The vegetative and root development of the species of Brazilian orchids Cattleya intermedia (John Lindley) and Miltonia clowesii (John Lindley) using different substrates and pyroligneous extract were assessed. Seedlings from in vitro propagation were transferred to vessels with: T1 - Pinus bark (Pi), coconut fibers (Co) and charred peel of rice (Cr); T2 – PiCoCr and vegetal charcoal (Ch), T3 – PiCoCrCh with the charcoal treated with pyroligneous extract (PE); T4 - CoCrChEP and T5 - CoCrCh. In Iran, a large number of Rosa species have been exposed to extinction and therefore preservation techniques are necessary to safeguard their future. In the present investigation, the objectives were to optimize the micropropagation and medium-term conservation of one of these wild species, Rosa pulverulenta. At proliferation stage, the maximum number of new leaves (9.6) were produced on the medium containing 4 µM BAP + 0 µM GA3, whereas the maximum number of axillary shoots (4.1) were observed in the medium containing 4 µM BAP + 3 µM GA3. Este trabalho teve como objetivo verificar o efeito de seis concentrações de ácido indolbutírico (IBA) no enraizamento de estacas herbáceas de figueira (Ficus carica L.) sob nebulização intermitente, em três épocas diferentes. Os ensaios foram instalados em maio/2004, setembro/2004 e janeiro/2005, e, em cada época, o delineamento experimental utilizado foi o inteiramente casualizado, com seis concentrações de IBA (0, 500, 1.000, 1.500, 2.000, 2.500 e 3.000 mg L-1). ABSTRACT Genetic diversity allows identification of potential intraspecific genotypes in the genus Passiflora. The objective of this study was to examine the morphological and genetic diversity of auxin-induced Passiflora mucronata. The experiments were arranged in a complete randomized block design, with a 9 x 2 factorial arrangement (nine genotypes x presence and absence of auxin, indole-3-butyric acid (IBA)), with four replicates of 16 cuttings. The induction and improvement of in vitro rhizogenesis of microshoots of Prosopis chilensis (Mol.) Stuntz and Nothofagus alpina (Poep. et Endl. Oerst.) were compared using Agrobacterium rhizogenes (Ar) versus indole-3-butyric acid (IBA) in the culture media. Microshoots of P. chilensis (1-2 cm length), coming from in vitro grown seedlings, were cultivated in a modified Broadleaved Tree Medium (BTMm) containing half salt concentration of macronutrients and 0.05 mg.L-1 benzilaminopurine (BAP). Callus growth and plant regeneration from long-term callus cultures were studied in two elite clones of Asparagus officinalis cv. Argenteuil, to establish a suitable protocol for a prospective in vitro selection program. Callus initiation and growth was evaluated on MS medium with 3% sucrose, 0.9% agar, 1 mg.l-1 kinetin, and three levels of 2,4-D. The highest callus relative growth was obtained on medium with 1.5 mg.l-1 2,4-D and 1 mg.l-1 kinetin. The effects of plant growth promoting rhizobacteria (PGPR) on the rooting and root growth of semi-hardwood and hardwood kiwifruit stem cuttings were investigated. The PGPR used were Bacillus RC23, Paenibacillus polymyxa RC05, Bacillus subtilis OSU142, Bacillus RC03, Comamonas acidovorans RC41, Bacillus megaterium RC01 and Bacillus simplex RC19. All the bacteria showed indole-3-acetic acid (IAA) producing capacity. Among the PGPR used, the highest rooting ratios were obtained at 47.50% for semi-hardwood stem cuttings from Bacillus RC03 and Bacillus simplex RC19 treatments and 42.50% for hardwood stem cuttings from Bacillus RC03. The work was carried out to study the effects of some auxins and boron in the rooting of kiwi (Actinidia chinensis Planch cv Bruno) stem cuttings.. These cuttings were treated on the base individually with H2O; NAA 300 mg.L-1; IBA 300 mg.L-1; NAA 300 mg.L-1 + Boron; IBA 300 mg.L-1 + Boron; NAA 0,5%-talc and IBA 0,5%-talc. After the treatments, the cuttings were placed in styrofoam trays with vermiculite under moist conditions for 120 days. Starting from semihardwood branches of peach and plum rootstocks (Prunus cerasifera E.) cv. MRS2/5, plum rootstocks (Prunus cerasifera × P. salicina) × (P. cerasifera × P. persica) cv. Ishtarà and olive tree (Olea europaea L.) cv. Frantoio, cuttings were collected and treated with 0 mg.L-1 and 2500 mg.L-1 of indolebutyric acid (IBA) by immersion in the base of the cuttings. From in vitro cultured adult material of Corylus avellana L. cv. Negretta, adventitious rooting of microshoots was evaluated. Rhizogenic induction mediated by two strains of wild-type Agrobacterium rhizogenes (A477 and A478) and indolbutyric acid (IBA) were compared under two light conditions (16:8 h photoperiod and complete darkness). An in vitro culture procedure was developed to induce shoots and roots of Guarianthe skinneri (Orchidaceae) plantlets regenerated from seed-derived protocorms on a Murashige and Skoog (MS) médium supplemented with 6-benzyladenine (BA), indole-3-acetic acid (IAA), a-naftalenacetic acid (NAA) and gibberellic acid (GA3). A máximum of 10.6 shoots was obtained with 16.1 μM NAA, 17.1 μM IAA, 6.3 x 10-9 μM GA3 and 0.0023 μM BA. A máximum of 4.0 roots on each shoot with 5.4 μM NAA, 17.1 μM IAA, 0.001 μM AG3 and 4.6 x 10-9 μM BA. Máximum shoot and root length was obtained with a minimum of GA3, but a máximum IAA.
