Assessment of genetic diversity among Pakistani wheat (Triticum aestivum L.) advanced breeding lines using RAPD and SDS-PAGE Electron. J. Biotechnol.
Ahmed,Muhammad Farooq; Iqbal,Muhammad; Masood,Muhammad Shahid; Rabbani,Malik Ashiq; Munir,Muhammad.
Genetic diversity was assessed among 32 advanced wheat breeding lines included in the National Uniform Wheat Yield Trials (2006-07) of Pakistan using molecular (DNA) and biochemical (SDS-PAGE) markers. Of the 72 RAPD primers used for initial screening, 15 were found polymorphic. A total of 140 bands (61.4% polymorphic) were generated by the 15 random decamer primers. Genetic similarity coefficients ranged from 0.81 to 0.94 for rainfed and from 0.70 to 0.93 for the normal seeding date group. Cluster analysis using the unweighted pair group method of arithmetic averages (UPGMA) clustered the 32 advanced wheat breeding lines into one major and three small groups. Maximum level of polymorphism (90%) was observed for the primer OPA-05. Lines N9 and N11 showed...

Genetic variability for waxy genes in Argentinean bread wheat germplasm Electron. J. Biotechnol.
Vanzetti,Leonardo Sebastián; Pflüger,Laura Alicia; Rodríguez-Quijano,Marta; Carrillo,José Maria; Helguera,Marcelo.
Amylose and amylopectin are the two polysaccharides that constitute starch in bread wheat and the enzyme GBSSI (Granule-bound starch synthase I), also known as waxy protein, is responsible for amylose synthesis in storage tissues. Decrease of the amylose content in starch has been associated with the lack of waxy protein(s). In this work, different sets of PCR markers were used to characterize the genetic variability of waxy loci from 103 Argentinean bread wheat cultivars. For the Wx-A1 locus, Wx-A1a and a novel molecular allele designed Wx-A1g were detected. Wx-B1 locus showed three alleles (Wx-B1a, Wx-B1b, Wx-B1e), and Wx-D1 locus showed only the Wx-D1a allele. Novel single-locus allele specific markers for Wx-A1b, Wx-B1b and Wx-D1b null alleles were...

Identification of croplands of winter cereals in Rio Grande do Sul state, Brazil, through unsupervised classification of normalized difference vegetation index images REA
Junges,Amanda H.; Fontana,Denise C.; Pinto,Daniele G..
This study aimed to propose methods to identify croplands cultivated with winter cereals in the northern region of Rio Grande do Sul State, Brazil. Thus, temporal profiles of Normalized Difference Vegetation Index (NDVI) from MODIS sensor, from April to December of the 2000 to 2008, were analyzed. Firstly, crop masks were elaborated by subtracting the minimum NDVI image (April to May) from the maximum NDVI image (June to October). Then, an unsupervised classification of NDVI images was carried out (Isodata), considering the crop mask areas. According to the results, crop masks allowed the identification of pixels with greatest green biomass variation. This variation might be associated or not with winter cereals areas established to grain production. The...

IDENTIFICATION OF HOMOGENEOUS RAINFALL ZONES DURING GRAIN CROPS IN PARANÁ, BRAZIL REA
Lopes,Allan R.; Marcolin,Jonatas; Johann,Jerry A.; Boas,Márcio A. Vilas; Schuelter,Adilson R..
ABSTRACT The aim of this study is to identify homogeneous rainfall zones in the winter and summer 1st and 2nd crops, in the state of Paraná, Brazil. The zones were defined by clustering using the expectation-maximization (EM) algorithm to transform seasonal rainfall series. Monthly average rainfall data collected from 157 weather stations for 20 years (1996 to 2015) were employed. The results show that the number of homogeneous zones varied among growing seasons. The summer crop presented two clusters, with rainfall averages of 1489 and 1925 mm; the second crop presented four clusters, with averages of 1849, 1004, 1454, and 1182 mm; and the winter crop had three clusters, with averages of 969, 1498, and 1171 mm. Clustering was a useful instrument to...

Molecular cytogenetic characterization of parental genomes in the partial amphidiploid Triticum aestivum x Thinopyrum ponticum Genet. Mol. Biol.
Brasileiro-Vidal,Ana Christina; Cuadrado,Angeles; Brammer,Sandra P.; Benko-Iseppon,Ana Maria; Guerra,Marcelo.
The wheat line PF 839197 and six hybrid derivatives from a cross between PF 839197 and Thinopyrum ponticum were cytologically characterized by fluorescent in situ hybridization (FISH). Probes for the 5S and 45S rDNA genes (pTa794 and pTa71, respectively), a highly repetitive rye sequence (pSc119.2), the synthetic oligonucleotide (AAG)5, and total genomic DNA from Th. ponticum and rye were used. In the wheat line, a 1RS.1BL translocation was revealed by the labeling patterns produced with pSc119.2 and (AAG)5, and confirmed by genomic in situ hybridization (GISH) using rye genomic DNA as a probe. Analyses of partial amphiploids confirmed previous results indicating mitotic instability, with a tendency to stabilize at 2n = 42 or 56. GISH with Th. ponticum...

Combining ability analysis for grain filling duration and yield traits in spring wheat (Triticum aestivum L. em. Thell.) Genet. Mol. Biol.
Kamaluddin,; Singh,Rishi M.; Prasad,Lal C.; Abdin,Malik Z.; Joshi,Arun K..
A diallel analysis of wheat (Triticum aestivum L. em. Thell) parents (n = 11) and their F1 (n = 55) and F2 (n = 55) offspring was carried out for the following four traits: grain filling duration (GFD), GFD for growing degree days (GDD), 1000 seed weight and seed yield per plant. Analysis of variance for general combining ability (GCA) and specific combining ability (SCA) displayed significant F1 and F2 general and specific combining ability effects for the four traits studied. For all the traits the GCA effects were relatively more important than the SCA effects, indicating that additive genetic effects were predominant. Crosses displaying high SCA effects for grain filling duration, seed weight and yield were observed to be derived from parents having...

Quantitative inheritance of some wheat plant traits Genet. Mol. Biol.
Novoselovic,D.; Baric,Marijana; Drezner,G.; Gunjaca,J.; Lalic,A..
The objective of this study was to estimate gene effects and genetic variability for some quantitative traits of two winter wheat crosses (Soissons/Zitarka and Soissons/Sana) by generation mean analysis. In most cases a digenic epistatic model was sufficient to explain variation in generation means. The additive-dominance model was adequate for plant height and grain weight per spike of the longest culm. In two cases (grain yield per plant and single grain weight) these models failed to explain variation in generation means, implying the presence of higher order interactions or interactions between linked loci. Dominance effects and additive x additive epistasis were more important than additive effects and other epistatic components. Only complementary...

Molecular characterization of Indian pathotypes of Puccinia striiformis f. sp. tritici and multigene phylogenetic analysis to establish inter- and intraspecific relationships Genet. Mol. Biol.
Aggarwal,Rashmi; Kulshreshtha,Deepika; Sharma,Sapna; Singh,Vaibhav K.; Manjunatha,Channappa; Bhardwaj,Subhash C.; Saharan,Mahender S..
Abstract Stripe rust caused by Puccinia striiformis f. sp. tritici (Pst) is one of the most devastating diseases of wheat (Triticum spp.) worldwide. Indian isolates were characterised based on their phenotypic reaction on differential hosts carrying different Yr genes. Based on virulence/avirulence structure, isolates were characterised into ten different pathotypes viz. 70S0-2, 67S64, 70S4, 66S0, 70S64, 66S64-1, 38S102, 47S102, 46S119, and 78S84. These Indian pathotypes of P. striiformis f. sp. tritici and 38 pathotypes of other rust species (P. graminis tritici and P. triticina) were used in this study to analyze their molecular phylogenetic relationship. The nucleotides of rDNA-ITS, partial β-tubulin and ketopantoate reductase genes of all the...

Comparison of radiosensitivity response to acute and chronic gamma irradiation in colored wheat Genet. Mol. Biol.
Hong,Min Jeong; Kim,Dae Yeon; Ahn,Joon-Woo; Kang,Si-Yong; Seo,Yong Weon; Kim,Jin-Baek.
Abstract We aimed to investigate the biological responses induced by acute and chronic gamma irradiation in colored wheat seeds rich in natural antioxidants. After acute and chronic irradiation, the phenotypic effects on plant growth, germination rate, seedling height, and root length were examined, and the biochemical changes were investigated by analyzing the expression of antioxidant enzyme-related genes, antioxidant enzyme activities, and total antioxidant capacity. High dosages of chronic radiation reduced plant growth compared with the controls. Electron spin resonance measurement and 2,2-diphenyl-1-picrylhydrazyl activity analysis showed lower amount of free radicals in colored wheat seeds on chronic irradiation with low dosage of gamma rays...

WRKY transcription factors have been extensively characterized in the past 20 years, but in wheat, studies onWRKY genes and their function are lagging behind many other species. To explore the function of wheat WRKY genes, we identified a TaWRKY68 gene from a common wheat cultivar. It encodes a protein comprising 313 amino acids which harbors 19 conserved motifs or active sites. Gene expression patterns were determined by analyzing microarray data of TaWRKY68 in wheat and of orthologous genes from maize, rice and barley using Genevestigator. TaWRKY68 orthologs were identified and clustered using DELTA-BLAST and COBALT programs available at NCBI. The results showed that these genes, which are expressed in all tissues tested, had relatively higher levels in...

Molecular characterization of a cryptic wheat-Thinopyrum intermedium translocation line: evidence for genomic instability in nascent allopolyploid and aneuploid lines Genet. Mol. Biol.
Yingshan,Dong; Xiuling,Bu; Yushi,Luan; Mengyuan,He; Bao,Liu.
A putative translocation line (#32), together with a disomic addition line (TAI27) and an octo-amphiploid line (Zhong3) of common wheat and Thinopyrum intermedium were characterized by restriction fragment length polymorphism (RFLP) analysis, using probes covering all seven homoeologous groups of Triticeae. Line 32 was confirmed to be a cryptic translocation line, based on the detection of multiple introgressed hybridization fragments specific to Th. intermedium in the RFLP patterns, and the absence of a hybridization signal in GISH analysis. In addition, extensive genomic changes, as compared to the wheat parent, were detected on all three lines studied, with a great majority of changes showing concordance among the lines. Our data is consistent with the...

Microsatellite polymorphism in wheat from Brazilian cultivars; inter- and intra-varietal studies Genet. Mol. Biol.
Lima,Vitor Lopes de Abreu; Seki,Homiko Abreu; Rumjanek,Franklin David.
Eleven samples of wheat (Triticum aestivum) from different Brazilian cultivars and six American varieties were compared for polymorphism, using primers for nine different STR loci. STR analysis of DNA from single grains of the Brazilian varieties showed that for most loci there was very little intra-cultivar polymorphism. The polymorphic variation observed for Brazilian cultivars was similar to that seen in the American varieties. For the Brazilian cultivars PCR analysis could be performed on only one half of a grain. The American samples required more seeds for analysis. The nucleotide sequences of five amplified microsatellites selected at random from the Brazilian samples were also determined and compared to those of the Chinese Spring variety. Although...

Genetic structure of Argentinean hexaploid wheat germplasm Genet. Mol. Biol.
Vanzetti,Leonardo S.; Yerkovich,Nadia; Chialvo,Eugenia; Lombardo,Lucio; Vaschetto,Luis; Helguera,Marcelo.
The identification of genetically homogeneous groups of individuals is an ancient issue in population genetics and in the case of crops like wheat, it can be valuable information for breeding programs, genetic mapping and germplasm resources. In this work we determined the genetic structure of a set of 102 Argentinean bread wheat (Triticum aestivum L.) elite cultivars using 38 biochemical and molecular markers (functional, closely linked to genes and neutral ones) distributed throughout 18 wheat chromosomes. Genetic relationships among these lines were examined using model-based clustering methods. In the analysis three subpopulations were identified which correspond largely to the origin of the germplasm used by the main breeding programs in Argentina.

Antioxidant responses of wheat plants under stress Genet. Mol. Biol.
Caverzan,Andréia; Casassola,Alice; Brammer,Sandra Patussi.
Abstract Currently, food security depends on the increased production of cereals such as wheat (Triticum aestivum L.), which is an important source of calories and protein for humans. However, cells of the crop have suffered from the accumulation of reactive oxygen species (ROS), which can cause severe oxidative damage to the plants, due to environmental stresses. ROS are toxic molecules found in various subcellular compartments. The equilibrium between the production and detoxification of ROS is sustained by enzymatic and nonenzymatic antioxidants. In the present review, we offer a brief summary of antioxidant defense and hydrogen peroxide (H2O2) signaling in wheat plants. Wheat plants increase antioxidant defense mechanisms under abiotic stresses, such...

Abstract The D-genome progenitor of wheat (Triticum aestivum), Aegilops tauschii, possesses numerous genes for resistance to abiotic stresses, including drought. Therefore, information on the genetic architecture of A. tauschii can aid the development of drought-resistant wheat varieties. Here, we evaluated 13 traits in 373 A. tauschii accessions grown under normal and polyethylene glycol-simulated drought stress conditions and performed a genome-wide association study using 7,185 single nucleotide polymorphism (SNP) markers. We identified 208 and 28 SNPs associated with all traits using the general linear model and mixed linear model, respectively, while both models detected 25 significant SNPs with genome-wide distribution. Public database searches...

Epistasis and genotype-by-environment interaction of grain protein content in durum wheat Genet. Mol. Biol.
Bnejdi,Fethi; Gazzah,Mohamed El.
Parental, F1, F2, BC1 and BC2 generations of four crosses involving four cultivars of durum wheat (Triticum durum Desf.) were evaluated at two sites in Tunisia. A three-parameter model was found inadequate for all cases except crosses Chili x Cocorit 71 at site Sidi Thabet and Inrat 69 x Karim at both sites. In most cases a digenic epistatic model was sufficient to explain variation in generation means. Dominance effects (h) and additive x additive epistasis (i) (when significant) were more important than additive (d) effects and other epistatic components. Considering the genotype-by-environment interaction, the non-interactive model (m, d, h, e) was found adequate. Additive variance was higher than environmental variance in three crosses at both sites....
