Background Escherichia coli does not produce n-butanol naturally, but can be butanologenic when related enzymes were expressed using inducible elements on plasmids. In this study we attempted to confer E. coli strain capability of automatic excretion of the chemical by employing a native anaerobic promoter. Also, a novel DNA kit was designed for PCR preparation of linear DNA fragments to perform strain modification. The kit is primarily composed of two mother vectors, co-transformation of linear DNAs into E. coli can simultaneously introduce two butanol synthetic operons into the chromosome and create two in-frame gene deletions at targeted native loci. Results E. coli strain Bw2V carries plasmid pCNA-PHC and pENA-TA, both utilizes native anaerobic... |

A detailed study of putative recombination events and their evolution frequency in the whole genome of the currently known members of the family Tombusviridae, comprising 79 accessions retrieved from the international databases, was carried out by using the RECCO and RDP version 3.31β algorithms. The first program allowed the detection of potential recombination sites in seven out of eight virus genera (Aureusvirus, Avenavirus, Carmovirus, Dianthovirus, Necrovirus, Panicovirus, and Tombusvirus), the second program provided the same results except for genus Dianthovirus. On the other hand, both methods failed to detect recombination breakpoints in the genome of members of genus Machlomovirus. Furthermore, based on Fisher's Exact Test of Neutrality, positive... |

Alu-PCR is a relatively simple technique that can be used to investigate genomic instability in cancer. This technique allows identification of the loss, gain or amplification of gene sequences based on the analysis of segments between two Alu elements coupled with quantitative and qualitative analyses of the profiles obtained from tumor samples, surgical margins and blood. In this work, we used Alu-PCR to identify gene alterations in ten patients with invasive ductal breast cancer. Several deletions and insertions were identified, indicating genomic instability in the tumor and adjacent normal tissue. Although not associated with specific genes, the alterations, which involved chromosomal bands 1p36.23, 1q41, 11q14.3, 13q14.2, occurred in areas of... |

The productive performance of four Holstein-Gir genetic groups (1/2H, 1/4H, 5/8H, 5/8Hinter se) and the effects of non-genetic factors on production traits was evaluated using a data set of 7,951 test-day samples collected between January 1980 and December 1999 from the Arcoverde Experimental Station in the Brazilian state of Pernambuco (Empresa Pernambucana de Pesquisa Agropecuária). The statistical model included the fixed effects of calving year (1980 to 1999) and month (January to December), genetic group of the cow, age at months of calving (33 to 190) and days in milk at each test-day. Random effects were sire, cow and residual with test-days treated as repeated records within each cow within each lactation. For the four genetic groups the milk yield... |

Vibrio parahaemolyticus and Vibrio cholerae are ubiquitous to estuarine and marine environments. Thes two species found in Mediterranean coastal systems can induce infection in humans. Environmental isolates of V. cholerae (n = 109) and V. parahaemotycius (n = 89) sampled at different dates, stations and water salinities were investigated for virulence genes and by a mutilocus sequence-based analysis (MLSA). V. cholerae isolate were all ctxA negative and only one isolate of V. parahaemolyticus displayed trh2 gene. Most Sequence Types (ST) corresponded to uique ST isolated at one date or one station. Frequent recombination events were detected among different pathogenic species, V. parahaemotyticus, V. cholerae, Vibrio mimicus, and Vibrio metoecus.... |

Background: The inhabitants of deep-sea hydrothermal vents occupy ephemeral island-like habitats distributed sporadically along tectonic spreading-centers, back-arc basins, and volcanically active seamounts. The majority of vent taxa undergo a pelagic larval phase, and thus varying degrees of geographical subdivision, ranging from no impedance of dispersal to complete isolation, often exist among taxa that span common geomorphological boundaries. Two lineages of Bathymodiolus mussels segregate on either side of the Easter Microplate, a boundary that separates the East Pacific Rise from spreading centers connected to the Pacific-Antarctic Ridge. Results: A recent sample from the northwest flank of the Easter Microplate contained an admixture of northern... |

Begomoviruses are whitefly-transmitted, single-stranded DNA viruses that are often associated with weed plants. The aim of this study was to further characterize the diversity of begomoviruses infecting weeds (mostly Sida spp.) in Brazil. Total DNA was extracted from weed samples collected in Viçosa (Minas Gerais state) and in some municipalities of Alagoas state in 2009 and 2010. Viral genomes were amplified by RCA, cloned and sequenced. A total of 26 DNA-A clones were obtained. Sequence analysis indicated the presence of 10 begomoviruses. All viral isolates from Blainvillea rhomboidea belonged to the same species, Blainvillea yellow spot virus (BlYSV ), thereby suggesting that BlYSV may be the only begomovirus present in this weed species. Four isolates... |
