Chloroplast localization of Cry1Ac and Cry2A protein- an alternative way of insect control in cotton Biol. Res.
Muzaffar,Adnan; Kiani,Sarfraz; Khan,Muhammad Azmat Ullah; Rao,Abdul Qayyum; Ali,Arfan; Awan,Mudassar Fareed; Iqbal,Adnan; Nasir,Idrees Ahmad; Shahid,Ahmad Ali; Husnain,Tayyab.
BACKGROUND: Insects have developed resistance against Bt-transgenic plants. A multi-barrier defense system to weaken their resistance development is now necessary. One such approach is to use fusion protein genes to increase resistance in plants by introducing more Bt genes in combination. The locating the target protein at the point of insect attack will be more effective. It will not mean that the non-green parts of the plants are free of toxic proteins, but it will inflict more damage on the insects because they are at maximum activity in the green parts of plants. RESULTS: Successful cloning was achieved by the amplification of Cry2A, Cry1Ac, and a transit peptide. The appropriate polymerase chain reaction amplification and digested products confirmed...

Transgenic lettuce seedlings carrying hepatitis B virus antigen HBsAg BJID
Marcondes,Jackson; Hansen,Ekkehard.
The obtainment of transgenic edible plants carrying recombinant antigens is a desired issue in search for economic alternatives viewing vaccine production. Here we report a strategy for genetic transformation of lettuce plants (Lactuca sativa L.) using the surface antigen HBsAg of hepatitis B virus. Transgenic lettuce seedlings were obtained through the application of a regulated balance of plant growth regulators. Genetic transformation process was acquired by cocultivation of cotyledons with Agrobacterium tumefaciens harboring the recombinant plasmid. It is the first description of a lettuce Brazilian variety "Vitória de Verão" genetically modified.

Metabolism and root exudation of organic acid anions under aluminium stress Braz. J. Plant Physiol.
Mariano,Eduardo D.; Jorge,Renato A.; Keltjens,Willem G.; Menossi,Marcelo.
Numerous plant species can release organic acid anions (OA) from their roots in response to toxic aluminium (Al) ions present in the rooting medium. Hypothetically OA complex Al in the root apoplast and/or rhizosphere and thus avoid its interaction with root cellular components and its entry in the root symplast. Two temporal patterns of root OA exudation are observed. In pattern I, OA release is rapidly activated after the contact of the root with Al ions while in pattern II there is a lag phase between the addition of Al and the beginning of OA release. Compounds other than OA have been detected in root exudates and are also correlated with Al resistance in plants. Plant species like buckwheat and tea show mechanisms of Al tolerance, which confer them...

Genotypic variation of Agrobacterium-mediated transformation of Italian ryegrass Electron. J. Biotechnol.
Lee,Ki-Won; Choi,Gi Jun; Kim,Ki-Yong; Yoon,Sei Hyung; Ji,Hee Chung; Park,Hyung Soo; Lim,Young Chul; Lee,Sang-Hoon.
In the present study, genotypic variation of Agrobacterium-mediated transformation of Korean Italian ryegrass has been evaluated. Mature seed-derived calli of seven cultivars were inoculated and co-cultured with Agrobacterium tumefaciens carrying the binary vector pCAMBIA1301, which contains a reporter gene (gus) and a plant selectable marker gene conferring resistance to hygromycin (hpt) in the T-DNA region. The effects of several factors such as callus type and callus age on transformation effectiveness and the expression of the GUS gene were investigated. The highest transformation effectiveness (6.7%) was obtained with the Hwasan 101 cultivar when 9-week-old calli (type-I) were inoculated with Agrobacterium. The overall transformation rates of the...

Utilization of genes encoding osmoprotectants in transgenic plants for enhanced abiotic stress tolerance Electron. J. Biotechnol.
Khan,Mohammad Sayyar; Ahmad,Dawood; Khan,Muhammad Adil.
Global agriculture in the context of growing and expanding populations is under huge pressure to provide increased food, feed, and fiber. The recent phenomenon of climate change has further added fuel to the fire. It has been practically established now that the global temperature has been on the increase with associated fluctuations in annual rainfall regimes, and the resultant drought and flood events and increasing soil and water salinization. These challenges would be met with the introduction and utilization of new technologies coupled with conventional approaches. In recent years, transgenic technology has been proved very effective in terms of production of improved varieties of crop plants, resistant to biotic stresses. The abiotic stresses such as...

Introduction of antifungal genes in sunflower via Agrobacterium Electron. J. Biotechnol.
Radonic,Laura Mabel; Zimmermann,Julián Marcos; Zavallo,Diego; López,Nilda; López Bilbao,Marisa.
There is evidence that overexpression of transgenes codifying antifungal proteins may confer protection to pathogen attack, and that this protection is increased due to the synergic effect of the expression of two or more genes. On the other hand it is well known that sunflower is a recalcitrant specie, highly difficult to be genetically transformed. In this context, the final aim of this project was to obtain sunflower plants expressing at least two antifungal genes, avoiding sequential transformation. The antifungal genes used encode for two enzymes that degrade the fungal wall (glucanase and chitinase), an osmotin and a ribosome inhibitor protein. Two types of transformation vectors were used: a more traditional system with a double cassette and a novel...

Soybean genetic transformation: a valuable tool for the functional study of genes and the production of agronomically improved plants Genet. Mol. Biol.
Homrich,Milena Schenkel; Wiebke-Strohm,Beatriz; Weber,Ricardo Luís Mayer; Bodanese-Zanettini,Maria Helena.
Transgenic plants represent an invaluable tool for molecular, genetic, biochemical and physiological studies by gene overexpression or silencing, transposon-based mutagenesis, protein sub-cellular localization and/or promoter characterization as well as a breakthrough for breeding programs, allowing the production of novel and genetically diverse genotypes. However, the stable transformation of soybean cannot yet be considered to be routine because it depends on the ability to combine efficient transformation and regeneration techniques. Two methods have been used with relative success to produce completely and stably transformed plants: particle bombardment and the Agrobacterium tumefaciens system. In addition, transformation by Agrobacterium rhizogenes...

Effect of soybean ureases on seed germination and plant development Genet. Mol. Biol.
Rechenmacher,Ciliana; Wiebke-Strohm,Beatriz; Oliveira-Busatto,Luisa A. de; Polacco,Joseph C.; Carlini,Célia R.; Bodanese-Zanettini,Maria H..
Abstract Urease catalyzes the hydrolysis of urea to ammonia and carbon dioxide. The ammonia (nitrogen (N) product of urease activity) is incorporated into organic compounds. Thus, urease is involved in N remobilization, as well as in primary N assimilation. Two urease isoforms have been described for soybean: the embryo-specific, encoded by the Eu1 gene, and the ubiquitous urease, encoded by Eu4. A third urease-encoding gene was recently identified, designated Eu5, which encodes the putative protein product SBU-III. The present study aimed to evaluate the contribution of soybean ureases to seed germination and plant development. Analyses were performed using Eu1/Eu4/Eu5-co-suppressed transgenic plants and mutants of the Eu1 and Eu4 urease structural genes,...
