Radiopharmaceuticals play a critical role in modern medicine primarily for diagnostic purposes, but also for monitoring disease progression and response to treatment. As the use of image has been increased, so has the use of prescription medications. These trends increase the risk of interactions between medications and radiopharmaceuticals. These interactions which have an impact on image by competing with the radiopharmaceutical for binding sites for example can lead to false negative results. Comparative biodistribution profile of [131I]VIP and [131I]VIP10-28 BABT

Various tumor cells express significantly higher amounts of VIP receptors (VIPR) that provided the basis for the clinical use of radiolabeled VIP for the in vivo localization of tumors. This work studied the labeling of VIP and VIP10-28 with iodine-131 to compare the biological distribution of the labeled compounds in Nuce mice and the affinity for tumor cells. Both VIP and VIP10-28 peptides contain two tyrosine residues, in positions 10 and 22, that are theoretically equally susceptible to radioiodination employing oxidative electrophilic substitution using oxidizing agents like Chloramine T. Nuclear medicine has an important role in aiding the diagnosis of particularly deep-seated infections such as abscesses, osteomyelitis, septic arthritis, endocarditis, and infections of prosthetic devices. The evidence that natural and synthetic drugs can affect radiolabeling or bioavailability of radiopharmaceuticals in setting of nuclear medicine clinic is already known. However, this drug interaction with radiopharmaceuticals (DIR) is not completely understood. Several authors have described the effect of drugs on the labeling of blood elements with technetium-99m (99mTc) and on the biodistribution of radiopharmaceuticals. Radiopharmaceuticals for imaging chronic lymphocytic inflammation BABT

In the last few decades, a number of radiopharmaceuticals for imaging inflammation have been proposed that differ in their specificity and mechanism of uptake in inflamed foci as compared to the traditional inflammation imaging agents. Radiolabelled cytokines represent a reliable tool for the preclinical diagnosis of chronic inflammatory processes, even before anatomical and functional changes occur in affected tissues. Moreover, the introduction of radiolabelled monoclonal antibodies and sophisticated technique like PET/CT now make the field of inflammation imaging highly specific and accurate. In this review, different approaches of the established and experimental radiopharmaceuticals for imaging of chronic inflammation are discussed.

Effect of antimalarial drugs on the bioavailability of the methylenediphosphonic acid labeled with technetium99m (99mTc-MDP) in Wistar rats BABT

The aim of this work was to study the effect of antimalarial drugs on the bioavailability of 99mTc-MDP in rats. Mefloquine (MQ) and artemisinin (AM) were administered in two treated groups (T) and sorbitol in control group (C) for 7 days. Then, 99mTc-MDP was injected in all groups and %ATI was calculated. A significant increase of %ATI in MQ group, from C to T, occurred in spleen (0.35±0.10to0.58±0.13), liver (1.69±0.28to3.31±0.07) and blood (0.79±0.17to2.09±0.53). The %ATI increased significantly in AM group:femur (2.76±0.59to5.98±0.70), liver (1.69±0.28to4.59±0.68), lungs (0.29±0.05to6.22±0.86), spleen (0.35±0.10 to0.86±0.15) and blood (0.79±0.17 to4.65 ±0.74). Importance of the nutritional status for the interpretation of nuclear medicine examinations BABT

Malnutrition is very prevalent in the Third World, but still in developing Countries and is found in certain communities in developed Countries. Several laboratories examinations are affected by malnutrition. Recently, gestational or neonatal malnutrition were considered to contribute to the development of chronic diseases in adulthood, this phenomena was named programming or metabolic imprinting. Similar consideration were suggested for Nuclear Medicine examinations. Here we review the literature about this aspect and present our own data showing changes in biodistribution of a radiopharmaceutical compound in different animal models of adult malnutrition or caused by maternal malnutrition programming.

Activities, procedures and doses in pediatric patients due to radiopharmaceuticals BABT

An investigation performed between 2003 and 2005 in sixteen selected public and private institutions in Northeast, Southeast and South geographical regions of Brazil evaluated average organ doses and effective doses in 2,411 pediatric patients due to diagnostic procedures with radiopharmaceuticals. For 1 year, effective doses were greater than literature. For 5 years, differences were noticed between present work and literature for bone scintigraphy, thyroid scintigraphy and 67Ga citrate scintigraphy. These differences may be attributed to the uncertainties in internal dose calculations. Knowledge of the radiochemical purity of radiopharmaceuticals is mandatory and can be evaluated by several methods and techniques. Planar chromatography is the technique normally employed in nuclear medicine since it is simple, rapid and usually of low cost. In the present study we evaluated the binding of the radiopharmaceuticals sodium pertechnetate (Na 99mTcO4), methylenediphosphonic acid (99mTc-MDP) and glucoheptonate acid (99mTc-GHA) to blood elements using centrifugation and radioautographic techniques. Heparinized blood was incubated with the labelled compounds for 0, 1, 2, 3, 4, 6 and 24 h. Plasma (P) and blood cells (BC) were isolated and precipitated with 5% trichloroacetic acid (TCA), and soluble (SF) and insoluble fractions (IF) were separated. Blood samples were prepared (0 and 24 h) and coated with LM-1 radioautographic emulsions and percent radioactivity (%rad) in P and BC was determined. Synthesis and biological evaluation of 125I-erythropoietin as a potential radiopharmaceutical agent for tumours BJPS

Erythropoietin (EPO) is a glycoprotein hormone responsible for regulating erythropoiesis. Expression of EPO and EPO receptors (EPOr) has recently been demonstrated in some neoplastic cell lines and tumours, suggesting a potential new target for therapy. In this work, EPO was labeled with iodine-125 using the lactoperoxidase method, known to prevent damage to protein during radioiodination, and labeling conditions were optimized. In vitro stability studies have shown that 125I-EPO is radiochemically stable for 20 days after radiolabeling. In vitro cell binding studies have demonstrated very low binding (<2%) of EPO to normal and neoplastic cell lines tested. Influence of radiation on endotoxin test using the PTS TM for 18-FDG radiopharmaceutical BJPS

F-18 FDG (2-[18-F] fluoro-2-deoxy-D-glucose) is the most frequently used radiopharmaceutical for PET and PET CT imaging exams. The FDA recently approved the use of the PTS TM (Portable Test System) as an alternative to the standard test proposed by the United States Pharmacopeia using the LAL (Limulus Amebocyte Lysates), that takes longer to perform (about 1h) than the PTS TM (15 min). Recent studies have demonstrated that radiation could interfere with the PTS TM test. In order to study the effects of radiation on the PTS TM test and/or equipment, 27 batches of F-18 FDG produced in the Nuclear Engineering Institute were analyzed. The results showed that no direct correlation with radiation was found in any of the cases.
