Presença de Staphylococcus spp. produtores de enterotoxinas e da toxina da síndrome do choque tóxico em manipuladores de queijo de cabra Arq. Bras. Med. Vet. Zootec.
Rapini,L.S.; Cerqueira,M.M.O.P.; Carmo,L.S.; Veras,J.F.; Souza,M.R..

The use of mice as animal model for testing acute toxicity (LD-50) of toxic shock syndrome toxin Arq. Bras. Med. Vet. Zootec.
Dias,R.S.; Carmo,L.S.; Heneine,L.G.D.; Rocha,P.H.; Barbosa,C.F.; Rodrigues,R.J.; Linardi,V.R..
Acute toxicity test (LD-50) using toxic shock syndrome toxin (TSST-1) was tested in BALB/c, C57BL/6 and Swiss mice. Animals (n = 10) were intraperitoneally injected with TSST-1 (0.01-10.0µg/mouse) followed 4h later by potentiating dose of lipopolysaccharide (75.0µg of LPS - E. coli O111:B4) and cumulative mortality was recorded over 72h. Control animals received either TSST-1 or LPS alone. The data were submitted to qui-Square test and acute toxicity test was calculated by probit analysis (confidence limits expressed as µg toxin/kg). BALB/c mice was the most sensitive (20.0µg/kg, 95% confidence limits: 9.0-92.0) followed by C57BL/6 (38.5µg/kg, 95% confidence limits: 9.11- 401.6). Data from Swiss mice was not conclusive, indicating only low sensitivity....

Characterization of Staphylococcus spp. strains in milk from buffaloes with mastitis in Brazil: the need to identify to species level to avoid misidentification Arq. Bras. Med. Vet. Zootec.
Coimbra-e-Souza,V.; Brito,M.A.V.P.; Chamon,R.C.; Laport,M.S.; Giambiagi-deMarval,M..
ABSTRACT Mastitis is an inflammation of the mammary gland that affects dairy cattle worldwide causing economic losses. Coagulase-negative staphylococci (CNS) are the predominant cause of this type of infection. We have recently showed that coagulase-positive staphylococci could be misidentified. So, the aim of this study was to characterize the Staphylococcus spp. strains initially classified as coagulase-negative Staphylococci, isolated from buffalo with subclinical mastitis. Milk of buffaloes with mastitis in herds was collected and 9 strains were identified as CNS by phenotypic tests. Molecular methodologies latter identified the strains as coagulase-negative Staphylococcus chromogenes (5), coagulase-positive Staphylococcus hyicus (2) and...

Mastite subclínica por Staphylococcus coagulase negativa em ovinos de corte Arq. Bras. Med. Vet. Zootec.
Tejada,T.S.; Silva,D.T.; Dias,P.A.; Conceição,R.C.S.; Müller Neto,H.; Timm,C.D..
Microorganisms that cause mastitis were isolated in a herd of sheep and their sensitivity to antimicrobials was tested. Twenty-one sheep had their mammary glands monitored during lactation. Eighty-eight milk sample from CMT-positive sheep were analyzed in order to isolate the etiologic agent of mastitis. Coagulase negative Staphylococcus were isolated from 29.5% of samples. Ampicillin and penicillin were the antimicrobials with higher number of resistant strains. The importance of coagulase negative staphylococci has been demonstrated, especially S. lentus and S. sciuri as a cause of ovine mastitis.

Anti-staphylococcal effectiveness of nisaplin in refrigerated pizza doughs BABT
Freitas,Wilma Carla de; Souza,Evandro Leite de; Sousa,Cristina Paiva de; Travassos,Antônio Eustáquio Resende.
This study evaluated the effectiveness of nisaplin, commercial product having nisin as active component, in decreasing the staphylococcal population in refrigerated pizza doughs. The refrigerated pizza dough pieces randomly chosen were dipped in the solutions with nisaplin concentrations of 1.0 x 10-3 g and 1.0 x 10-2 g nisaplin/mL named for the treatment A and B and kept under refrigeration (7 °C ± 1 °C). On times 0, 15 and 30 days post treatment the Staphylococcus spp. count was carried out. The results showed that both nisaplin treatments were able to reduce the Staphylococcus spp. count (CFU/g) in the refrigerated pizza doughs. However, only treatment B showed statistically significant reducer effect (p < 0.05) on the count providing a decrease of...

Vancomycin serum concentrations in pediatric oncologic/hematologic intensive care patients BJID
Silva,Dáfne Cardoso Bourguignon da; Seixas,Gláucia Toribio Finoti; Araujo,Orlei Ribeiro de; Arduini,Rodrigo Genaro; Carlesse,Fabianne Altruda de Moraes Costa; Petrilli,Antonio Sergio.
BACKGROUND: Usual treatment regimens with vancomycin often fail to provide adequate serum levels in patients with severe infections. METHODS: Retrospective analysis of vancomycin trough serum measurements. The following parameters were calculated by Bayesian analysis: vancomycin clearance, distribution volume, and peak estimated concentrations. The area under the concentration curve (AUC) (total daily dose/24 h clearance of vancomycin) was used to determine the effectiveness of treatment through the ratio of AUC/minimum inhibitory concentration (MIC) above 400, using MIC = 1 µg/mL, based on isolates of Staphylococci in cultures. RESULTS: Sixty-one vancomycin trough measurements were analyzed in 31 patients. AUC/MIC > 400 was obtained in 34 out of 61...

Methicillin-resistant Staphylococcus lugdunensis carrying SCCmec type V misidentified as MRSA BJID
Pereira,Eliezer Menezes; Schuenck,Ricardo Pinto; Nouér,Simone Aranha; Santos,Kátia Regina Netto dos.
Staphylococcus lugdunensis is a rare cause of severe infections and clinical manifestations are similar to those related to S. aureus infection. We describe a hospital-acquired bacteremia due to methicillin-resistant Staphylococcus lugdunensis, misidentified as methicillin-resistant S. aureus. The oxacillin MIC was 16 µg/mL and the mecA gene and SCCmec type V were determined by PCR. Although treatment had been appropriated, the patient died after rapid progressive respiratory failure and another nosocomial sepsis. It is important not only to identify S. lugdunensis in view of its clinical course, but also to determine its susceptibility to oxacillin by detecting the mecA gene or its product.

Phenotypic and molecular characterization of resistance to macrolides, lincosamides and type B streptogramin of clinical isolates of Staphylococcus spp. of a university hospital in Recife, Pernambuco, Brazil BJID
Pereira,Jussyêgles Niedja da Paz; Rabelo,Marcelle Aquino; Lima,Jailton Lobo da Costa; Neto,Armando Monteiro Bezerra; Lopes,Ana Catarina de Souza; Maciel,Maria Amélia Vieira.
Abstract Introduction There is a mechanism of macrolide resistance in Staphylococcus spp. which also affects the lincosamides and type B streptogramins characterizing the so-called MLSB resistance, whose expression can be constitutive (cMLSB) or inducible (iMLSB) and is encoded mainly by ermA and ermC genes. The cMLSB resistance is easily detected by susceptibility testing used in the laboratory routine, but iMLSB resistance is not. Therapy with clindamycin in cases of infection with isolated iMLSB resistance may fail. Objective To characterize the phenotypic (occurrence of cMLSB and iMLSB phenotypes) and molecular (occurrence of ermA and ermC genes) profiles of MLSB resistance of clinical isolates of susceptible and methicillin-resistant Staphylococcus...

Distribution of erm genes and low prevalence of inducible resistance to clindamycin among staphylococci isolates BJID
Coutinho,Vivian de Lima Spode; Paiva,Rodrigo Minuto; Reiter,Keli Cristine; de-Paris,Fernanda; Barth,Afonso Luis; Machado,Alice Beatriz Mombach Pinheiro.
INTRODUCTION: Resistance to macrolides, lincosamides and streptogramins B (MLS B antibiotics) in staphylococci may be due to modification in ribosomal target methylase encoded by erm genes. The expression of MLS B resistance lead to three phenotypes, namely constitutive resistance (cMLS B), inducible resistance (iMLS B), and resistance only to macrolides and streptogramins B (MS B). The iMLS B resistance is the most difficult to detect in the clinical laboratory. OBJECTIVE: This study investigated the expression of MLS B resistance and the prevalence of the erm genes among 152 clinical isolates of Staphylococcus aureus and coagulase-negative Staphylococcus (CNS) from Hospital de Clínicas de Porto Alegre. METHODS: Primary MLS B resistance was detected by...

Presence of Staphylococcus spp. and Candida spp. in the human oral cavity BJM
Martins,Clélia Aparecida de Paiva; Koga-Ito,Cristiane Yumi; Jorge,Antonio Olavo Cardoso.
The presence of yeasts and staphylococci in the oral cavity is important because they can act as supplementary microbiota and in certain situations can cause oral or systemic diseases. The aim of this work was to study the prevalence of Candida spp. and Staphylococcus spp. in the human oral cavity. Oral rinses were collected from sixty-eight individuals according to the technique described by Samaranayake and MacFarlane and then cultured on Sabouraud medium supplemented with chloramphenicol and Baird-Parker agar. After the incubation period, the microorganisms were isolated and identified through biochemical tests. The data obtained were statistically analysed by ANOVA. Candida spp. were isolated from 61.76% of the examined individuals and C. albicans was...

Staphylococcus spp. in the oral cavity and periodontal pockets of chronic periodontitis patients BJM
Loberto,Jussara Cia S.; Martins,Clélia Ap. de Paiva; Santos,Silvana S. Ferreira dos; Cortelli,José Roberto; Jorge,Antonio Olavo Cardoso.
Staphylococcus spp are not usually isolated from the oral cavity, and when this occurs, they are considered to belong to the transitory microbiota. Individuals with periodontal disease represent possible reservoirs of these opportunistic bacteria in the oral cavity. The use of antibiotics for treatment of periodontal disease or other infections may predispose to the increase of the number of Staphylococcus spp. in the oral cavity. These microorganisms easily become resistant to antibiotics, and may result in superinfection. The purpose of this study was to evaluate the presence of Staphylococcus spp. in the oral cavity and in periodontal pockets of patients with chronic periodontitis, identify the isolates and verify the relationship between the presence...

Habituation of enterotoxigenic Staphylococcus aureus to Origanum vulgare L. essential oil does not induce direct-tolerance and cross-tolerance to salts and organic acids BJM
Tavares,Adassa Gama; Monte,Daniel Farias Marinho do; Albuquerque,Allan dos Reis; Sampaio,Fábio Correia; Magnani,Marciane; Siqueira Júnior,José Pinto de; Souza,Evandro Leite de.
Enterotoxigenic Staphylococcus aureus strains that were isolated from foods were investigated for their ability to develop direct-tolerance and cross-tolerance to sodium chloride (NaCl), potassium chloride (KCl), lactic acid (LA) and acetic acid (AA) after habituation in sublethal amounts (1/2 of the minimum inhibitory concentration - 1/2 MIC and 1/4 of the minimum inhibitory concentration - 1/4 MIC) of Origanum vulgare L. essential oil (OVEO). The habituation of S. aureus to 1/2 MIC and 1/4 MIC of OVEO did not induce direct-tolerance or cross-tolerance in the tested strains, as assessed by modulation of MIC values. Otherwise, exposing the strains to OVEO at sublethal concentrations maintained or increased the sensitivity of the cells to the tested...
