Sucrose synthase molecular marker associated with sugar content in elite sugarcane progeny Genet. Mol. Biol.
Silva,Jorge A. da; Bressiani,Jose A..
We describe the development and application of an expressed sequence tag (EST)-derived restriction fragment length polymorphism (RFLP) marker for sugarcane elite genotypes which can be used for quantitative trait loci (QTL) tagging for sugar content. EST-derived RFLP markers for proteins involved in sucrose metabolism have been used in Southern analysis for mapping and gene tagging in elite sugarcane clones. A single dose marker, obtained from a sucrose synthase EST associated with sugar content at the alpha = 0.01 probability level, is presented for sugarcane breeding. Utilization of EST homologues to known genes for generation of molecular markers accelerated the identification of a QTL controlling an important trait-sugar content. Sugarcane bacterial...

Physiological and Metabolic Responses of Freshwater and Brackish-Water Strains of Microcystis aeruginosa Acclimated to a Salinity Gradient: Insight into Salt Tolerance ArchiMer
Georges Des Aulnois, Maxime; Roux, Pauline; Caruana, Amandine; Réveillon, Damien; Briand, Enora; Hervé, Fabienne; Savar, Veronique; Bormans, Myriam; Amzil, Zouher; Atomi, Haruyuki.
Proliferation of microcystin (MC)-producing Microcystis aeruginosa in brackish waters has been described in several locations and represents a new concern for public and environmental health. While the impact of a sudden salinity increase on M. aeruginosa physiology has been studied, less is known about the mechanisms involved in salt tolerance after acclimation. This study aims to compare the physiological responses of two strains of M. aeruginosa (PCC 7820 and PCC 7806), which were isolated from contrasted environments, to increasing salinities. After acclimation, growth and MC production rates were determined and metabolomic analyses were conducted. For both strains, salinity decreased the biovolume, growth, and MC production rates and induced the...

Carbon sources and polyethylene glycol on soybean somatic embryo conversion PAB
Körbes,Ana Paula; Droste,Annette.
Somatic embryogenesis is an efficient method for the production of target cells for soybean genetic transformation. However, this method still offers low percentages of plant regeneration, and perhaps is related to the maturation process and high morphological abnormalities of the matured embryos. This study aimed to identify a maturation medium that could contribute to the outcome of more efficient plant regeneration results. Embryogenic clusters, derived from cotyledons of immature seeds of the soybean cultivars Bragg and IAS5, were used as starting material for embryos development. Different maturation media were tested by using 6% maltose, 3% sucrose or 6% sucrose, combined with or without 25 g L-1 of the osmotic regulator polyethylene glycol...

Plant architecture of Paspalum vaginatum schwartz modified by nitrate and ammonium nutrition PAB
Beltrano,José; Ronco,Marta Guillermina; Barreiro,Roberto; Montaldi,Edgardo Raúl.
Paspalum vaginatum Schwartz plants were grown under greenhouse conditions in a continuous-flow hydroponic culture, containing NO3- or NH4+or NH4NO3 as nitrogen source. After 30 days, the size of aerial biomass and root system decreased significantly when plants were supplied with NH4+as exclusive nitrogen source. Compared to NO3- treatment, reducing and non-reducing sugars were decreasing together with a significant increase in amino acids content. NH4+-nutrition caused tillers to grow toward an orthogravitropic position (average angle of 68° with respect to the horizontal), and with NO3--nutrition, tillers tended to become diagravitropic (average angle of 23°). With NH4NO3 all the parameters measured had values in between those of the other two sources....

Assimilate partitioning in leaves of the raffinose-storing herb Lamium album L.: photosynthesis and carbon partitioning throughout the photoperiod Rev. Bras. Bot.
Kochhar,A.; Rees,T. Ap; Pollock,C.J..
Lamium album accumulates starch, sucrose and raffinose-family oligosaccharides (RFO) as the major products of photosynthesis. These products were measured in leaves throughout a sixteen-hour photoperiod and under various irradiance conditions. There was continuous accumulation of sucrose and starch. The rate of gas exchange was higher at 500 µEm² s-1 and 900 µEm²s-1 than at 300 µEm² s-1. The rate of photosynthesis did not decline over the sixteen-hour photoperiod, which suggested that there was no short-term feed back inhibition due to sucrose accumulation in this plant. When the products of photosynthesis were compared at the end of the photoperiod, only sucrose increased in abundance at high irradiance. The RFO pool in leaves was shown to contain...

Assimilate partitioning in leaves of the raffinose-storing herb Lamium album L.: the effects of altering source-sink balance Rev. Bras. Bot.
Kochhar,A.; Rees,T. Ap; Pollock,C.J..
In Lamium album, sucrose and raffinose-family oligosaccharides are the major products of photosynthesis that are stored in leaves. Using gas analysis and 14CO2 feeding, we compared photosynthesis and the partitioning of recently-fixed carbon in plants where sink activity was lowered by excision of flowers and chilling of roots with those where sink activity was not modified. Reduction in sink activity led to a reduction in the maximum rate of photosynthesis, to retention of fixed carbon in source leaves and to the progressive accumulation of raffinose-family oligosaccharides. This ultimately affected the extractable activities of invertase and sucrose phosphate synthase. At the end of the light period, invertase activity was significantly higher in treated...

Effects of preculture with sucrose and aba on cell suspensions water status and its relation with vitrification resistance Rev. Bras. Fisiol. Veg.
SEIJO,GUILLERMO.
Changes in cell water relations during precultures were followed in an attempt to understand the mechanism of cell hardening for cryopreservation by vitrification. Medium containing 0.4 M sucrose (psiw=-1.45 MPa) and containing 5 mg L-1 of ABA (MT psiw=-0.73 MPa and MS psiw=-0.48 MPa) were used to harden cell suspensions of orange and carrot. Preculture in these medium did not cause a significant decrease of cell viability, however, it improved the cell survival to PVS2 and liquid nitrogen expositions. When cells were inoculated into medium containing 0.4 M sucrose, the cell psiw decreased rapidly until reach the medium psiw , and turgor was also severely reduced or disappeared. Subsequently, cell psis began to decrease and after 48 h of treatment in low...

Cell suspension cultures of Bauhinia forficata Link, Curcuma zedoaria Roscoe and Phaseolus vulgaris L. were used to test plant ability to utilize an alternative to sucrose as carbon source and energy for growth. Glycerol, sorbitol and galactose were the alternative carbon sources tested. Cell suspension cultures established on liquid medium containing sucrose were transferred to culture medium supplemented with sucrose or glycerol, or sorbitol, or galactose as the sole carbon source. Fresh and dry weight increasing and protein content showed marked differences among the different carbon sources used. Sucrose was the best carbon source for all the three plant species tested. Galactose and glycerol promoted slow or no growth of the three studied species....

Sugarcane maturity estimation through edaphic-climatic parameters Scientia Agricola
Scarpari,Maximiliano Salles; Beauclair,Edgar Gomes Ferreira de.
Sugarcane (Saccharum officinarum L.) grows under different weather conditions directly affecting crop maturation. Raw material quality predicting models are important tools in sugarcane crop management; the goal of these models is to provide productivity estimates during harvesting, increasing the efficiency of strategical and administrative decisions. The objective of this work was developing a model to predict Total Recoverable Sugars (TRS) during harvesting, using data related to production factors such as soil water storage and negative degree-days. The database of a sugar mill for the crop seasons 1999/2000, 2000/2001 and 2001/2002 was analyzed, and statistical models were tested to estimate raw material. The maturity model for a one-year old...

Climatic effects on sugarcane ripening under the influence of cultivars and crop age Scientia Agricola
Cardozo,Nilceu Piffer; Sentelhas,Paulo Cesar.
The lack of information about the effects of cultivars, crop age and climate on the sugarcane (Saccharum ssp.) crop yield and quality has been the primary factor impacting the sugar-ethanol sector in Brazil. One of the processes about which we do not have a satisfactory understanding is sugarcane ripening and the effects of cultivars, crop age and climate on that. Sugarcane ripening is the process of sucrose accumulation in stalks, which is heavily influenced by several factors, mainly by climatic conditions such as air temperature and water deficits. Because it is a complex process, studies of the variables involved in sugarcane ripening can provide important information, resulting in a better use of commercial cultivars, bringing advantages to growers,...

Physiological model to estimate the maturity of sugarcane Scientia Agricola
Scarpari,Maximiliano Salles; Beauclair,Edgar Gomes Ferreira de.
Sugarcane (Saccharum spp.) grows in different weather and management conditions which directly affect crop maturation. These conditions lead to the necessity of quantifying crop response to different stimuli for planning purposes. Forecast models for the quality of raw material are important tools in sugarcane farming, especially the forecast curve of sucrose accumulation in shoots. The goal of these models is to supply yield estimates during the crop cycle, aiming to characterize alternatives and to increase the efficacy for management and strategic decisions. The objective of this project was to develop empiric models capable of obtaining estimates of total recoverable sugar (TRS), for the varieties RB72454, SP81-3250 and SP80-1842 during the crop cycle,...

Sucrose, salicylic acid and citric acid solutions to extent vase life of Vriesea incurvata Gaudich. (Bromeliaceae) floral scapes Acta Agron. (Palmira)
Pulido,Edwin; Rejane-Negrelle,Raquel; Cuquel,Francine Lorena.
Abstract Vriesea incurvata is a native bromeliad from Brazilian Atlantic Rainforest and commercialized as an ornamental pot plant. The morphological characteristics of its floral scape may also indicate it as a new product to use as a cut flower. However, its postharvest behavior was unknown. This study was conducted to determine its vase life by applying distilled water (control) and solutions containing sucrose (50 g L-1), salicylic acid (50 μM), and citric acid (50 g L-1) for periods of 8 and 24 h. Floral scapes maintained in solutions showed vase life greater than 16 days when compared to the control (distilled water). However, solutions with sucrose evidenced the best behaviors related to the maintenance of physiological and aesthetic features during...

Cultivation of immature Capsicum spp. embryos for incompatible-crossing embryo rescue Agronomy
Walter, Rafael; Carvalho, Virginia Silva; Generoso, Andressa Leal; Rodrigues, Rosana; Gravina, Geraldo de Amaral.
The in vitro culture of embryos is an important technique to enable the rescue of embryos from incompatible crosses. Studies analyzing the factors that affect the in vitro culture of zygotic embryos are scarce. This study evaluated the effect of the genotype (Capsicum baccatum and C. frutescens), the composition of the culture medium (MS and ½ MS medium with different concentrations of sucrose, IAA and GA3) and the stage of development (globular, cordiform, torpedo and cotyledonary) in the in vitro culture of immature embryos. Embryo germination was influenced primarily by the stage of development of the embryo and the composition of the culture medium. Regardless of the species, the most suitable culture medium for the germination of globular and...
