Low concentrations of doxycycline attenuates FasL-induced apoptosis in HeLa cells Biol. Res.
Yoon,Jung Mi; Koppula,Sushruta; Huh,Se Jong; Hur,Sun Jin; Kim,Chan Gil.
BACKGROUND: Doxycycline (DC) has been shown to possess non-antibiotic properties including Fas/Fas Ligand (FasL)-mediated apoptosis against several tumor types in the concentration range of 10-40 μg/mL. However, the effect of DC in apoptotic signaling at much low concentrations was not studied. METHODS: The present study investigated the attenuation effect of low dose of DC on FasL-induced apoptosis in HeLa cell by the methods of MTT assay, fluorescence microscopy, DNA fragmentation, flow cytometry analysis, and western blotting. RESULTS AND CONCLUSION: In the present findings we showed that low concentration of DC (<2.0 μg/mL) exhibited protective effects against FasL-induced apoptosis in HeLa cells. FasL treatment to HeLa cells...

Influence of tetracycline in the hepatic and renal development of rat's offspring BABT
Machado,Ana Lourdes da Silva; Brandão,Adriana Aigotti Haberbeck; Silva,Cláudia Maria de Oliveira Monteiro da; Rocha,Rosilene Fernandes da.
This study aimed on evaluating the possible effects of tetracycline administered to rats on the tenth day of pregnancy, on kidney and liver development of their offspring. The liver showed vacuolization, necrosis, inflammation and sinusoidal dilatations, more evident in the newborn. Mitosis, early increase of Kupffer cells population and hipertrophy of hepatocytes with greater synthesis of glycogen were present on the tenth and twentieth days of life. The kidney showed slight tubular vacuolizations and necrosis, more proeminent in the newborn, as well as signs of tubular regeneration on the tenth and twentieth days. These results suggested that the organs studied went through several transitory morphological changes during development but presented signs...

Biologically Synthesized Silver Nanoparticles for Enhancing Tetracycline Activity Against Staphylococcus aureus and Klebsiella pneumoniae BABT
Hussein,Elsayed Alsaied Masoud; Mohammad,Ali Al-Hajry; Harraz,Farid Abourageh; Ahsan,Mohd Faisal.
Abstract Phytochemical content of plant extracts can be used effectively to reduce the metal ions to nanoparticles in one-step green synthesis process. In this study, six plant extracts were used for the synthesis of silver nanoparticles (AgNPs). Biologically synthesized AgNPs was characterized using UV-Vis Spectrophotometer, Field Emission Scanning Electron Microscope (FE-SEM), X-ray diffraction (XRD), Energy Dispersive X-ray spectroscopy (EDX) and Fourier Transform Infrared (FTIR) spectroscopy. The individual and combined effects of AgNPs and tetracycline against S. aureus and K. pneumoniae were assessed. Ginger, onion and sidr extracts supported AgNPs formation while arak, garlic and mint extracts failed to convert the silver ions to AgNPs. The present...

Mini-Mu insertions in the tetracycline resistance determinant from Proteus mirabilis BJMBR
The inducible tetracycline resistance determinant isolated from Proteus mirabilis cloned into the plasmid pACYC177 was mutagenized by insertion of a mini-Mu-lac phage in order to define the regions in the cloned sequences encoding the structural and regulatory proteins. Three different types of mutants were obtained: one lost the resistance phenotype and became Lac+; another expressed the resistance at lower levels and constitutively; the third was still dependent on induction but showed a lower minimal inhibitory concentration. The mutant phenotypes and the locations of the insertions indicate that the determinant is composed of a repressor gene and a structural gene which are not transcribed divergently as are other known tetracycline determinants...

Presence of tetracycline resistant bacteria and genes in grassland-based animal production systems Ciencia e Investigación Agraria
López,Liliana; Santamaría,Johanna; Sánchez,Alexandra; Castro,Lida; Moreno,Jessica L.
This study assessed the presence of tetracycline-resistant bacteria in five different grassland-based production systems dedicated to raising dairy cattle located in the Colombian Andes. Animal (ruminal fluid and feces) and environmental (soil and water) samples were evaluated, and resistant heterotrophic bacteria were found in rumen fluid, feces, soil, and runoff water samples. The resistant bacteria were isolated and identified based on the 16S rDNA region. Subsequently, they were evaluated for the presence of tet genes, which encode ribosomal protection proteins and membrane efflux pumps. The most frequent phyla detected were Firmicutes and Proteobacteria. The most common resistance genes found were tet(W), tet(Q), and tet(M). The nucleotide sequences...
