Karyotype and nuclear DNA content of Trichomycterus areolatus (Siluriformes, Trichomycteridae) Genet. Mol. Biol.

Cytogenetic analysis of Trichomycterus areolatus, collected from the Tijeral and Huilma Rivers in southern Chile has shown a diploid chromosome number of 2n = 54, a fundamental number of FN = 106, and a karyotypic formula of 44m + 8sm + 2st. Intra-individual polymorphism of chromosome number (2n = 54, 55 and 56) in specimens from the Huilma River has also been documented, providing further evidence of the occurrence of this phenomenon in Trichomycterus. The karyotype exhibited large chromosome pairs: metacentric pairs 1 (relative length 7.54%), 2 (5.75%) and 3 (5.09%), submetacentric pair 23 (5.25%), and subtelocentic pair 27 (5.28%). In observations by confocal or conventional fluorescence microscopy, important factors should be considered in order to obtain accurate images. One of them, such as the fluorescence bleaching from highest intensity to lowest signal of fluorescence is a common problem with several DNA fluorochromes and especially for DAPI stain. The fluorescence of DAPI fades rapidly when it is exposed to UV light, under optimal conditions of observation. Although the fading process can be retarded using a mounting medium with antifading reagents, the photochemical process underlying the fluorescence decay has not yet been fully explained. Cytogenetic studies in some species of Passiflora L. (Passifloraceae): a review emphasizing Brazilian species BABT

The Passifloraceae is represented by species of tropical and subtropical origin. The Passiflora is the richest genus with approximately 450 species, 200 of them being native to Brazil. Recent karyological studies have reported the basic chromosome number for the Passiflora genus as x = 6, whereas x = 9, x = 10 and x = 12 were established as secondary basic numbers. High rates of fertility occur in most Passiflora species, since both meiotic index and pollen viability are above 90%. Unusual meiotic behavior has been described in some taxa. Flow cytometry allows to estimate the DNA content of a large number of plants quickly. However, inadequate protocols can compromise the reliability of these estimates leading to variations in the values of DNA content the same species. The objective of this study was to propose an efficient protocol to estimate the DNA content of Brachiaria spp. genotypes with different ploidy levels using flow cytometry. The development of homozygous breeding lines in maize may be accelerated through the use of haploids. Thus, the obtaining and prior identification of haploids generated by the haploid inducer lines is an important factor. The purpose of this study was to identify haploids by flow cytometry and to correlate the nuclear DNA content to the morphological and morphometric traits of the seeds that gave rise to them. In addition, molecular markers were used to confirm the androgenetic nature of the haploid. The seeds obtained were derived from the cross between the inbred line W23 and the commercial hybrid P30F90.
