Sources of paralytic shellfish toxin accumulation variability in the Pacific oyster Crassostrea gigas ArchiMer

This study was designed to assess the contribution of feeding behavior to inter-individual variability of paralytic shellfish toxin (PST) accumulation in the Pacific oyster Crassostrea gigas. For this purpose 42 oysters were exposed for 2 days to non-toxic algae and then for 2 other days to the PST producer Alexandrium minutum. Individual clearance rate (CR) of oysters was continuously monitored over the 4 days using an ecophysiological measurement system. Comparison of CR values when exposed to toxic and non toxic algae allowed to estimate a clearance rate inhibition index (CRII). Since 2008, the French oyster-farming sector has been confronted with exceptionally high rates of mortality affecting spats (less than one year old) of the Pacific oyster, C. gigas, of between 60 and 90% at all rearing sites and in Pertuis Charentais sea. The objective of this document is to provide an overview of the research and studies carried out by Ifremer in Poitou-Charentes since the emergence of this phenomenon. The actions presented were funded by IFREMER, and CPER Poitou-Charentes 2007-2013.

Combination of a pesticide exposure and a bacterial challenge: In vivo effects on immune response of Pacific oyster, Crassostrea gigas (Thunberg) ArchiMer

To assess the impact of pollution induced by pesticides on Pacific oyster, Crassostrea gigas, health in France, in vivo effects of combined pesticide exposure and bacterial challenge on cell activities and gene expression in hemocytes were tested using flow cytometry and real-time PCR. As a first step, an in vivo model of experimental contamination was developed. La conchyliculture, et principalement l’élevage de l’huître creuse, Crassostrea gigas, constitue la principale activité aquacole française. Cette activité repose, en grande partie, sur le recrutement naturel de l’espèce qui assure 60 à 70% des besoins en jeunes huîtres (naissain) : cette activité de collecte s’appelle le captage. Les deux principaux centres de captage en France sont les bassins d’Arcachon et de Marennes-Oléron. Infectious diseases are a limiting factor to the development and sustainability of aquaculture. For mollusc cultures, only prophylactic measures may be applied, their development usually facilitated by the reproduction of the disease under experimental conditions. To develop an experimental infection in Crassostrea gigas, Vibrio strains were isolated and their virulence was experimentally evaluated. Collaboration phenomena between some strains were observed in infection trial, leading to higher mortality rate when two strains are concomitantly inoculated. Haplosporidiosis in the pacific oyster Crassostrea gigas from the French Atlantic coast ArchiMer

Two cases of haplosporidian infection occurred during 1993 in Pacific oysters Crassostrea gigas from the French Atlantic coast. The localization and ultrastructure of the plasmodia are described. In situ hybridization of infected tissue sections was conducted with DNA probes for oyster-infecting haplosporidians. The Haplosporidium nelsoni-specific DNA probe MSX1347 hybridized with the C. gigas parasite, and the H, costale-specific probe SSO1318 did not hybridize. Ostreid herpesvirus type 1 (OsHV-1) can be considered one of the major infectious agents in Pacific oysters, Crassostrea gigas, in France and all around the world. Differences in terms of mortality among infected animals were reported suggesting a genetic basis of the susceptibility to the OsHV-1 infection. In this context, the main objective of this thesis was to understand the interactions between Pacific oysters and OsHV-1, in particular, the molecular basis of the viral cycle. Results showed that the virus is able to replicate in the host regardless of its stage of development or its susceptibility.
