RESUMO: Visando ao estabelecimento de um protocolo de assepsia para a micropropagação de diferentes cultivares de mangueira, foram coletadas no campo, por um período de dois anos (2000-2001), microestacas e folhas jovens e tratadas no laboratório com hipoclorito de sódio (v/v) 2% com Tween 80 a 0,025% por 15 minutos no caso de microestacas e, 0,5; 1,0; 1,5 e 2% com Tween 80 a 0,06 por 5 minutos no caso de folhas jovens. As microestacas, depois da assepsia com hipoclorito de sódio foram tratadas também com Benomyl 200mg/L por 30 minutos, ficando, em seguida, em magentas com vermiculita umedecidas em água estéril. RESUMO: A eliminação de microrganismos é uma condição essencial para o estabelecimento de um processo de regeneração in vitro, pois eles além de competirem pelos nutrientes também produzem toxinas que causam a morte do explante. O trabalho visou desenvolver metodologias de controle de crescimento de microrganismos provenientes de explantes de mangueira inoculados em meio de cultura, como fase inicial do estabelecimento de um protocolo de micropropagação de gemas laterais de mangueira para apoiar o programa de melhoramento desenvolvido na Embrapa. Abstract: The objective of this work was to evaluate the effects of temperature and osmotic agents on the in vitro conservation of Cape gooseberry (Physalis peruviana). Temperatures at 18 and 25°C, as well as the osmotic agents sucrose, mannitol, and sorbitol were tested. A short-term in vitro conservation of Cape gooseberry can be achieved at 18°C, using 30 g L-1 sucrose. |

ABSTRACT: The objective of the present research was to develop a protocol for micropropagation of Anthurium bonplandii and Anthurium maricense by direct organogenesis. Nodal segments, with two or three nodes, were used as explants. The cultures were kept in a growth chamber at a temperature of 25±2ºC, under a photoperiod of 16 hours and a luminosity of 30μmol m-2 s-1. At 60 days, the number of regenerated buds per explant (NBE) was counted.
