Genetic linkage map and mapping of the locus of biological nitrogen fixation inefficiency in cowpea Agronomy

The objectives of the present study were to construct a cowpea genetic map using the F2 population resulting from the cross IC-1 x BRS Marataoã, based on single nucleotide polymorphism (SNP) markers, and to map the cpi gene, with additional reference to introgression with the consensus map of species, aiming to identify markers for assisted selection to develop more efficient cultivars for BNF. The parents and 89 F2 plants were genotyped with 51,128 SNP markers, of which 910 polymorphic markers were used to construct the map. A fixação biológica do nitrogênio é de fundamental importância na sustentabilidade agrícola, e a caracterização fenotípica rizobial contribui para o conhecimento da sua diversidade em coleções e para a utilização posterior em sistemas agrícolas. O objetivo deste trabalho foi avaliar a diversidade fenotípica de rizóbio isolados de solos da Amazônia e suas eficiências simbióticas quando inoculados em feijão caupi (Vigna unguiculata L. Walp.). Os isolados nativos apresentaram diversidade quanto às características morfológicas e fisiológicas. Foi verificado que a inoculação com isolados de rizóbio nativos contribuiu, de forma significativa, para o aumento da biomassa, nodulação de caupi e em eficiência simbiótica.

Azospirillum sp . inoculation in wheat, barley and oats seeds greenhouse experiments BABT

The aim of the present work was to evaluate the inoculation effects of Azospirillum sp. RAM-7 strain, in seeds of wheat, barley and oats cultures on yield and total nitrogen content of the grains in greenhouse experiments. Seeds inoculation with Azospirillum sp. RAM-7 strain enhanced grain yields, even so, the responses vary among the evaluated cultures. For wheat, significant increases were obtained when the inoculation was associated to 100% of the recommended nitrogen, although the treatment that received only inoculation produced 7.4% above the control, however this difference wasn't significant. Growth of Crotalaria juncea L. supplied with mineral nitrogen BABT

Plants of Crotalaria juncea inoculated with Rhizobium were treated with nutrient solution containing 10 or 20mg of either N/NO3 or N/NH4.plant-1.week-1 . The control plants received nutrient solution without N. An investigation was conducted on the effect of these sources of N on growth and nitrogen fixation of plants with 30, 60 and 90 days after sowing (DAS). Those that received mineral N presented higher growth than -N plants, but the presence of nodules occurred in all the treatments. Plants treated with NH4 presented higher N content until 60 days. The highest concentrations of leghemoglobin and protein in nodules were found at 30 DAS and there was no difference in leghemoglobin content between treatments for any age and in protein from 60 DAS....

Azospirillum brasilense is a diazotroph found in association with important agricultural crops. In this organism, the regulation of nitrogen fixation by ammonium ions involves several proteins including the uridylyltransferase/uridylyl-removing enzyme, GlnD, which reversibly uridylylates the two PII proteins, GlnB and GlnZ, in response to the concentration of ammonium ions. In the present study, the uridylylation/deuridylylation cycle of A. brasilense GlnB and GlnZ proteins by GlnD was reconstituted in vitro using the purified proteins. Azospirillum brasilense is a diazotroph that associates with important agricultural crops and thus has potential to be a nitrogen biofertilizer. The A. brasilense transcription regulator NifA, which seems to be constitutively expressed, activates the transcription of nitrogen fixation genes. It has been suggested that the nitrogen status-signaling protein GlnB regulates NifA activity by direct interaction with the NifA N-terminal GAF domain, preventing the inhibitory effect of this domain under conditions of nitrogen fixation. Application of representational difference analysis to identify genomic differences between Bradyrhizobium elkanii and B. Japonicum species BJM

Bradyrhizobium elkanii is successfully used in the formulation of commercial inoculants and, together with B. japonicum, it fully supplies the plant nitrogen demands. Despite the similarity between B. japonicum and B. elkanii species, several works demonstrated genetic and physiological differences between them. In this work Representational Difference Analysis (RDA) was used for genomic comparison between B. elkanii SEMIA 587, a crop inoculant strain, and B. japonicum USDA 110, a reference strain. Nitrogenase activity of Beijerinckia derxii is preserved under adverse conditions for its growth BJM

In order to evaluate the response of Beijerinckia derxii ICB-10 to different environmental factors, growth curves and specific nitrogenase activity were studied. Tested conditions were as follows: a) media with different pH values (2.5, 2.8, 4.2 and 5.7); b) medium supplemented with 230 µM aluminium sulphate; c) media with two different potassium phosphate concentrations (50 mM and 100 mM); d) shaken or still cultures; e) medium supplemented with 40 mM sodium thiosulphate. Torr., se recolectó suelo de cinco localidades 1) Salinas, San Luis Potosí 2) Ejido, Jacalón, Salinas, San Luis Potosí; 3) Calera, Zacatecas; 4) Santa María del Oro, Durango y 5) Crucero La Zarca, Durango. Abstract Cyanobacteria is a remarkable group of prokaryotic photosynthetic microorganisms, with several genera capable of fixing atmospheric nitrogen (N2) and presenting a wide range of morphologies. Although the nitrogenase complex is not present in all cyanobacterial taxa, it is spread across several cyanobacterial strains. The nitrogenase complex has also a high theoretical potential for biofuel production, since H2 is a by-product produced during N2 fixation. ABSTRACT: Soybean [Glycine max (L.) Merr.] is the most important legume cropped worldwide, with an economic value attributed mainly to the high protein content of the grain, which, in turn, demands a heavy supply of nitrogen. An environmentally friendly source of nitrogen at low cost to farmers is represented by the biological fixation of atmospheric nitrogen that takes place is association with symbiotic bacteria.
