In 2005, draft sequences of the genomes of Trypanosoma brucei, Trypanosoma cruzi and Leishmania major, also known as the Tri-Tryp genomes, were published. These protozoan parasites are the causative agents of three distinct insect-borne diseases, namely sleeping sickness, Chagas disease and leishmaniasis, all with a worldwide distribution. Despite the large estimated evolutionary distance among them, a conserved core of ~6,200 trypanosomatid genes was found among the Tri-Tryp genomes. Extensive analysis of these genomic sequences has greatly increased our understanding of the biology of these parasites and their host-parasite interactions. A study was conducted in 24 gilts to determine the effect of Trypanosoma brucei infection on their reproductive efficiency. The infected gilts developed clinical trypanosomosis following a prepatent period of 2-3 days with 1.8 x 10⁶ trypanosomes per gilt. The clinical signs were observed intermittent fever, pale mucus membranes, short moist cough, moist rales, mucopurulent ocular discharges and hyperemia of the skin, reduced feed intake, and loss of body condition, recumbency, uncoordinated movements, posterior paresis and death of gilts. The cause of death in the pigs was pneumonia caused by Escherichia coli. Grossly, the lungs were severely congested and had undergone gray hepatization. With the hypothesis that African trypanosomes could have in vivo specific genes for adaptation to host’s environment, the present study was conducted by using suppressive subtractive hybridization (SSH) technique to seek the highly expressed genes especially in host. A total of 328 clones from the in vivo SSH library and that of 160 clones from the in vitro SSH library were analyzed in order to determine their expression levels, but none of the above-mentioned genes showed differential expression. This indicates that no trypanosome genes could be differentially expressed either the in vivo or in vitro propagated trypanosomes.
