The role of phytochromes in cadmium stress responses in tomato Bragantia
Gaion,Lucas Aparecido; Lorevice,Paulo Guilherme; Monteiro,Carolina Cristina; Gavassi,Marina Alves; D’Amico-Damião,Victor; Gratão,Priscila Lupino; Gasparino,Eduardo Custódio; Carvalho,Rogério Falleiros.
ABSTRACT It is well known that phytochromes mediate a wide range of photomorphogenic processes in plants. In addition, many studies have demonstrated the involvement of phytochromes as part of abiotic stress signaling responses. However, little is known about cadmium (Cd) stress regulation by phytochromes. Thus, in this study, we used the phyA (far red-insensitive; fri), phyB1 (temporary redinsensitive; tri) and phyB2 (phyB2) tomato (Solanum lycopersicum L.) mutants to investigate the roles of these three phytochromes on Cd stress responses. The plants were grown over a 21-d period in the presence of Cd. We evaluated plant growth, Cd and chlorophyll content and anatomical changes in the leaves. The results indicated that all genotypes were affected by Cd...

Improvement of xylanase production by a parasexual cross between Aspergillus niger strains BABT
Loera,Octavio; Córdova,Jesús.
A diploid strain (D4) isolated via parasexual recombination between two Aspergillus niger xylanase overproducing mutants was characterised in terms of enzyme production and catabolite repression by glucose. This strain increased xylanase production (607 nkat/ml), which was nearly 100% higher than titers achieved by the wild type strain (305 nkat/ml) and 28% higher than the best mutant used to induce parasexual cycle. Diploid D4 was also less sensitive to carbon catabolite repression by glucose, since xylanolytic activity was detected under conditions normally repressing production by the wild type strain. No decrease in maximal xylanase levels was observed in the presence of glucose for diploid D4.

Increased expression of keratinase and other peptidases by Candida parapsilosis mutants BJMBR
Duarte,T.R.; Oliveira,S.S.; Macrae,A.; Cedrola,S.M.L.; Mazotto,A.M.; Souza,E.P.; Melo,A.C.N.; Vermelho,A.B..
Keratinases are enzymes of great importance involved in pathogenic processes of some fungi. They also have a widespread ecological role since they are responsible for the degradation and recycling of keratin. On the one hand, studying them furthers our knowledge of pathogenicity mechanisms, which has important implications for human health, and on the other hand, understanding their ecological role in keratin recycling has biotechnological potential. Here, a wild-type keratinolytic Candida parapsilosis strain isolated from a poultry farm was treated with ethyl methanesulfonate in order to generate mutants with increased keratinase activity. Mutants were then cultured on media with keratin extracted from chicken feathers as the sole source of nitrogen and...

DNA polymorphism and total protein in mutants of Metarhizium anisopliae var. Anisopliae (Metsch.) Sorokin strain E9 BJM
Freire,Laurineide Lopes de Carvalho; Costa,Ana Bolena Lima da; Góes,Larissa Brandão; Oliveira,Neiva Tinti de.
Five mutants (MaE10, MaE27, MaE24, MaE41 e MaE49) of Metarhizium anisopliae wild strain E9 were analysed for DNA profile through the RAPD technique and for changes in total protein content by spectrophotometry, polyacrylamide gel electrophoresis and densitometry. The pattern of RAPD markers showed genetic polymorphism among the strains: out of twenty primers seven were selected, producing 113 bands. Forty seven bands were present in all strains (41.6% of monomorphic bands) and 66 showed polymorphism (58.4%). The mean coefficient of similarity among all strains was 0.75 (75%). The total protein content varied, staining in the interval of 6.0-8.0 µg/µl. The electrophoresis analysis, through zymogram and protein fraction profiles by densitometry, allowed the...

Lysine catabolism: flow, metabolic role and regulation Braz. J. Plant Physiol.
Fornazier,Ricardo Francisco; Azevedo,Ricardo Antunes; Ferreira,Renato Rodrigues; Varisi,Vanderlei Aparecido.
Lysine is an essential amino acid, synthesized in plants in the aspartic acid pathway. The lysine catabolism is performed by the action of two consecutive enzymes, lysine 2-oxoglutarate reductase (LOR) and saccharopine dehydrogenase (SDH). The steady state of lysine is controlled by both, synthesis and catabolism rates, with the final soluble lysine concentration in cereal seeds a direct result of these processes. In the last 40 years, the enzymes involved in lysine biosynthesis have been purified and characterized from some plant species such as carrot, maize, barley, rice, and coix. Recent reports have revealed that lysine degradation might be related to various physiological processes, for instance growth, development and response to environmental...

Analysis of the SHP2 enhancer for the use of tissue specific activation tagging in Arabidopsis thaliana Genet. Mol. Biol.
Chalfun-Junior,Antonio; Mes,Jurriaan J.; Busscher,Marco; Angenent,Gerco C..
Activation tagging is a powerful tool to identify new mutants and to obtain information about possible biological functions of the overexpressed genes. The quadruple cauliflower mosaic virus (CaMV) 35S enhancer fragment is a strong enhancer, which is most commonly used for this purpose. However, the constitutive nature of this enhancer may generate lethal mutations or aberrations in different plant organs by the same overexpressed gene. A tissue-specific activation tagging approach may overcome these drawbacks and may also lead more efficiently to the desired phenotype. For this reason the SHATTERPROOF2 (SHP2) promoter fragment was analysed for enhancer activity. The SHP2 gene is involved in dehiscence zone development and expressed during silique...

Effect of soybean ureases on seed germination and plant development Genet. Mol. Biol.
Rechenmacher,Ciliana; Wiebke-Strohm,Beatriz; Oliveira-Busatto,Luisa A. de; Polacco,Joseph C.; Carlini,Célia R.; Bodanese-Zanettini,Maria H..
Abstract Urease catalyzes the hydrolysis of urea to ammonia and carbon dioxide. The ammonia (nitrogen (N) product of urease activity) is incorporated into organic compounds. Thus, urease is involved in N remobilization, as well as in primary N assimilation. Two urease isoforms have been described for soybean: the embryo-specific, encoded by the Eu1 gene, and the ubiquitous urease, encoded by Eu4. A third urease-encoding gene was recently identified, designated Eu5, which encodes the putative protein product SBU-III. The present study aimed to evaluate the contribution of soybean ureases to seed germination and plant development. Analyses were performed using Eu1/Eu4/Eu5-co-suppressed transgenic plants and mutants of the Eu1 and Eu4 urease structural genes,...

New wheat genotypes tolerant to aluminum toxicity obtained by mutation induction PAB
Tulmann Neto,Augusto; Alves,Marcelo Correa; Camargo,Carlos Eduardo de Oliveira; Castro,Jairo Lopes de; Ferreira Filho,Wilson Penteado.
Seed from the sensitive wheat (Triticum aestivumL.) cultivar Anahuac was treated to gamma-ray irradiation and eleven Al3+ tolerant mutants selected. The objective was to compare these mutants to the original Anahuac and to the tolerant wheat cultivars IAC-24 and IAC-60 from 1994 to 1996 in acid (Capão Bonito) and limed (Monte Alegre do Sul) soil field trials, in the State of São Paulo, Brazil. Grain yield and agronomic characteristics were analyzed. All the mutant lines yielded higher than the sensitive Anahuac cultivar in the acid soils of Capão Bonito. Under limed soil conditions, 10 mutants had a similar yield to the original sensitive cultivar and one a lower yield. The majority of the mutants were similar in yield to the tolerant cultivars IAC-24 and...

Nitrate levels and stages of growth in hypernodulating mutants of Lupinus albus. I. N2 fixation potential Rev. Microbiol.
Burity,Hélio Almeida; Chamber-Perez,Manuel; Lyra,Maria do Carmo Catanho Pereira de; Figueiredo,Márcia do Vale Barreto.
This work aimed to evaluate physiological parameters, nodulation response and N2 fixation rate in mutants of Lupinus albus in comparison with the standard Multolupa cultivar. Two nitrate levels (0 and 5mM) and two evaluation periods (7 and 10 weeks) were used. Significant differences were observed among genotypes, in relation to fresh nodule weight, nitrate levels and growth stages. The overall average for nitrate level differed between them where 5mM severely inhibited the number of nodules, reaching a 49.5% reduction in relation to treatment without nitrate. There were no behaviour differences among genotypes, nor among evaluation periods. Although the level of nitrate did not influence the production of shoot dry matter in relation to the average among...

Longevity of africanized worker honey bees (Apis mellifera) carrying eye color mutant alleles Scientia Agricola
Almeida,Rosana de; Soares,Ademilson Espencer Egea.
The dark coloration of insects eyes is attributed to the accumulation of the brown pigment insectorubin, a mixture of ommochromes, xanthommatin and several ommins, biosynthesized from tryptophan. When any of the events in the synthesis chain is interrupted, formation and accumulation of pigments other than insectorubin occurs, and a new eye color will appear. The aim of the present work is to evaluate the longevity of worker honey bees Apis mellifera, homozygous and heterozygous for the mutant alleles cream (cr), snow-laranja (s la) and brick (bk). Eye pigmentation and average longetivity of bees are very closely related. Mutant bees carrying lighter eye pigmentation are unable to return to the hive; there is, therefore, a close association between the eye...
