The effects of polluted water at three sites in the Marinho River, Brazil, on Oreochromis niloticus (Nile tilápia) were investigated using histological, hematological and biochemical approaches. Fish exposed to the impacted water demonstrated that histological changes in gills were accompanied by nuclear and micronuclei abnormalities in cells. The aim of this work was to evaluate the physiological aspects of polyphosphate metabolism of Cunninghamella elegans grown in presence of copper. The growth profile was obtained by means of biomass yields, orthophosphate consumption, polyphosphate accumulation and phosphatases activities. The results revealed the influence of copper on the growth, observed by biomass yields. Orthophosphate consumption was faster in cells grown in the presence of copper. The presence of copper in the culture medium induced polyphosphate accumulation. The polyphosphate level was almost constant in the beginning of control culture growth, and could be related to the exponential growth phase. The effect of arsenite (As2O3) in situ on the level of the phosphate pool and activities of phosphohydrolytic enzymes was examined in rice (Oryza sativa L.) seedlings grown for 5-20 d in sand cultures. The effects were manifested via a decline in phosphate content and inhibition of the activities of key phosphatases. Application of 50 µM As2O3 in situ resulted in 34 to 77% inhibition of acid phosphatase activity in roots and about 38 to 50% inhibition of activity in shoots of 15-20-d-old seedlings. Similarly, alkaline phosphatase activity was inhibited in shoots under in situ As (III) toxicity. Acid phosphatases (3.1.3.2) are a group of enzymes widely distributed in nature, which catalyze the hydrolysis of a variety of phosphate esters in the pH range of 4-6. We confirmed the presence of acid phosphatases in seedlings of cucumber (Cucumis sativus), radish (Raphanus sativus) and rocket salad (Eruca vesicaria) under different assay conditions using a rapid and simple preparation. The results showed that the optimum pH and temperature used for all species were close to 5.5 and 35°C, respectively. The enzyme was inhibited by molybdate, fluoride, azide, levamisole, orthovanadate, Zn2+ and Cu2+. Suramin had no effect on enzyme activity.
