Influence of dietary phospholipids on early ontogenesis of fish ArchiMer
Cahu, Chantal; Gisbert, Enric; Villeneuve, Laure A. N.; Morais, Sofia; Hamza, Neila; Wold, Per-avid; Zambonino, Jose-luis.
The aim of this paper is to provide explanations of how dietary phospholipid (PL) globally improves fish larval development, including growth and survival, digestive functions and skeletal development, and to propose optimal PL levels and sources in fish larval diets. Dietary incorporation of 8-12% PL related to dry matter (d.m.) promotes growth and enhanced survival in various species. Marine source PL, incorporating highly unsaturated fatty acids, was most efficient than soybean lecithin. This beneficial effect was explained by an enhancement in digestive functions, assessed by digestive enzyme activities and histomorphology. Nevertheless, 1.5-2.5% highly unsaturated fatty acids related to diet d.m. supplied by PL improved growth, survival and skeletal...

Improvement of lipid and phospholipid recoveries from sardine (Sardina pilchardus) viscera using industrial proteases ArchiMer
Dumay, Justine; Donnay-moreno, Claire; Barnathan, G; Jaouen, P; Berge, Jean-pascal.
Enzymatic hydrolysis of sardine viscera by three broad spectrum proteases was investigated using the pHstat method (24 h, pH 8, 50 degrees C). After hydrolysis, three fractions (sludge, aqueous phase and oily phase) were collected. For each fraction, lipids and phospholipids were quantified and molecular weights of aqueous phase peptides were determined. Under these conditions, the degree of hydrolysis (DH) varied from 1.9% (Flavourzyme), 3.1% (Protamex) and to 3.3% (Alcalase). Dry matter distribution indicated that all hydrolysis performed increased solubilisation of proteins: protein recovery for each hydrolysis ranged from 57.4% to 61.2%. Furthermore, the total lipid content in the liquid parts (oily and aqueous phases) increased (at least 85% of lipids...

Acumulación de ácido araquidónico en fosfolípidos y triglicéridos y producción de prostaglandinas con relación a estrés térmico en semilla de ostión Crassostrea gigas ArchiMer
Duran Encinas, Yazmin.
Crassostrea gigas is a marine invertebrate that inhabits the intertidal zone and therefore, is commonly exposed to multiple environmental stressors, including temperature. In recent years mass mortality events have been reported in Mexico in C. gigas associated with the increase in temperature during the summer. Organisms under thermal stress present an increase in prostaglandins (PG) production by cyclooxygenases (COX), an enzymatic route using arachidonic acid (ARA) present in the phospholipids (FL) of the cell membrane as substrate, but PG can also increase non-enzymatically by reactive oxygen species (ROS) that are produced during increased metabolism. It has been reported that some organisms exposed to stress accumulate a higher concentration of ARA...

Substitution of live food by formulated diets in marine fish larvae ArchiMer
Cahu, Chantal; Zambonino, Jose-luis.
Until recently, it was considered impossible to feed newly hatched marine fish species with a compound diet. Substituting a compound diet for live prey was performed several weeks after hatching, depending on the species. Compound diets were well ingested at the early stage but larvae died with a gut full of food, suggesting that larvae were unable to digest the compound diet. The hypothesis was that younger larvae have insufficient digestive enzymes to thrive on compound diets, and that exogenous enzymes, provided from live prey, are necessary for early stages. The organogenesis of marine fish larvae is not completely achieved at hatching and histological studies have revealed that the anatomy of the digestive tract undergoes developmental changes over...

Nutritional components affecting skeletal development in fish larvae ArchiMer
Cahu, Chantal; Zambonino, Jose-luis; Takeuchi, Toshio.
Marine fish larvae undergo major functional and morphological changes during the developmental stages and several factors can interfere with the normal development of larvae and affect fry quality. Skeletal malformations, such as spinal malformation-scoliosis, lordosis, coiled vertebral column-, missing or additional fin rays, bending opercle or jaw malformations, are frequently observed in hatchery-reared larvae. This paper reviews the effects of some nutritional components on skeletal development in larvae of a number of fish species. In the dietary lipid fraction, for instance, it was proven that the phospholipid concentration affected the spinal malformation rate in sea bass fed a compound diet from mouth opening onwards. Phosphatidylinositol, in...

Differential protein expression profile in the liver of pikeperch (Sander lucioperca) larvae fed with increasing levels of phospholipids ArchiMer
Hamza, Neila; Silvestre, Frederic; Mhetli, Mohamed; Ben Khemis, Ines; Dieu, Marc; Raes, Martine; Cahu, Chantal; Kestemont, Patrick.
A comparative proteomic approach was used to assess the protein expression profile in the liver of 34 days old pikeperch larvae fed from day 10 post hatching, with three isoproteic and isolipidic formulated diets varying by their phospholipid (PL) contents (% dry diet weight): 1.4% (PL1), 4.7% (PL5) and 9.5% (PL9). Using 2D-DIGE minimal labelling of liver extracts, we were able to show 56 protein spots with a differential intensity (p<0.05) depending on the dietary PL content. Among these spots, 11 proteins were un-ambiguously identified using nanoLC-MS/MS tandem mass spectrometry. In the PL9 larvae, our results indicate that the glycolytic pathway could be down-regulated due to the under-expression of the fructose biphosphate aldolase B and the...

Ontogenèse des fonctions digestives et besoins nutritionnels chez les larves de poissons marins ArchiMer
Cahu, Chantal; Zambonino, Jose-luis.
The production of fish larvae by hatchery shows an intensive increase for several species, but the specific nutritional requirements of larvae were poorly known until some years. We have conducted several experiments aimed to understand the specificities in larvae digestion, and to formulate a compound diet taking into account the differences compared to juveniles. The chosen species was the European sea bass, Dicentrarchus labrax, and the data were validated on different species. Larvae were fed live preys or experimental compound diet, especially formulated for the experiment. It appeared that pancreatic digestive enzymes, such as trypsin and amylase could be assayed in early ontogenesis, before mouth opening, and increase dramatically during the first...

Remodeling of membrane lipids in gills of adult hard clam Mercenaria mercenaria during declining temperature ArchiMer
Parent, G; Pernet, Fabrice; Tremblay, R; Sevigny, J; Ouellette, M.
In a previous study, a major remodeling of lipids, consistent with the theory of homeoviscous adaptation, was observed in the juvenile hard clam Mercenaria mercenaria during a temperature reduction from 24 to -1 degrees C. In addition, the lipid remodeling varied between genetically distinct lines of hard clams. The present study examined whether adult hard clams originating from different locations and a selectively bred variety show differences related to their genetic characteristics in the remodeling of lipids that normally occur during decreasing temperatures. Wild hard clams from 4 locations in Atlantic Canada and the selectively bred M mercenaria var. notata were held at an aquaculture growout site located at the northern distribution limit of the...

Dietary phospholipids are more efficient than neutral lipids for long-chain polyunsaturated fatty acid supply in European sea bass Dicentrarchus labrax larval development ArchiMer
Gisbert, Enric; Villeneuve, Laure; Zambonino, Jose-luis; Quazuguel, Patrick; Cahu, Chantal.
We evaluated the effects of dietary lipid class (phospholipid vs. neutral lipid) and level of n-3 long-chain PUFA (LC-PUFA) on the growth, digestive enzymatic activity, and histological organization of the intestine and liver in European sea bass larvae. Fish were fed from the onset of exogenous feeding at 7 to 37 d post-hatch with five isoproteic and isolipidic compound diets with different levels of EPA and DHA. Diet names indicated the percentage of EPA and DHA contained in the phospholipids (PL) and neutral lipids (NL), as follows: PL5, PL3, PL1, NL1, and NL3. Histological observations showed different patterns of lipid absorption and accumulation in the intestinal mucosa depending on the level and nature of the dietary lipid fraction. Fish fed high...

Relation between lipid and fatty acid composition of eggs and larval survival in white pacific shrimp (Penaeus vannamei, Boone, 1931) ArchiMer
Palacios, Elena; Rehault, Jean Pierre; Heras, Horacio; Marty, Yannick; Moal, Jeanne; Samain, Jean-francois.
In this study, lipid classes and fatty acid composition were determined in eggs of P. vannamei as a function of survival to zoea III stage. Spawns were reared individually to zoea III and grouped, according to their final survival to this larval stage, into spawns of high and low survival. Eggs of individual spawns were analyzed for lipid and fatty acid composition of neutral and polar lipids and the results were then grouped according to survival to zoea III. The lipids within each group (high and low survival to zoea III) were pooled for the separation of phospholipids and the analysis of their fatty acid composition. Higher levels of triglycerides, carotenoids, and linoleic (18: 2n-6) acid in eggs were associated with improved survival to zoea III....

Lipid composition of the pacific lion-paw scallop, Nodipecten subnodosus, in relation to gametogenesis - 2. Lipid classes and sterols ArchiMer
Palacios, Elena; Racotta, I.s.; Arjona, O; Marty, Y; Le Coz, Jean-rene; Moal, Jeanne; Samain, Jean-francois.
Knowledge on the energy distribution during the reproductive cycle of the giant lion-paw scallop, Nodipecten subnodosus is limited, but necessary for aquaculture purposes. In a previous work, we found that natural food availability can sustain the full cost of gametogenesis in optimal environmental conditions. The present study analyses the lipid and sterol classes' composition of several tissues during gametogenesis. In female gonads, the quantity of triacylglycerol (TG) increased parallel to gametogenesis from December (spawned gonads, 0.7 mg g(-1) or 7% of total lipids) to June (ripe gonads, 52 mg g(-1) or 66% of total lipids). In male gonads sampled in June, the concentration of TG (0.7 mg g(-1) or 3.5% of total lipids) and esterified sterols was...

Martínez-Aispuro,José Alfredo; Mendoza,German David; Cordero-Mora,José Luis; Ayala-Monter,Marco Antonio; Sánchez-Torres,María Teresa; Figueroa-Velasco,José Luis; Vázquez-Silva,Gabriela; Gloria-Trujillo,Adrian.
ABSTRACT An experiment was carried out to evaluate the effect of an herbal choline feed plant additive on the productive parameters and blood metabolites of finishing lambs. Forty male Hampshire × Suffolk lambs (initial body weight of 30.36±3.75 kg) were assigned according to a completely randomized design. Treatments consisted of dietary inclusion of the herbal additive BioCholine at 0, 3, 6, and 9 g/kg dry matter for 56 days. A linear response for herbal choline dose was observed for daily gain, final body weight, feed conversion, blood glucose, and blood phosphatidylcholine as herbal choline level was increased in the diet. Total protein, globulins, and low-density lipoproteins showed a quadratic effect, and there were no effects on intake, Longissimus...
