We present the karyotypic characterization of 26 specimens of the side-necked turtle Hydromedusa tectifera collected in the upper Iguaçu River, Paraná state, Brazil. The turtles were cytogenetically analyzed using Giemsa staining and other banding techniques (C, G, Ag-NOR and CMA3) as well as fluorescence in situ hybridization (FISH) with a rDNA 18S probe. All the specimens showed a diploid number of 58 composed of 22 macro and 36 microchromosomes. The Ag-NOR, CMA3 and FISH techniques permitted the identification and characterization of the chromosome pairs bearing nucleolus organizer regions (NORs), while G-banding facilitated a better recognition and pairing of macrochromosomes. The localization of peroxidase activity in different cell regions is used as a criterion for the classification of the stage of maturation of mammalian mononuclear phagocytes with a positive peroxidase reaction indicating the presence of monoblasts, promonocytes, monocytes and macrophages. In this study it was evaluated the peroxidase activity of blood mononuclear phagocytes of this turtle detected at different stages of differentiation. The present observations suggest that, in turtles, the differentiation of mononuclear phagocytes occur in the blood circulation, in contrast to animals, where only are monocytes in circulating blood and macrophage differentiation occurs in other body compartments |

カメの体幹部は甲羅に覆われているため、単純X線撮影では得られる情報が少ない。そこで今回、ミシシッピーアカミミガメに対するX線CTによる断層撮影検査の有用性について検討した。まず基礎的検討として、臨床的に健康な6匹を用い、軟部組織、肺および骨のCT値を計測し、その結果からそれぞれの組織の観察に適したウインドウを設定した。また、病理学的検索の結果、全ての個体において肝臓の脂肪変性が認められ、肝臓のCT値が低い個体ほど変性の程度が大きく、CT値の高い個体ほど変性の程度が小さかった。このことから、肝臓のCT値の差により脂肪変性の程度を診断することが可能であった。さらに、臨床例についてCT検査を実施したところ、それぞれ、消化管内異物、肺炎および卵塞の診断が可能であった。以上の成績より、ミシシッピーアカミミガメの臨床診断において従来の単純X線撮影では読影が困難であった体幹部軟部組織について、CTにより詳細な情報を得ることが可能であった。よって、カメの臨床診断にはCTによる断層画像診断の積極的な利用が望まれる。 In turtles,conventional radiographic techniques give little information on the interna1 organs because of the density of the shell. Phrynops geoffroanus (Schweigger, 1812) is the freshwater turtle with the widest geographical distribution in South America. During 2006, physical examination and hematological evaluation were performed on free-ranging turtles from two polluted rivers, the Piracicaba River (n = 51) and its tributary Piracicamirim (n = 42), in southeastern Brazil. Red blood cell and thrombocyte counts, mean corpuscular volume and mean corpuscular hemoglobin levels differed in turtles from the two water courses. Although free-ranging turtles showed ectoparasites and boat propeler lesions, animals apparently had no signs of clinical disease. The biology and ecology of South American turtles is still poorly known, particularly, for the Brazilian species. Laboratory studies are essential to understand the life cycles of aquatic turtles species and to help in formulating management plans for their conservation. As a contribution to the knowledge of Podocnemis erythrocephala species, we give a description of its species-typical behaviors, categorized as: maintenance, locomotion, feeding, agonistic and reproduction, based on captives observations of four pairs of turtles in an aquarium in Manaus, Brazil. Similarities and differences with the repertoires of other turtle species are discussed, concluding that turtles have much more complex adaptative strategies and social life than was believed. |
