Three selective enrichment broths - selenite cystine (SC), Muller-Kauffmann tetrathionate (MKT) and Rappaport-Vassiliadis (RV) - were compared, for Salmonella Typhimurium isolation from rectal swabs of a calf experimentally infected. The bacteriological procedure involved pre-enrichment in Hajna-GN broth (only for the samples inoculated in RV broth), selective enrichment (SC, MKT and RV broths), culture in modified brilliant green agar (BGA), presumptive biochemistry tests (using triple-sugar-iron agar and lysine-agar) and slide agglutination test with poli-O and poli-H Salmonella antisera. The intraperitoneal inoculation of 3.8 x 10(9) CFU of alive Salmonella typhimurium caused 100% mortality in rats. In these animals a small number of neutrophils migrated to the peritoneal cavity. The intraperitoneal inoculation of the animals with thioglycollate (Tg, 90 mg), carrageenin (Cg, 500 mu g), or killed S. typhimurium (KS, 1.9 x 10(9) CFU) 24 hours before challenge with alive S. typhimurium caused a significant neutrophil migration to the peritoneal cavity (determined 6 hours after challenge) and protected the animals against the lethal effect of the bacteria at a rate of 50%, 83% and 80%, respectively. However, similar treatment of the animals with brain heart infusion broth (BHI, S. typhimurium grout medium) protected the animals by only 23%.... |

El estudio tuvo como objetivo estandarizar y validar la técnica de Reacción en Cadena de la Polimerasa (PCR) para la detección de Salmonella typhimurium (St) en carne de cerdo, res y pollo, optimizando el tiempo y la sensibilidad con relación al método microbiológico; además se realizó el diagnóstico en estos tipos de carnes para evaluar su calidad microbiológica en la Ciudad de Texcoco. Un total de 60 muestras de carne (20 de cerdo, 20 de res y 20 de pollo) de 250 g fueron recolectadas en puntos de venta de la Ciudad de Texcoco. Two constructions in pHS164 vector were designed to permit expression of OV7A and OV4A inserts encoding the N-terminal and C-terminal portion of an aspartic protease from Onchocerca volvulus, respectively. A novel 39 kD protein ompA-OV7A fusion protein was stably expressed as ompA fusion in a modified strain of Salmonella typhimurium strain SL5000 and E.coli strain JM109. Expression of the fusion protein in bacterial strains harboring the constructs were evaluated by western blotting. E.coli and Salmonella lysates were fractionated by 10% SDS-PAGE gel and then immobilized to nitrocellulose membrane by electroblotting.
