Prior to abscission, nutrients are redeployed from leaves and redistributed to other parts of the plant. Data comparing nutrient resorption to soil fertility and leaf life span remains controversial in the literature. We compared nitrogen (N) conservation mechanisms among four legume trees with different leaf life spans (Hymenaea courbaril L. var. stilbocarpa (Hayne) Lee et Lang., Lonchocarpus guilleminianus (Tul.) Malme, Enterolobium contortisiliquum (Vell.) Morong and Peltophorum dubium (Spreng.) Taub.), from a semideciduous tropical forest, remnant of the Atlantic Forest. We hypothesized that these mechanisms differ among the four species and are affected by their leaf life span and by the availability of N, both as a mineral in the soil and, in the... |

The objectives of the present study were to construct a cowpea genetic map using the F2 population resulting from the cross IC-1 x BRS Marataoã, based on single nucleotide polymorphism (SNP) markers, and to map the cpi gene, with additional reference to introgression with the consensus map of species, aiming to identify markers for assisted selection to develop more efficient cultivars for BNF. The parents and 89 F2 plants were genotyped with 51,128 SNP markers, of which 910 polymorphic markers were used to construct the map. The results revealed 11 linkage groups, with an average of 82 markers per chromosome and average distance of 1.26 cM between markers. Recombination analysis of the SNPs indicated that markers 2_12850 and 2_00188, located in linkage... |

The aim of the present work was to evaluate the inoculation effects of Azospirillum sp. RAM-7 strain, in seeds of wheat, barley and oats cultures on yield and total nitrogen content of the grains in greenhouse experiments. Seeds inoculation with Azospirillum sp. RAM-7 strain enhanced grain yields, even so, the responses vary among the evaluated cultures. For wheat, significant increases were obtained when the inoculation was associated to 100% of the recommended nitrogen, although the treatment that received only inoculation produced 7.4% above the control, however this difference wasn't significant. For barley, the presence of the inoculant substituted 20% the recommended nitrogen fertilization. For oats the inoculation with Azospirillum sp. RAM-7 didn't... |

Azospirillum brasilense is a diazotroph found in association with important agricultural crops. In this organism, the regulation of nitrogen fixation by ammonium ions involves several proteins including the uridylyltransferase/uridylyl-removing enzyme, GlnD, which reversibly uridylylates the two PII proteins, GlnB and GlnZ, in response to the concentration of ammonium ions. In the present study, the uridylylation/deuridylylation cycle of A. brasilense GlnB and GlnZ proteins by GlnD was reconstituted in vitro using the purified proteins. The uridylylation assay was analyzed using non-denaturing polyacrylamide gel electrophoresis and fluorescent protein detection. Our results show that the purified A. brasilense GlnB and GlnZ proteins were uridylylated by... |

Sugarcane is an important agricultural product of Brazil, with a total production of more than 500 million tons. Knowledge of the bacterial community associated with agricultural crops and the soil status is a decisive step towards understanding how microorganisms influence crop productivity. However, most studies aim to isolate endophytic or rhizosphere bacteria associated with the plant by culture-dependent approaches. Culture-independent approaches allow a more comprehensive view of entire bacterial communities in the environment. In the present study, we have used this approach to assess the bacterial community in the rhizosphere soil of sugarcane at different times and under different nitrogen fertilization conditions. At the high taxonomic level, few... |

Azospirillum brasilense is a diazotroph that associates with important agricultural crops and thus has potential to be a nitrogen biofertilizer. The A. brasilense transcription regulator NifA, which seems to be constitutively expressed, activates the transcription of nitrogen fixation genes. It has been suggested that the nitrogen status-signaling protein GlnB regulates NifA activity by direct interaction with the NifA N-terminal GAF domain, preventing the inhibitory effect of this domain under conditions of nitrogen fixation. In the present study, we show that an N-terminal truncated form of NifA no longer required GlnB for activity and lost regulation by ammonium. On the other hand, in trans co-expression of the N-terminal GAF domain inhibited the... |

Bradyrhizobium elkanii is successfully used in the formulation of commercial inoculants and, together with B. japonicum, it fully supplies the plant nitrogen demands. Despite the similarity between B. japonicum and B. elkanii species, several works demonstrated genetic and physiological differences between them. In this work Representational Difference Analysis (RDA) was used for genomic comparison between B. elkanii SEMIA 587, a crop inoculant strain, and B. japonicum USDA 110, a reference strain. Two hundred sequences were obtained. From these, 46 sequences belonged exclusively to the genome of B. elkanii strain, and 154 showed similarity to sequences from B. japonicum genome. From the 46 sequences with no similarity to sequences from B. japonicum, 39... |

Native rhizobia are ideal for use as commercial legume inoculants. The characteristics of the carrier used to store the inoculants are important for the survival and symbiotic potential of the rhizobia. The objective of this study was to investigate the effects of peat (PEAT), perlite sugarcane bagasse (PSB), carboxymethyl cellulose plus starch (CMCS), and yeast extract mannitol supplemented with mannitol (YEMM) on the survival, nodulation potential and N2 fixation capacity of the native strains Sinorhizobium mexicanum ITTG R7T and Rhizobium calliandrae LBP2-1T and of the reference strain Rhizobium etli CFN42T. A factorial design (4 × 3) with four repetitions was used to determine the symbiotic potential of the rhizobial strains. The survival of the... |

Abstract Aneurinibacillus aneurinilyticus strain CKMV1 was isolated from rhizosphere of Valeriana jatamansi and possessed multiple plant growth promoting traits like production of phosphate solubilization (260 mg/L), nitrogen fixation (202.91 nmol ethylene mL-1 h-1), indole-3-acetic acid (IAA) (8.1 µg/mL), siderophores (61.60%), HCN (hydrogen cyanide) production and antifungal activity. We investigated the ability of isolate CKMV1 to solubilize insoluble P via mechanism of organic acid production. High-performance liquid chromatography (HPLC) study showed that isolate CKMV1 produced mainly gluconic (1.34%) and oxalic acids. However, genetic evidences for nitrogen fixation and phosphate solubilization by organic acid production have been reported first time... |

Abstract Plant-microbe interactions in the rhizosphere are the determinants of plant health, productivity and soil fertility. Plant growth-promoting bacteria (PGPB) are bacteria that can enhance plant growth and protect plants from disease and abiotic stresses through a wide variety of mechanisms; those that establish close associations with plants, such as the endophytes, could be more successful in plant growth promotion. Several important bacterial characteristics, such as biological nitrogen fixation, phosphate solubilization, ACC deaminase activity, and production of siderophores and phytohormones, can be assessed as plant growth promotion (PGP) traits. Bacterial inoculants can contribute to increase agronomic efficiency by reducing production costs... |

ABSTRACT: Soybean [Glycine max (L.) Merr.] is the most important legume cropped worldwide, with an economic value attributed mainly to the high protein content of the grain, which, in turn, demands a heavy supply of nitrogen. An environmentally friendly source of nitrogen at low cost to farmers is represented by the biological fixation of atmospheric nitrogen that takes place is association with symbiotic bacteria. For the soybean, the association occurs mainly with bacteria belonging to the species Bradyrhizobium japonicum and Bradyrhizobium elkanii, but our knowledge about gene expression associated with the symbiosis is still poor. This work - part of an effort by the Soybean Genome Consortium (Genosoja)?was conducted with the aim of achieving better... |
