Attempting to improve reproduction performance and ichthyo prophylaxis, this study evaluated the effects of maintaining silver catfish (Rhamdia quelen) broodstock in different saline concentrations (0, 2, 4, 6 and 8‰) on gametes quality and reproductive viability. The results showed that sperm percent motility did not change between 0 and 4‰, but it was reduced at 6‰, and sperm became immotile at 8‰ salinity. Sperm motility time was increased (almost five fold) at 6‰. Salinities up to 4‰ prevented fertilization and hatching, proving their deleterious effects on oocytes and embryos. Therefore, media up to 4‰ salinity may be an alternative for icthyo prophylaxis, although fertilization and incubation must be done in freshwater medium |

Genus Salmonella bacteria are among the major pathogenic microorganisms in food. This bacterium pathogenicity is related to a number of virulence factors, among which its flagella. Flagellum expression is one of the virulence factors modulated by Quorum Sensing. Epinephrine produced by mammals uses the same signaling pathway of the 3 bacteria autoinducer. This study evaluated the effect of molecules inducer (epinephrine) and autoinducers (autoinducer 2 and autoinducer 3) and their association with the motility, growth and expression genes flhC, fliA, fliY, motA, motB e fliC of Salmonella Typhimurium (ST). Initially, ST was inoculated in BHI. Then, motility assays, growth curves and gene expression were performed by testing different concentrations of... |

High quality of sperm is essential to a high fertilization rate, especially post- cryopreservation. Assessment of sperm integrity, motility and energy reserves before cryopreservation is necessary for selection of milt with optimal fertilizing potential. We describe the effect of cryopreservation on the quality of black-lip pearl oyster, Pinctada margaritifera var. cumingii sperm. Evaluated quality indices of fresh and frozen/thawed P. margaritifera spermatozoa, included morphology, ultrastructure and motility characteristics relative to the energy content (ATP) and its capacity to be sustained by mitochondrial respiration. Morphology and ultrastructure were quantitatively evaluated using images obtained by optical microscopy assisted by the Image J... |

Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). At pH 6.4, percentage of motile spermatozoa was three times higher when they were activated in SW containing 30 mM NH4Cl, which alkalinizes internal pH (pHi) of spermatozoa, compared... |

Fertilization of the oocyte is a critical process of sexual reproduction depending among other factors on spermatozoa functionality. Mitochondria participate in many crucial processes for spermatozoa motility and fertilizing ability. The Pacific oyster, Crassostrea gigas, is characterized by a high inter-individual variability of its reproductive success which has been shown to be partially due to the variability of gamete quality. The present study explored spermatozoa characteristics such as: i) relationships among mitochondrial functionality (ATP content, mitochondrial membrane potential (MMP), reactive oxygen species production); sperm viability (percentages of live, dying, and dead); and motility; and ii) their involvement in inter-oyster variability... |

Many recent ecotoxicological studies suggest a relationship between freshwater contamination and increasing abundances of motile diatoms (potentially able to move). The capacity to escape would present advantages to species in polluted environments. However, actual motility as a response to toxicants had not been described and required experimental validation. We designed a specific experiment to assess how a field-isolated diatom (Gomphonema gracile) distributes energy to in situ resistance (increased population growth or photosynthesis) and escape (behavioral changes), when exposed to increasing concentrations of the herbicide metolachlor. We report here the dose-time dependent responses of G. gracile populations. They coped with low contamination by... |

Changes in sperm features during the movement phase are especially interesting to study in external fertilization species whose sperm duration movement is long because this implies a significant adaptation of moving cells to the external medium. This study describes the changes in tetraploid Pacific oyster sperm characteristics in relation to time post activation. Sperm individually collected on three tetraploid males were activated in seawater. Their features were analysed over a 24 h period and compared to a sperm pool collected on three diploid males as a reference. The percentage of motile spermatozoa, the intracellular ATP content, and the fine structure of spermatozoa were studied in relation to time post activation. Furthermore, the fertilisation... |

After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. The cost of such 'hyperactivity' is a very rapid consumption of intracellular ATP that outstrips the supply.... |

Sperm cryopreservation is a widely employed technique that promotes alternative techniques to contribute to broodstock management or restoration programs for species of commercial interest, endangered species or species with an interesting genotype. The preservation of genetic material from improved stocks or from the original population is extremely important for the oyster aquaculture industry to prevent the potential impacts of epidemic diseases and natural disasters. The Portuguese oyster, Crassostrea angulata, was the most important species commercialized by the shellfish industry. However, inadequate management of this industry and pathology occurrences resulted in a significant decrease in natural populations. For this reason, in this work a... |

Ten healthy male (boar) of large white pigs aged between 6.0 and 18.0 months, were used for this study. Spermiogram and morphological characteristics were studied in testes and in the different parts of the epididymis of the boar. The objective was to study the changes that occur in testes and epididymis during epididymal transit. Progressive motile spermatozoa were first observed in the testes (6-10%) in the caput (20-30%), corpus (40-50%) and highest at the caudal epididymis (85%). Cytoplasmic droplets were lost as the spermatozoa moved up the epididymis. The caput epididymis had a value of 20.05% of abnormal sperm cell, which is different significantly (p<0.5) from 15 80 % of the caudal epididymis. The total abnormal sperm cells in corpus are also... |

Goat spermatozoa were incubated in vitro in Tris-citrate buffer, pH 7.2, containing 0, 40, 80, or 160 mug of Naja haje venom/mL buffer for 4 hours. During incubation, the percentages of sperm motility were decreased, while percentages of dead spermatozoa were increased in a time and concentration-dependent manner. The effect of venom concentrations on the ultrastructure of incubated spermatozoa was examined hourly by scanning (SEM) and transmission (TEM) electron microscopy. SEM results showed plasma membrane wrinkling at heads of some spermatozoa after 2 h incubation with 40mug venom. Most spermatozoa suffered membrane wrinkling after 4 h incubation. However, incubation with 80 mug venom caused membrane fractures in most sperm heads after 1 h incubation.... |

This study evaluates the effects of extender osmolality and composition on the cooling of Prochilodus lineatussperm. Sperm was diluted in six extenders: two compositions (powdered coconut water(tm) = ACP; Beltsville Thawing Solution(tm) = BTS) x three osmolalities (285, 325, and 365 mOsm/kg) plus an undiluted control, and stored at 6-8°C. Motility rate and velocities (curvilinear, straight line, and average path) were determined every other day. Osmolality did not affect the quality of cooled sperm, thus data were pooled. Motility was higher on d 0 compared to the other days and diluted samples (85-90%) yielded higher motility than control (75%). On d 2, motility was higher in BTS-diluted samples and control, but on d 4 and 6, control yielded the highest... |

Cryoprotectant solutions are used to protect the sperm from alterations caused by the low temperature in the cryopreservation process. We evaluated the quality of Colossoma macropomum semen after freezing, using dimethyl sulfoxide (DMSO) as a cryoprotectant, combined with two extender solutions (T1 - Solution 1: Glucose 90.0 g/L, Sodium Citrate 6.0 g/L, EDTA 1.5 g/L, Sodium Bicarbonate 1.5 g/L, Potassium Chloride 0.8 g/L, Gentamycin Sulphate 0.2 g/L, and T2 - Solution 2: Glucose 90.0 g/L, ACP(r)-104 10.0 g/L). Motility rate and motility time did not differ between T1 and T2 and were lower than fresh semen. The number of normal sperm was significantly different in treatments T1 (15.1%) and T2 (21.9%), and both showed a reduction in the percentage of normal... |

This study was designed to investigate gastrointestinal dysfunctions, especially the effects of gastrointestinal prokinetic agents on digestive disorders involving lowered motility. The effects of α2-adrenergic agonists for treating acute abdominal disorders involving intestinal torsion in horses were also investigated. The results are as follows: (1) It appeared that the functions of migrating contractions (MC) in horses were to transfer the contents of the digestive tract as soon as possible to the cecum and colon, in addition to maintaining homeostatis in the digestive tract. (2) Motilin was seen to be involved in the regulation mechanism of intestinal motility in the horse as well as in humans and dogs, and brought about MC. (3) The intravenous... |

Abstract: The objective of this work was to evaluate the efficiency of extenders solutions in tambaqui (Colossoma macropomum) semen cooling at 6°C. The treatments consisted of semen dilutions with three extenders - BTS, HBSS, and ACP-104 -, and a control with undiluted semen. Cooled semen was evaluated by 24, 48, 72, 96, and 120 hours. There were differences for the duration of motility and fertilization rates among the semen dilutions. Motility duration proved a highly variable evaluation after 72 hours. Dilution testing can be performed by the three products, and cooling storage for 72 hours is most effective with BTS extender. |

ABSTRACT By using informetric tools, we evaluated the contribution and impact of the open source computer-assisted sperm analysis (CASA) software on sperm studies, especially pertaining to fish. The article was cited 125 times between 2007 and 2014 in four publication types (articles, doctoral dissertations, master’s theses, and books). Among articles, the number of citations increased at rate of three citations per year and the calculated h-index was 17. Of the studies that cited Wilson-Leedy and Ingermann (2007), fish were the main group of organisms studied (61 articles); of these, 34 articles used the open source CASA, covering 23 marine or freshwater species. Studies conducted in 12 countries were published in 21 journals. Four ways of knowledge... |
