Neutralizing antibodies to bovine herpesviruses types 1 (BHV-1) and 5 (BHV-5) induced by an experimental, oil-adjuvanted, BHV-1 vaccine Braz. J. Vet. Res. Anim. Sci.
PETZHOLD,Sylio Alfredo; RECKZIEGEL,Paulo Estanislao; PRADO,José Antonio Pires; TEIXEIRA,João Carlos; WALD,Vera Beatriz; ESTEVES,Paulo Augusto; SPILKI,Fernando Rosado; ROEHE,Paulo Michel.
An experimental oil-adjuvanted, inactivated vaccine against bovine herpesvirus type 1 (BHV-1.1), was produced and evaluated in its capacity to induce neutralizing antibodies against bovine herpesvirus type 1 (BHV-1, subtypes 1.1 and 1.2) and bovine herpesvirus type 5 (BHV-5). Cattle were vaccinated and revaccinated 90 days later. Antibodies were measured at days 0, 30, 90, 120, 180, 270 and 450 days after the first dose of vaccine (DPV). Antibody titres to BHV-1.1 and BHV-1.2 were significantly higher than to BHV-5 throughout the experiment. While all calves seroconverted to BHV-1.1 and BHV-1.2 after the first dose of vaccine, only two out of 23 (8,7%) calves seroconverted to BHV-5. However, after the booster injection all animals seroconverted to the...

Mimotopes selected by biopanning with high-titer HIV-neutralizing antibodies in plasma from Chinese slow progressors BJID
Zhang,Xiaoli; Han,Xiaoxu; Dai,Di; Mingjia,Bao; Zhang,Zining; Zhang,Min; Bice,Tristan; Zhao,Min; Cao,Yaming; Shang,Hong.
OBJECTIVE: One approach to identifying HIV-1 vaccine candidates is to dissect the natural antiviral immune response in treatment-naïve individuals infected for over ten years, considered slow progressor patients (SPs). It is suspected that SP plasma has strongly neutralizing antibodies (NAb) targeting specific HIV viral epitopes. METHODS: NAbs levels of 11 HIV-1-infected SPs were detected by PBMC-based neutralization assays. To investigate SP NAb epitope, this study used a biopanning approach to obtain mimotopes of HIV-1 that were recognized by SP plasma NAbs. IgG was purified from hightiter NAb SP plasma, and used as the ligand for three rounds of biopanning to select HIV-specific mimotopes from a phage-displayed random peptide library. Double-antibody...

Induction of neutralizing antibodies in mice immunized with scorpion toxins detoxified by liposomal entrapment BJMBR
The possibility of producing neutralizing antibodies against the lethal effects of scorpion toxins was evaluated in the mouse model by immunization with an immunogen devoid of toxicity. A toxic fraction (5 mg) from the venom of the scorpion Tityus serrulatus was entrapped in sphingomyelin-cholesterol liposomes. The liposomes were treated for 1 h at 37oC with a 1% (w/w) trypsin solution in 0.2 M sodium carbonate buffer, pH 8.3. This treatment led to a strong reduction in venom toxicity. Immunization was performed as follows: mice were injected sc with 20 µg of the liposome-entrapped toxic fraction on days 1 and 21 and a final injection (20 µg) was administered ip on day 36. After injection of the immunogen, all mice developed an IgG response which was shown...

Three leptospiral bacterins, produced with different serovars of Serogroup Sejroe, namely the hardjo (bacterin A), wolffi (bacterin B) and guaricura (bacterin C), were evaluated in male hamsters (Mesocricetus auratus) by comparing the agglutinating and neutralizing antibodies titers using microscopic agglutination (MAT) and in vitro growth inhibition (GIT) tests. The immunization schedule was based on two 1.0 mL doses of non-diluted formalininactivated whole culture bacterin given through subcutaneous route with 10-day interval. The challenge was performed ten days after the second vaccine dose, when the animals were inoculated with 0.2 mL of non-inactivated cultures of each serovar through intraperitoneal route. On the 21st post-challenge day (PCD), all...

Immunogenicity in sheep of Uruguayan commercial vaccines against bovine alphaherpesvirus 1, 5 and bovine pestiviruses Ciência Rural
Merchioratto,Ingryd; Aurélio,Alana de Almeida; Villela,Janice Machado; Stone,Nicole Vieira; Roman,Isac Junior; Traesel,Carolina Kist; Brum,Mário Celso Sperotto.
ABSTRACT: The serological responses induced by four commercial inactivated Uruguayan vaccines against bovine alphaherpesviruses (BoHV)-1 and -5 and bovine pestiviruses (BVDV-1, BVDV-2, and HoBiPeV) were evaluated in sheep. Thirty-seven sheep were immunized twice (day 0 and 25) and their serum samples were tested at different intervals (days 0, 25, 40, 60, and 90) post-vaccination (PV). Among the four vaccines tested, only one (G4) could induce the production of moderate neutralizing antibody titers against BoHV-1 and -5 and BVDV-1 and -2. The G3 vaccine showed a neutralizing serological response against the bovine alphaherpesviruses only. The G1 and G2 vaccines produced extremely low levels of antibodies in a few vaccinated animals only (geometric mean...

Serological response to rabies virus induced by commercial vaccines in cattle Ciência Rural
Martins,Mathias; Quadros,João Motta de; Flores,Eduardo Furtado; Weiblen,Rudi.
ABSTRACT: The antibody response to rabies virus (RABV) induced by commercial vaccines in heifers was investigated. For this, 84 heifers were vaccinated twice (30 days interval) with each of four vaccines (G1 = 14 animals; G2 = 24; G3 = 22 and G4 = 24) and received a booster vaccination 360 days later. Serum samples collected at different intervals after vaccination and 30 days after booster were submitted to a virus neutralizing (VN) assay for RABV antibodies. Thirty days after the second vaccine dose, 92% of the immunized animals presented VN titers ≥0.5UI/mL (geometric medium titers [GMT] 1.7 to 3.8UI/mL). At the day of the booster (360 days post-vaccination); however, the percentage of animals harboring antibody titers ≥0.5UI/mL had dropped to 31%...

da Silva, Viviane Alcântara; Gamon, Thais Helena Martins; da Silva, Andréa de Cássia Rodrigues; Caporale, Graciane Maria Medeiros; Chaves, Luciana Botelho; Scheffer, Karin Corrêa.
The National Programme for Rabies Prevention aims to maintainimmunogenic protective levels in vaccinated animals, with titersof neutralizing antibodies (VNA) ≥ 0.5 IU / mL. The aim of thisstudy was to evaluate, according to age, race, and the periodof vaccination and blood collection, the immune response indogs that received a single dose of rabies vaccine virus in cellculture. In this study 432 samples received at the Pasteur Instituteof São Paulo in the triennium 2009-2011 were analyzed. Datawere analyzed and the evaluation of the VNA to rabies virus wasperformed by rapid fluorescent inhibition test foci (RFFIT). Of thetotal samples analyzed, 21.76% did not have protective titers.Among these, 67.02% samples were considered puppies andwhen the interval...
