Summer mortality of hatchery-produced Pacific oyster spat (Crassostrea gigas). II. Response to selection for survival and its influence on growth and yield ArchiMer

Response to divergent selection for "high" and "low" survival during the summer period, from July to October, was investigated in juvenile (six-month-old) Pacific oysters, Crassostrea gigas, by producing two sets of progenies in 2002 (Generation 2) and three sets of progenies in 2003 (Generation 3). A strict between-family approach was used and resistance of these selected progenies to summer mortality was assessed in three sites along French coasts, to determine their response to selection and estimate realized heritability of the trait. Effects of age and environment on survival of summer mortality by two selected groups of the Pacific oyster Crassostrea gigas ArchiMer

This study investigated the effects of age (first or second summer of life) and environment (three study sites on French coasts) on summer mortality in Crassostrea gigas, using three generations of oysters produced during the Morest project. For each generation, two groups of oysters, selected either as 'resistant' (R) or 'susceptible' (S) to summer mortality at the age of 6 months in Rivière d'Auray (RA, South Brittany), were evaluated. In Ronce (Marennes-Oléron Bay) and RA, R oysters had low mortality during their first and second summers, while mortality was higher during the second summer in Baie des Veys (BDV, Normandy). Hemocyte characteristics in families of oysters, Crassostrea gigas, selected for differential survival during summer and reared in three sites ArchiMer

High variability among individuals is often encountered when hemocyte characteristics are measured in bivalves. Such variability is suspected to result partly from genetic factors. During the MOREST project (2001–2006), oyster summer mortality was extensively investigated in Crassostrea gigas in France. However, significant increases in magnitude and geographic distribution of summer mortality episodes have been reported since 2008. This work contributed to the development of specific genomic tools for the Pacific oyster Crassostrea gigas, aiming to identify molecular markers associated with summer survival of this species. To do so, 29745 ESTs obtained from different sequencing programs were grouped in a unique database. 9058 unigenes have been used to produce a cDNA microarray, to compare lines selected to be Resistant (R) or Sensitive (S) to summer mortality. 34 genes were observed as differentially expressed before the mortality event. Mortality of oysters Crassostrea gigas occurred along the coasts of France during summer 2008, causing a serious crisis in French oyster production. During spring to summer 2008, 5 groups of 1-yr old oysters of different origins and ploidy levels deployed in the Thau lagoon (France) were sampled for biometrical and biochemical analyses; environmental parameters were monitored in the same area. Mortality occurred in 2 episodes: 13 May to 10 June and 24 June to 9 July. Wild-collected oysters showed mortality sooner than other groups but total overall mortality was 85% in all groups. Summer mortality of Crassostrea gigas (Thunberg) in relation to environmental rearing conditions ArchiMer

The purpose of this study is to investigate summer mortality of the cupped oyster, Crassostrea gigas, in relation to culture practices in the traditional oyster production region of Marennes-Oleron (France). Four oyster rearing conditions, varying culture location ("on-" or "off-bottom"), and site depth (65% to 80% daily immersion termed "deep" or 45% to 65%, termed "shallow") were studied to compare biologic performance and maturation status of oysters, in relation to sediment and water column parameters. The most severe mortality occurred in June to July in "on-bottom" reared oysters (25%), as compared with 10% mortality in "off-bottom" cultured oysters. Recherche et identification de bactéries pathogènes au sein d'une écloserie nurserie d'huître creuse, Crassostrea gigas, en relation avec la mortalité estivale du naissain ArchiMer

Summer mortality has been observed on oyster spat Crassostrea gigas since 1991 on french coasts. In our study, we deseribed symptoms observed during the mortality, especially adductor muscle weakness and we isolated strains on Zobell medium and TCBS from moribund spat. Strains isolated during a high mortality (60 %) were selected by experimental pathology on spat. Inoculation by bath led us to select a pathogenic strain (TNEMF6) for oyster spat. Phenotypic identification, phylogenetic study on 16S rDNA and quantitative DNA/DNA hybridisation showed that this strain belongs to Vibrio splendidus biovar II. Phenotyping and ribotyping with strains isolated during the study showed that only TNNIII7 and TNMB5 are similar to the pathogenic strain TNEMF6....

Summer mortality of the Pacific cupped oyster Crassostrea gigas in the Bay of Marennes-Oleron (France) ArchiMer

A comprehensive research program has been developed since 1995 to focus on C. gigas abnormal mortality rates occurring regularly during the summer period in the Bay of Marennes Oleron (France). Several subprojects were carried out including in-vitro ecophysiological studies to asses l) the relationship between reproductive status, scope for growth and mortality, 2) effects of stress temperature on physiological functions. Meanwhile, field studies concerned monitoring surveys of calibrated oyster populations at various emersion time and geographic sites. Summer mortality of adults and juveniles has been reported in the Pacific oyster, Crassostrea gigas, for many years and in several countries. The French multidisciplinary program "Morest" aims to investigate the causes of the summer mortality in Crassostrea gigas. Within this program, we designed multi-site field experiments to assess to what extent genetic variability exists for summer mortality in French populations of C. gigas and to determine whether selective breeding could improve survival.

Recently, the MOREST project ((MORtalitEStivale) a multidisciplinary project (2001-2005) focused on his problem led to a better understanding of this phenomenon in France. This study reports mortality under laboratory conditions in unselected controls and two lines of juvenile Pacific oysters Crassostrea gigas, previously selected for their high or low survival in the field during the summer period. Oysters were also deployed in field conditions and mortality between both conditions was then compared. In the laboratory, mortality was observed in all experiments and it always lasted for a week indicating that mortality under laboratory conditions was a short-term event.
