Rapid detection of Mycobacterium tuberculosis complex (MTBC) is a critical step in controlling tuberculosis (TB). In this study, we used IS6110 as the specific identification target to develop a novel hybridization signal amplification method (HSAM) for the rapid and direct detection of MTBC from clinical sputum specimens. This system consists of magnetic bead-linked capture probes for target isolation, dextranbased nanoparticles for amplifying the reporter molecule (biotinylated-FITC), and detection probes (2B-DNA) for binding the nanoparticles. Assessment of the efficiency of SimPlate™ total plate count color indicator (TPC CI) to quantify mesophilic aerobic microorganisms in pasteurized milk BJM

The SimPlate™ TPC CI system is a rapid method to count mesophilic aerobic microorganisms (MAM) in foods, based on the use of resazurine to indicate bacterial growth. Its efficiency in pasteurized milk was evaluated using 142 pasteurized milk samples (38 type A, 43 type B and 61 type C) collected in Londrina, PR. The standard plating method, using Plate Count Agar (PCA) was used for comparison. The plates of both systems were incubated at 35ºC and read after 24h and 48h. The occurrence of false-positive and false-negative wells and the predominant microorganisms in them were also evaluated. The results were compared by simple correlation and mean variance analyses. Detection of salmonella sp in chicken cuts using immunomagnetic separation BJM

The immunomagnetic separation (IMS) is a technique that has been used to increase sensitivity and specificity and to decrease the time required for detection of Salmonella in foods through different methodologies. In this work we report on the development of a method for detection of Salmonella in chicken cuts using in house antibody-sensitized microspheres associated to conventional plating in selective agar (IMS-plating). First, protein A-coated microspheres were sensitized with polyclonal antibodies against lipopolysacharide and flagella from salmonellae and used to standardize a procedure for capturing Salmonella Enteritidis from pure cultures and detection in selective agar. Performance of two ready-to-use systems for enumeration of aerobic mesophilic microorganisms in frozen goat milk BJM

Enumeration of aerobic microorganisms in Brazilian pasteurized bovine milk using PetrifilmTM AC and SimplateTM TPC was shown to be influenced by the nature of autochthonous microbiota. This study was conducted to assess if the same occurs in pasteurized goat milk. Pasteurized frozen goat milk (n=62) was purchased in retail stores in São Paulo, SP, Brazil, and submitted to mesophilic aerobic counts using PetrifilmTM AC and SimplateTM TPC, comparing results with those of conventional pour-plates. Correlation indexes above 0.90 were ranked "excellent"; those between 0.80 and 0.89, "good", those from 0.79 to 0.50, "fair", and those below 0.50, "poor".
