One Perkinsus species may hide another: characterization of Perkinsus species present in clam production areas of France ArchiMer
Arzul, Isabelle; Chollet, Bruno; Michel, Justine; Robert, Maeva; Garcia, Celine; Joly, Jean-pierre; Francois, Cyrille; Miossec, Laurence.
Although clam populations in France are known to be infected with protozoans of the genus Perkinsus, no molecular characterization was previously performed on these parasites. Considering that several members of this genus have been associated with mortalities of molluscs worldwide, a study was undertaken in order to characterize these parasites in France. For that purpose, clams, Ruditapes philippinarum and R. decussatus, collected from different production areas and found to be infected with Perkinsus sp. in thioglycolate culture medium, were selected for PCR-RFLP tests and sequencing. Perkinsus olseni was detected in all the investigated areas and results also suggested the presence of P. chesapeaki in Leucate, a lagoon on the Mediterranean coast and in...

Determination of diet in Manila clams by spatial analysis of stable isotopes ArchiMer
Dang, C.; Sauriau, Pierre-guy; Savoye, N.; Caill-milly, Nathalie; Martinez, P.; Millaret, C.; Haure, Joel; De Montaudouin, X..
Spatial changes in the dietary regime of Ruditapes philippinarum from intertidal sediments of Arcachon Bay were depicted by analyzing stable isotope ratios in both its adductor muscles and potential trophic sources. Manila clams were collected from 50 sites in May to June 2006. Dietary reconstruction was based on the IsoSource mixing model, considering trophic enrichments of 3.5%. for carbon and 3.0%. for nitrogen, which were determined experimentally. delta C-13 and delta N-15 values, respectively, averaged -28.2 and 5.2 parts per thousand for riverine particulate organic matter (POM), -20.6 and 4.7 parts per thousand for inner bay phytoplankton, -21.0 and 5.6%. for outer bay phytoplankton, -16.8 and 4.8 parts per thousand. for microphytobenthos, -18.4...

Experimental shift of diet and DIC stable carbon isotopes: Influence on shell delta C-13 values in the Manila clam Ruditapes philippinarum ArchiMer
Poulain, C.; Lorrain, Anne; Mas, R.; Gillikin, D. P.; Dehairs, F.; Robert, Rene; Paulet, Yves-marie.
The influences of diet and seawater dissolved inorganic carbon (DIC) on the carbon isotope composition of shell aragonite (Cinched in the Manila clam Ruditapes philippinarum reared under laboratory conditions were investigated. Clams were exposed to two successive negative carbon isotope shifts: a first shift in diet (delta C-13(phytoplankton)) and a second shift, 35 days later, in DIC (delta C-13(DIC)). Both successive shifts induced a decrease in delta C-13(shell). These results are the first to experimentally confirm an incorporation of respired carbon derived from food and carbon from DIC into shell carbonate of adult bivalves. Skeletal delta C-13 responded to changes in the delta C-13 of both diet and DIC in less than 7 days. Consequently, proxies...

Brown muscle disease (BMD), an emergent pathology affecting Manila clam Ruditapes philippinarum in Arcachon Bay (SW France) ArchiMer
Dang, C; De Montaudouin, X; Gonzalez, P; Mesmer Duldons, N; Caill-milly, Nathalie.
We describe an emerging pathology, brown muscle disease (BMD), which specifically affects the Manila clam Ruditapes philippinarum in Arcachon Bay (France). BMD induces a transformation of the posterior adductor muscle, which becomes infused by conchiolin and calcified, reducing the ability of clams to bury. The disease affects both types of muscular tissue, with striated muscle becoming affected to a higher degree than smooth muscle. Two indices were created to quantify the symptoms: the Muscle Print Index, used for empty and live shells, and the Final Disease Index, utilized for live clams only. Histological sections were made and observed under light microscopy to examine the muscular damage and to investigate a causal agent. Sections revealed an...

Spatio-temporal patterns of perkinsosis in the Manila clam Ruditapes philippinarum from Arcachon Bay (SW France) ArchiMer
Dang, Cecile; De Montaudouin, Xavier; Caill-milly, Nathalie; Trumbic, Zeljka.
Pathogens belonging to the genus Perkinsus infect many bivalve molluscan species around the world, including the Manila clam Ruditapes phihppinarum. We investigated the spatial distribution of this parasite at 34 stations throughout Arcachon Bay (SW France). Prevalence of perkinsosis was 93% and mean infection abundance was 96 x 10(3) cells g(-1) wet gill. Lowest mean abundances were found close to the Leyre River mouth and a significant negative correlation was observed between mean abundance and salinity. Perkinsosis was rare at the oceanic site where salinities and other environmental parameters were stable. A second aim of this study was to survey perkinsosis during annual cycles at 4 sites within Arcachon Bay. Prevalence and intensities (+/-SE) of the...

Diter, Alain.
Methods of gynogenesis, androgenesis and polyploidy induction were developed with the aim of genetic improvement in the rainbow trout, the Pacific oyster, the Manila clam and the black scallop. In the trout: · haploid androgenesis was induced by means of a pressure shock applied on diploid eggs, probably at the time of karyogamy. Diploid androgenetic individuals were directly produced from eggs fertilized with sperm from tetraploid male. · a heat shock treatment effective for suppressing the first egg mitosis was developed. This treatment allows the production of homozygous gynogenetic lines (checked using enzyme electrophoresis technique) from eggs inseminated with ultraviolet irradiated sperm. The treatment also induced tetraploidy in eggs normally...

Bodoy, Alain; Riva, Alain; Maitre-allain, Thierry.
Measurements of oxygen consumption in the two species Ruditapes decussatus and Ruditapes phi/ippinarum were perfonned using hatchery-reared juveniles, grown under controlled conditions. The temperature range varied from 10 to 30°C. The linear relations between the logarithms of the dry weight and of the oxygen consumption were calculated for each species. Rates were compared in terms of standardized clam tissue weight calculated from the regression of respiration on tissue weight. Such a technique allows to make comparisons between the species by avoiding any interference from the variations in individual weight. Within this range of temperature, oxygen consumption of R. philippinarum and of R. decussatus exhibit the same variations, with a maximum at...

The Manila clam population in Arcachon Bay (SW France): Can it be kept sustainable? ArchiMer
Dang, Cecile; De Montaudouin, Xavier; Gam, Meriame; Paroissin, Christian; Bru, Noelle; Caill-milly, Nathalie.
The venerid clam Ruditapes philippinarum is the most prominent suspension-feeding bivalve inhabiting muddy intertidal seagrass beds in Arcachon Bay (SW France). It is exploited by fishermen, and Arcachon Bay ranks number one in France in terms of production and total biomass of this species. Previous studies revealed a decrease in the standing stock of R. philippinarum since 2003 and unbalanced length-frequency distributions with a lack of juveniles and of adults >40 mm. Consequently, the population dynamics of this bivalve were studied at four intertidal sites and one oceanic site in Arcachon Bay. As clam size structure did not allow classical dynamics computations. field monitoring was coupled with field experiments (tagging-recapture) over two years....

Asari clam (Ruditapes philippinarum) in France: history of an exotic species 1972 – 2015 ArchiMer
De Montaudouin, Xavier; Arzul, Isabelle; Caill-milly, Nathalie; Khayati, Alice; Labrousse, Jean-michel; Lafitte, Céline; Paillard, Christine; Soudant, Philippe; Goulletquer, Philippe.
In 1972, France underwent an oyster (Crassostrea angulata) crisis and urgently needed to diversify its aquaculture. Thus, Asari clam (Ruditapes philippinarum) was introduced at that time for aquaculture purpose, concomitantly with the introduction of Pacific oyster (Crassostrea gigas). All Asari clam adults and spat originated from the same area (Puget Sound, WA, USA). After a promising start and the implementation of a national Research & Development program, Asari clam culture rapidly faced a series of concomitant handicaps: firstly, in spite the fact that cultural practices were optimized and locking points addressed, leasing ground availability was limited during the 1980s’ due to certain reluctance from oyster farmers to share their leases and/or...

Caill-milly, Nathalie; Duclercq, Benoit; Morandeau, Gilles.
Between 1980 and 1986, some Arcachon oyster farmers took up breeding the Japanese clam on the foreshore (Robert et Deltreil, 1990). This production was short lived. The steep fall in prices due to economic competition from Spain, Portugal, Italy and Tunisia, predator action such as the trigger fish (Popovsky et al., 1999), added to the significant amount of mortality that occurred in the summer and autumn of 1992 that affected two-thirds of the total stock, led to clam farming being abandoned at the end of the 1980s. In the Arcachon basin, the Japanese clam (Ruditapes philippinarum) population, now natural to the area, has found favourable conditions for development and is expanding rapidly there. This species has been supplanting the local species...

Baud, Jean-pierre; Haure, Joel; Garnier, Jacqueline.
A bulk production of phytoplankton (Skelelonema costatum) has been obtained in concrete tanks from saline groundwater. This was used in various concentrations to feed an intensive cultivation of the Manila clam (Ruditapes philippinarum) in ponds. No difference was observed in growth rates of molluscs, with food concentrations between 31 to 126 µg/l Chl a. However, the growth rates obtained from March to November with a food supplement were 1.5 times higher than those observed in the traditional, extensive cultivation. The biomass doubled and the increase in flesh dry weight was 2.5 times higher. The condition index reached a value of 100 in the intensive cultivation. During growth, the proximate composition analysis revealed that the lipid and carbohydrate...

Asari (= Manila) clam, Ruditapes philippinarum, is the second bivalve mollusc in terms of production in the world and, in many coastal areas, can beget important socio-economic issues. In Europe, this species was introduced after 1973. In Arcachon Bay, after a decade of aquaculture attempt, Asari clam rapidly constituted neo-naturalized population which is now fished. However, recent studies emphasized the decline of population and individual performances. In the framework of a national project (REPAMEP), some elements of fitness, stressors and responses in Arcachon bay were measured and compared to international data (41 publications, 9 countries). The condition index (CI=flesh weight/shell weight) was the lowest among all compared sites. Variation in...

Japanese clam (Ruditapes philippinarum) and European clam (Ruditapes decussates) farming based on spat produced in hatcheries has gone beyond the experimental stage on the Atlantic coasts since the research of Latrouite et Claude (1976), Lucas (1977), Flassch (1979), Dréno (1979) and Gras (soon to be published). It has thus become necessary to develop farming procedures reliable enough to be handed over to the professionals in the business.

Dang, Cecile.
The Manila clam Ruditapes philippinarum was introduced into Arcachon Bay at the beginning of the 1980s for aquaculture purposes. It rapidly naturalized in all intertidal flats of the lagoon. Nowadays, Arcachon Bay ranks at the first French place in terms of exploitable stock. However, recent stock assessments have shown an unbalance size structure with a deficit in juvenile and adult clams (> 38 mm shell length). These alarming patterns asked many questions on the sustainability of Manila clam populations within Arcachon Bay and have motivated the present thesis. This study aimed to better understand the population dynamics of this species and also some factors controlling this dynamic. The final objective was to improve the Manila clam fishing...
