ABSTRACT High salinity affects plants due to stimulation of osmotic stress. Cell signaling triggered by nitric oxide (NO) and hydrogen sulfide (H2S) activates a cascade of biochemical events that culminate in plant tolerance to abiotic and biotic stresses. For instance, the NO/H2S-stimulated biochemical events that occur in plants during response to high salinity include the control of reactive oxygen species, activation of antioxidant system, accumulation of osmoprotectants in cytosol, induction of K+ uptake and Na+ cell extrusion or its vacuolar compartmentation among others. Due to the lack of a complete urea cycle, uricotelic species, such as broilers, are not able to synthesize de novo arginine (Arg), thus depending exclusively on dietary Arg. High levels of dietary lysine (Lys) increase the demand for Arg because of the antagonistic relationship between these amino acids. The Arg-Lys antagonism promotes an expressive increase in the renal Arg activity and consequently induces the degradation of Arg and the decrease in the activity of glycine amidinotransferase, an enzyme that uses Arg in the synthesis of muscle creatin. In the present study, the antileishmanial effect induced by the ethanol extract of stem barks from Z. rhoifolium (ZR-EEtOH) and its n-hexane fraction (ZR-FHEX) on infection and infectivity of murine macrophages by promastigote forms of Leishmania amazonensis were investigated. Major modifications of immune system have been observed in African trypanosomiasis. These immune reactions do not lead to protection and are also involved in immunopathology disorders. The major surface component (variable surface glycoprotein,VSG) is associated with escape to immune reactions, cytokine network dysfunctions and autoantibody production. Most of our knowledge result from experimental trypanosomiasis. Innate resistance elements have been characterised. In infected mice, VSG preferentially stimulates a Th 1-cell subset. ABSTRACT Post-germination desiccation tolerance (DT) was studied in two wheat cultivars. Effect of pretreatment of abscisic acid (ABA)/ osmotic/ salt/ heat stress was also studied. One day (d)-old seedlings of wheat cultivars PBW 644 (drought tolerant) and PBW 343 (drought susceptible) were exposed to ABA/stress treatment for next 1 d, desiccated for 4 d and subsequently rehydrated for 4 d. Biomass, protein, water, protein carbonyls (oxidative toxicity) and nitric oxide (NO) levels were measured in 2 d (treated), 6 d (desiccated), 10 d (rehydrated) seedlings. Removal of the cortical projections alters expression of NOS in the different cell types of the superficial layers of the superior colliculus in rats Anais da ABC (AABC)

Nitric oxide has several biological roles and nitric oxide synthase (NOS) is expressed in the nervous system, and co-localizes with NADPH-diaphorase. The superficial layers of the superior colliculus (SC), which receive retinal and cortical inputs, present NADPH-d staining in a sub-population of neurons that include all cell types. We have previously shown, by NADPH-diaphorase, that eye enucleation alters the intracellular distribution of NOS. Here, we studied the effect of cortical ablation on NOS expression by neurons in collicular superficial layers. Our results show that cortical ablation alters the proportion of different NOS-positive cell types, but not the intracellular distribution of the enzyme.

Effects of rotational culture on morphology, nitric oxide production and cell cycle of endothelial cells Biocell

Devices for the rotational culture of cells and the study of biological reactions have been widely applied in tissue engineering. However, there are few reports exploring the effects of rotational culture on cell morphology, nitric oxide (NO) production, and cell cycle of the endothelial cells from human umbilical vein on the stent surface. This study focuses on these parameters after the cells are seeded on the stents. Results showed that covering of stents by endothelial cells was improved by rotational culture. NO production decreased within 24 h in both rotational and static culture groups. A capacitative Ca2+ entry (CCE) pathway, activated by depletion of intracellular Ca2+ stores, is thought to mediate much of the Ca2+ entry evoked by receptors that stimulate phospholipase C (PLC). However, in A7r5 vascular smooth muscle cells, vasopressin, which stimulates PLC, empties intracellular Ca2+ stores but simultaneously inhibits their ability to activate CCE. Effect of nitric oxide and plant antioxidants on microsomal content of lipid radicals Biol. Res.

The antioxidant ability of nitric oxide (NO) generated by a chemical donor and of commercially available antioxidant preparations was assayed. SNAP (S-Nitroso-N-acetylpenicilamine) was used as the NO donor, and Ginkgo biloba, wheat and alfalfa preparations were tested. Lipid peroxidation was assayed by EPR employing a reaction system consisting of rat liver microsomes, ADP, FeCl3, NADPH and POBN in phosphate buffer, pH=7.4. In vitro NO exposure decreased microsomal lipid peroxidation in a dose-dependent manner. In the present study, we investigated whether cellular damage, as demonstrated by lactate dehydrogenase (LDH) release in the human fallopian tube (FT) infected by Neisseria gonorrhoeae (Ngo), correlated with high levels of nitric oxide synthase (NOS) mRNA and enzyme activity. Infection with Ngo induced a significant increase (~35-fold) in mRNA transcripts of the inducible isoform of NOS. Paradoxically, a reduction in NOS enzyme activity was observed in infected cultures, suggesting that gonococcal infection possibly influences translation of iNOS mRNA to the enzyme. This study investigated the activation in resident, inflammatory macrophages and J774 cells in vitro treated with lipopolysaccharide (LPS), soluble Leishmania antigen (SLA), calcium ionophore (CaI) and magnesium (Mg2+) alone or combined. Endothelial dysfunction is one of the earliest events in atherogenesis. A consequence of endothelial damage is a lower availability of nitric oxide (NO), the most potent endogenous vasodilator. NO inhibits platelet aggregation, smooth muscle cell proliferation and adhesion of monocytes to endothelial cells. Endothelial dysfunction is present in patients with cardiovascular disease and/or coronary risk factors, such as hypertension, dyslipidemia, diabetes, smoking or hyperhomocysteinemia.
