Transcriptional activity of the 5'-flanking region of the thyroid transcription factor-1 gene in human thyroid cell lines Genet. Mol. Biol.
Yamada,Hiroya; Takano,Toru; Matsuzuka,Fumio; Watanabe,Mikio; Miyauchi,Akira; Iwatani,Yoshinori.
Thyroid transcription factor-1 (TTF-1, NKX2-1) is a homeodomain-containing transcriptional factor that binds to and activates the promoters of thyroid and lung-specific genes, such as thyroglobulin, thyroid peroxidase, and thyroid stimulating hormone receptor. TTF-1 is known to play a key role in the development of the thyroid. However, the precise mechanism of TTF-1 gene transcription in human thyroid cells has not been studied. The expression of transcriptional activity in various lengths of the 5'-flanking region of the human TTF -1 gene was studied in TTF-1 positive and negative human thyroid cell lines. Increased transcriptional activity was observed in thyroid cell lines containing plasmids that coded for a sequence proximal to the transcription...

The Incidence of the Foramen Thyroideum in the South African Population International Journal of Morphology
Ramsaroop,L; Hurrinarain,K; Partab,P; Satyapal,K. S.
The foramen thyroideum is described as an occasional opening existing in one or both laminae of the thyroid cartilage which may or may not contain a neurovascular component. Foramen thryoideum was first described in the literature by Segond in 1847. Some authors consider its existence a structural variation rather than an anomaly, with classical texts of anatomy providing little detail when describing this foramen. This study was undertaken to investigate the incidence and characteristics of the foramen thyroideum in the South African population. A total of 80 formalin fixed cadaveric laryngeal specimens (obtained from the Department of Clinical Anatomy, University of KwaZulu-Natal, South Africa) were dissected. Larynges were dissected with the aid of a...

Biochemical Parameters and Histopathological Findings in the Forced Molt Laying Hens Rev. Bras. Ciênc. Avic.
Mert,N; Yildirim,BA.
ABSTRACT The aim of this study was to evaluate the effects of forced molting using biochemical parameters and histopathological findings in laying hens. 36 Hyline W36 strain laying hens, 90 weeks old were chosen for this research. Eight of these chickens were randomly selected and placed in a cage as the control group before the molting program began. All the others 28 chickens were used for the forced molting program. Eight laying hens were slaughtered at the end of the molting program named as molting group. The remaining 20 hens were fed for 37 days, weighted and slaughtered when they reached the maximum egg production (80%) as postmolting group. Then, blood was analyzed for malondialdehyde, glutathione, catalase, glucose, calcium, phosphorus, albumin,...

Model of induction of thyroid dysfunctions in adult female mice Arq. Bras. Med. Vet. Zootec.
Ferreira,E.; Silva,A.E.; Serakides,R.; Gomes,A.E.S.; Cassali,G.D..
It is described the elaboration of a protocol to induce hyperthyroidism and hypothyroidism in mice by administrating thyroxin and propylthiouracil, respectively, in the drinking water. The drugs were administered to adult female mice of the Swiss strain for 30 days in order to obtain a systemic status of thyroid dysfunction. The induction of hyperthyroidism and hypothyroidism in the animals was confirmed by the histomorphological analysis of the thyroid in the end of the experiment, when the state of gland dysfunction in the animals submitted to the treatment was observed.

Histomorphometric and functional studies of the rat thyroid throughout the estrous cycle Arq. Bras. Med. Vet. Zootec.
Serakides,R.; Nunes,V.A; Nascimento,E.F.; Serra,G.V..
The influence of the estrous cycle on the thyroid gland was studied. Twenty one five-to-seven-month-old Wistar female rats were divided according to the stage of the estrous cycle in two groups: metaestrus-diestrus and proestrus-estrus. After gross inspection, the thyroids were weighed, sampled, and processed for staining with hematoxilyn-eosin. Seric concentrations of total T4, free T4, total T3, TSH, progesterone, and estradiol were measured. The values of estradiol were significantly higher in the proestrus-estrus stage. However, no significant differences in the plasmatic concentrations of progesterone, free T4, total T4 and TSH throughout the cycle were found. The results of the morphometric study of the thyroid did not indicate any significant...

Propylthiouracil-induced hypothyroidism delays apoptosis during the first wave of spermatogenesis Biol. Res.
Silva,Doris; Lizama,Carlos; Tapia,Verónica; Moreno,Ricardo D.
Mammalian germ cell apoptosis plays a key role in controlling the correct number of germ cells supported by Sertoli cells during the first wave of spermatogenesis in mammalian puberty. However, little is known about hormonal factors that could influence the rate of germ cell apoptosis during puberty or adulthood. In this work we evaluate germ cell apoptosis under hypothyroidism induced by goitrogen propylthiouracil (PTU) during the first wave of spermatogenesis. Neonatally administered PTU promoted a delay in the differentiation of Sertoli cells as evaluated by the expression of clusterin using immunohistochemistry and RT-PCR. Clusterin had different expression levels in control and PTU-treated animals, but under both conditions the highest levels were...

Thyroid cell irradiation by radioiodines: a new Monte Carlo electron track-structure code BABT
Champion,Christophe; Elbast,Mouhamad; Wu,Ting-Di; Colas-Linhart,Nicole.
The most significant impact of the Chernobyl accident is the increased incidence of thyroid cancer among children who were exposed to short-lived radioiodines and 131-iodine. In order to accurately estimate the radiation dose provided by these radioiodines, it is necessary to know where iodine is incorporated. To do that, the distribution at the cellular level of newly organified iodine in the immature rat thyroid was performed using secondary ion mass microscopy (NanoSIMS50). Actual dosimetric models take only into account the averaged energy and range of beta particles of the radio-elements and may, therefore, imperfectly describe the real distribution of dose deposit at the microscopic level around the point sources. Our approach is radically different...

Changes in the thyroid gland during the reproductive cycle of the male vespertilionid bat, Scotophilus heathi BJB
KRISHNA,A.; SINGH,K..
The aim of present study was to compare the changes in thyroid gland with the reproductive cycle of S. heathi. Thyroid showed marked seasonal variation in weight, quantity of colloid and follicular epithelial height, suggesting the thyroid gland to be inactive during quiescence and winter dormancy and active during the time of recrudescence and breeding similarly to the testicular cycle. Plasma thyroxin (T4) concentration showed a significant seasonal change with high concentration during breeding and post-breeding and low concentration during quiescence. However, the T4 concentration increased from breeding to post-breeding phase, when the testes weight was declining. It is suggested that in S. heathi the positive correlation between thyroid and...

Thyroid peroxidase activity is inhibited by amino acids BJMBR
Carvalho,D.P.; Ferreira,A.C.F.; Coelho,S.M.; Moraes,J.M.; Camacho,M.A.S.; Rosenthal,D..
Normal in vitro thyroid peroxidase (TPO) iodide oxidation activity was completely inhibited by a hydrolyzed TPO preparation (0.15 mg/ml) or hydrolyzed bovine serum albumin (BSA, 0.2 mg/ml). A pancreatic hydrolysate of casein (trypticase peptone, 0.1 mg/ml) and some amino acids (cysteine, tryptophan and methionine, 50 µM each) also inhibited the TPO iodide oxidation reaction completely, whereas casamino acids (0.1 mg/ml), and tyrosine, phenylalanine and histidine (50 µM each) inhibited the TPO reaction by 54% or less. A pancreatic digest of gelatin (0.1 mg/ml) or any other amino acid (50 µM) tested did not significantly decrease TPO activity. The amino acids that impair iodide oxidation also inhibit the TPO albumin iodination activity. The inhibitory amino...

Thyroid function in post-weaning rats whose dams were fed a low-protein diet during suckling BJMBR
This study was designed to evaluate the thyroid and pituitary hormone levels in post-weaning rats whose dams were fed a low-protein diet during suckling (21 days). The dams and pups were divided into 2 groups: a control group fed a diet containing 22% protein that supplies the necessary amount of protein for the rat and is the usual content of protein in most commercial rat chow, and a diet group fed a low-protein (8%) diet in which the protein was substituted by an isocaloric amount of starch. After weaning all dams and pups received the 22% protein diet. Two hours before sacrifice of pups aged 21, 30 and 60 days, a tracer dose (0.6 µCi) of 125I was injected (ip) into each animal. Blood and thyroid glands of pups were collected for the determination of...

Effect of 30 mCi radioiodine on multinodular goiter previously treated with recombinant human thyroid-stimulating hormone BJMBR
Paz-Filho,G.J.; Mesa-Junior,C.O.; Olandoski,M.; Woellner,L.C.; Goedert,C.A.; Boguszewski,C.L.; Carvalho,G.A.; Graf,H..
Recombinant human thyroid-stimulating hormone (rhTSH) enhances 131I uptake, permitting a decrease in radiation for the treatment of multinodular goiter (MNG). Our objective was to evaluate the safety and efficacy of a single 0.1-mg dose of rhTSH, followed by 30 mCi 131I, in patients with MNG. Seventeen patients (15 females, 59.0 ± 13.1 years), who had never been submitted to 131I therapy, received a single 0.1-mg injection of rhTSH followed by 30 mCi 131I on the next day. Mean basal thyroid volume measured by computed tomography was 106.1 ± 64.4 mL. 131I 24-h uptake, TSH, free-T4, T3, thyroglobulin, anti-thyroid antibodies, and thyroid volume were evaluated at regular intervals of 12 months. Mean 131I 24-h uptake increased from 18.1 ± 9.7 to 49.6 ± 13.4%...

Estradiol decreases iodide uptake by rat thyroid follicular FRTL-5 cells BJMBR
Furlanetto,T.W.; Nunes Jr.,R.B.; Sopelsa,A.M.I.; Maciel,R.M.B..
Estradiol has well-known indirect effects on the thyroid. A direct effect of estradiol on thyroid follicular cells, increasing cell growth and reducing the expression of the sodium-iodide symporter gene, has been recently reported. The aim of the present investigation was to study the effect of estradiol on iodide uptake by thyroid follicular cells, using FRTL-5 cells as a model. Estradiol decreased basal iodide uptake by FRTL-5 cells from control levels of 2.490 ± 0.370 to 2.085 ± 0.364 pmol I-/µg DNA at 1 ng/ml (P<0.02), to 1.970 ± 0.302 pmol I-/µg DNA at 10 ng/ml (P<0.003), and to 2.038 ± 0.389 pmol I-/µg DNA at 100 ng/ml (P<0.02). In addition, 4 ng/ml estradiol decreased iodide uptake induced by 0.02 mIU/ml thyrotropin from 8.678 ± 0.408 to...

Biphasic modulation of insulin receptor substrate-1 during goitrogenesis BJMBR
Grozovsky,R.; Morales,M.M.; Carvalho,D.P..
Insulin receptor substrate-1 (IRS-1) is the main intracellular substrate for both insulin and insulin-like growth factor I (IGF-I) receptors and is critical for cell mitogenesis. Thyrotropin is able to induce thyroid cell proliferation through the cyclic AMP intracellular cascade; however, the presence of either insulin or IGF-I is required for the mitogenic effect of thyroid-stimulating hormone (TSH) to occur. The aim of the present study was to determine whether thyroid IRS-1 content is modulated by TSH in vivo. Strikingly, hypothyroid goitrous rats, which have chronically high serum TSH levels (control, C = 2.31 ± 0.28; methimazole (MMI) 21d = 51.02 ± 6.02 ng/mL, N = 12 rats), when treated with 0.03% MMI in drinking water for 21 days, showed...

Pituitary-thyroid axis in short- and long-term experimental diabetes mellitus BJMBR
Short-term experimental diabetes mellitus (DM) produces a significant decrease in serum thyroid hormones, a decreased or normal serum thyroid-stimulating hormone (TSH) and a reduction in hepatic and renal T4-5'-deiodination. However, little is known about the effects of chronic diabetes mellitus on the pituitary-thyroid axis function. We evaluated the changes induced by very short-term (6 days), short-term (15 days) and chronic (6 months) streptozotocin-induced diabetes mellitus in 3-month old female Dutch-Miranda rat serum T4, serum TSH and T4-5'-deiodinase activity in the thyroid and pituitary glands. Serum hormones were determined by specific radioimmunoassays. Iodothyronine-5'-deiodinase activities were assayed in the thyroid and pituitary microsomal...

Autoradiographic thyroid evaluation in short-term experimental diabetes mellitus BJMBR
Nascimento-Saba,C.C.A.; Brito,A.C.; Pereira,M.J.S.; Carvalho,J.J.; Rosenthal,D..
Previous studies have shown that in vitro thyroid peroxidase (TPO) iodide oxidation activity is decreased and thyroid T4-5'-deiodinase activity is increased 15 days after induction of experimental diabetes mellitus (DM). In the present study we used thyroid histoautoradiography, an indirect assay of in vivo TPO activity, to determine the possible parallelism between the in vitro and in vivo changes induced by experimental DM. DM was induced in male Wistar rats (about 250 g body weight) by a single ip streptozotocin injection (45 mg/kg), while control (C) animals received a single injection of the vehicle. Seven and 30 days after diabetes induction, each diabetic and control animal was given ip a tracer dose of 125I (2 µCi), 2.5 h before thyroid excision....

Lack of mutations of exon 2 of the MEN1 gene in endocrine and nonendocrine sporadic tumors BJMBR
Costa,S.C.; Nascimento,L.S.; Ferreira,F.J.; Mattos,P.S.; Camara-Lopes,L.H.; Ward,L.S..
In addition to the mutations that underlie most cases of the multiple endocrine neoplasia type 1 (MEN1) syndrome, somatic mutations of the MEN1 gene have also been described in sporadic tumors like gastrinomas, insulinomas and bronchial carcinoid neoplasm. We examined exon 2 of this gene, where most of the mutations have been described, in 148 endocrine and nonendocrine sporadic tumors. DNA was obtained by phenol/chloroform extraction and ethanol precipitation from 92 formalin-fixed, paraffin-embedded samples, and from 40 fresh tumor tissue samples. We used 5 pairs of primers to encompass the complete coding sequence of exon 2 of the MEN1 gene that was screened by the polymerase chain reaction-single-stranded conformation polymorphism (PCR-SSCP) technique...
