Etude ecologique des vibrions du bassin d'Arcachon ArchiMer

Water and sediment samples monthly collected from Arcachon Bay (Gironde, France) were examined during three years for the presence of marine vibrios. V. cholerae serotype 01 was never detected. On the other hand, the Eyre River and mouth contain non-agglutinating V. cholerae (127 strains) principally in water. The low temperatures slow their development down without stopping it completely. V. parahaemolyticus (257 strains) present in the same areas is particularly isolated from water when the aquatic medium temperature rises to 15 degree C. V. alginolyticus (638 strains) settles the whole Bay, except the river, in any season. The low temperatures promote growth of V. metschnikovii (203 strains) very tolerant towards salinity. Lysozyme gene expression and hemocyte behaviour in the Mediterranean mussel, Mytilus galloprovincialis, after injection of various bacteria or temperature stresses ArchiMer

The aim of the present study was to evaluate the expression of the Mytilus galloprovincialis lysozyme gene in different in vivo stress situations, including injection of bacteria Vibrio splendidus LGP32, Vibrio anguillarum or Micrococcus lysodeikticus, as well as heat shock at 30 C and cold stress at 5 C. Injection of V. splendidus LGP32 resulted in: (i) a general down-regulation of lysozyme gene expression, as quantified by Q-PCR; (ii) reduction in the number of circulating hemocytes; (iii) decrease in the percentage of circulating hemocytes expressing lysozyme mRNA which was now restricted to only small cells, as observed by ISH; and (iv) accumulation of hemocytes expressing lysozyme in the muscle sinus where injection took place. Effect of dietary water content on European sea bass (Dicentrarchus labrax) growth and disease resistance ArchiMer

The effect of dietary water content on Dicentrarchus labrax growth parameters and resistance to Vibrio anguillarum infection was investigated using commercial pellets with identical energy contents and different moisture levels. The first experiment hypothesis was that moisturizing pelleted ration can have an impact on Dicentrarchus labrax growth performance by the osmoregulation energy cost reduction. In a second time, the experiment explores the effect of water addition in pellets on the fish resistance to a disease. A specific device was built to uniformly moisturize dry pellets to different moisture levels, i.e. 8%, 20%, 40% and 60%. Bacterial infections occurred during the development of marine fish farming of species such as turbot, sea-bass, sea-bream or salmon and trout. The main-specific bacteria involved in these diseases observed belong to 3 groups: the Vibrionaceae for Vibrio anguillarum (type 408) and Aeromonas salmonicida , the causative agents of respectively vibriosis and furinculosis; the Corynebacteriaceae for Renibacterium salmoninarum , responsible for bacterial kidney disease (BKD), the Enterobacteriaceae for Yersinia ruckeri , the causative agent of enteric redmouth disease. Uptake of <i>Vibrio anguillarum</i> vaccine by <i>Artemia salina</i> as a potential oral delivery system to fish fry OMA

An ELISA to detect <i>Vibrio anguillarum</i> antigen in <i>Artemia salina</i> was developed. This assay was then used to determine the time course of antigen uptake by <i>Artemia</i> during incubations with <i>Vibrio</i> vaccine with a view to using <i>Artemia</i> as a vaccine carrier for oral vaccination of fish fry. The rate of uptake of vaccine by <i>Artemia</i> was shown to be concentration dependent. At a dose of 1.5x10<sup>7</sup> cfu ml<sup>-1</sup>, 1 mg wet weight of <i>Artemia</i> took up 1x10<sup>5</sup> cfu within 30min and reached maximum of 6x10<sup>6</sup> cfu after 60min. The use of API 20E for the identification of <i>Vibrio anguillarum</i> and <i>V. ordalii</i> OMA

Sixty-eight strains of <i>Vibrio anguillarum</i>, five of <i>V. ordalii</i> and the type strains of <i>V. alginolyticus, V. carchariae, V. damsela</i> and <i>V. parahaemolyticus</i> were compared using the API 20E gallery. Within the <i>V. anguillarum</i> strains, distinct groups could be separated mainly on the basis of their reaction on indole production and the fermentation of amygdalin and arabinose. <i>Vibrio ordalii</i>, the former <i>V. anguillarum</i> biotope 2, could easily be separated from <i>V. anguillarum</i> and from the other fish pathogenic <i>Vibrio</i> spp.

Extended serotyping scheme for <i>Vibrio anguillarum</i> with the definition and characterization of seven provisional O-serogroups OMA

The present paper summarizes the serotyping scheme of the fish pathogenic bacterium <i>Vibrio anguillarum</i> and defines seven additional O-serogroups. Strains, collected in our laboratories that were nontypable with antisera against the previously defined 16 O-serotypes, were used for generating new antisera and were characterized further by means of LPS profiles, Western blots, and serological reactions. On the basis of the results, it is suggested that the seven new O-serogroups are to be included in the existing serotyping system as serotypes O17-O<sub>2</sub>3. There is a need for a standard in vitro procedure to infect larval fish with bacteria. Such a procedure could be used as a tool to evaluate fish quality, verify fish sensitivity to a certain bacterial strain or evaluate the pathogenicity of a bacterium towards a given fish species. The availability of a challenge test is a prerequisite in studies dealing with the effect of different medicinal treatments on diseased fish. An oral challenge test would offer the possibility to investigate the routes of infection via the digestive tract, i.e. after feeding contaminated feeds. Comparative serology of the marine fish pathogen <i>Vibrio anguillarum</i> OMA

The different serotyping systems, based on thermostable O antigens, reported for <i>Vibrio anguillarum</i> and <i>V. ordalii</i> were compared by quantitative agglutination, sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and subsequent silver staining or Western blotting (immunoblotting) of purified lipopolysaccharide (LPS), using polyclonal rabbit antisera.
