 1188 were cultured on PCG and SCG-2 media, for calli growth. Somatic embryos were formed on the surface of embryogenic calli after transfer to embryo development (ED) medium. Clone TSH 565 showed a higher embryogenic potential than TSH 1188. The best combination of carbon source for embryo induction in ED medium was genotype-specific. Embryogenic callus formations increased in micropore tape-sealed Petri dishes, irrespective of cacao genotype. Mature somatic embryos were successfully converted into plantlets.

Tissue culture parameters in sweet orange cultivars PAB

The objective of this work was to establish tissue culture parameters for gene transfer in sweet orange cultivars. Epicotyl explants with different ages were cultured with 6-benzylaminopurine (BAP), kanamycin and hygromycin. Shoots were cultured with alpha-naphthaleneacetic acid (NAA) alone or in combination with indole-3-butyric acid (IBA). The requirement of BAP for shoot development was genotype-specific. Epicotyl explants from 35-day-old seedlings produced significantly more shoots per explant in 'Pêra'. Kanamycin inhibited shoot regeneration for the most cultivars. The percentage of shoots that produced roots in 'Pêra' was significantly higher in medium with NAA and IBA than with NAA alone.

A reliable protocol for plant regeneration of Mullein Nightshade (Solanum donianum Walp.) was developed from in vitro leaf explants. They were cultured on Murashige and Skoog semisolid medium, supplemented with several combinations of zeatin riboside and 1-naphthaleneacetic acid for shoot regeneration. The formulation that originated most shoots per explant was 8.5 µM zeatin riboside in absence of 1-naphthalene acetic acid. Explants with shoots were transferred to Murashige and Skoog medium, with half the normal salt concentration and without plant growth regulators for elongation. This is a comparative study of the ultrastructural characteristics of the cells involved in the organogenesis in vitro of Bauhinia forficata Link (indirect system) and Glycine max (L.) Merrill (direct system). B. forficata calli after 30 days culture and G. max meristemoids after 10 days culture were prepared for ultrastructural analysis using conventional methods. Concentrically arranged rough endoplasmic reticulum (RER) and plastids containing starch grains were seen during G. max and B. forficata organogenesis. The amitotic process, the presence of plastids around the nucleus and nuclear envelope with conspicuous pores were found in B. forficata.
