Genetic diversity analysis of Bemisia tabaci populations in Pakistan using RAPD markers Electron. J. Biotechnol.
Hameed,Saima; Hameed,Shahid; Sadia,Munazza; Malik,Salman A.
Background: The Bemisia tabaci is one of the most devastating pests of agricultural crops and ornamental plants worldwide. The genetic diversity and biotype status of the Bemisia tabaci in Pakistan was assessed by using random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR). A total 80 samples of B. tabaci collected from 14 districts of the Punjab province and 7 districts of the Sindh province were included. Results: All 10 primers screened in this study generated 151 scorable amplification products, of which 117 or 77% were polymorphic. Pairwise Nei and Li’s similarity had ranged from 0.25 to 0.88 among all individuals analyzed. Based on Nei and Li’s similarity coefficients Bemisia populations were grouped into 3 main clusters and clearly...

Diversity analysis of Bemisia tabaci biotypes: RAPD, PCR-RFLP and sequencing of the ITS1 rDNA region Genet. Mol. Biol.
Rabello,Aline R.; Queiroz,Paulo R.; Simões,Kenya C.C.; Hiragi,Cássia O.; Lima,Luzia H.C.; Oliveira,Maria Regina V.; Mehta,Angela.
The Bemisia tabaci complex is formed by approximately 41 biotypes, two of which (B and BR) occur in Brazil. In this work we aimed at obtaining genetic markers to assess the genetic diversity of the different biotypes. In order to do that we analyzed Bemisia tabaci biotypes B, BR, Q and Cassava using molecular techniques including RAPD, PCR-RFLP and sequencing of the ITS1 rDNA region. The analyses revealed a high similarity between the individuals of the B and Q biotypes, which could be distinguished from the BR individuals. A phylogenetic tree based on ITS1 rDNA sequence was constructed. This is the first report of the ITS1 rDNA sequence of Bemisia tuberculata and of the BR biotype of B. tabaci.

Genetic diversity of Bemisia tabaci (Genn.) Populations in Brazil revealed by RAPD markers Genet. Mol. Biol.
Lima,L.H.C.; Campos,L.; Moretzsohn,M.C.; Návia,D.; Oliveira,M.R.V. de.
Bemisia tabaci (Genn.) was considered a secondary pest in Brazil until 1990, despite being an efficient geminivirus vector in beans and soybean. In 1991, a new biotype, known as B. tabaci B biotype (=B. argentifolii) was detected attacking weed plants and causing phytotoxic problems in Cucurbitaceae. Nowadays, B. tabaci is considered one of the most damaging whitefly pests in agricultural systems worldwide that transmits more than 60 different plant viruses. Little is known about the genetic variability of these populations in Brazil. Knowledge of the genetic variation within whitefly populations is necessary for their efficient control and management. The objectives of the present study were to use RAPD markers (1) to estimate the genetic diversity of B....

A review of geminivirus diseases in vegetables and other crops in Brazil: current status and approaches for management Horticultura Brasileira
Inoue-Nagata,Alice K; Lima,Mirtes F; Gilbertson,Robert L.
ABSTRACT: Insect-transmitted viruses cause some of the most damaging and economically important diseases of crop plants, especially in tropical and subtropical regions. The geminiviruses, a group of single-stranded DNA viruses with unique twined icosahedral virus particles, are responsible for many of these diseases. Of particular importance are the geminiviruses transmitted by whiteflies (Bemisia tabaci), which are in the genus Begomovirus. This is the largest genus of plant viruses (in terms of number of species), and some are responsible for many devastating diseases in vegetable and fiber crops throughout the world. This review presents the current understanding of begomoviruses, the diseases they cause, and approaches for disease management. An...

Influence of trichomes on attractiveness and ovipositional preference of Bemisia tabaci (Genn.) B biotype (Hemiptera: Aleyrodidae) on tomato genotypes Neotropical Entomology
Oriani,Maria A de G; Vendramim,José D.
Brazil is one of the world's largest tomato producer, but considerable part of the production is lost due to the attack of Bemisia tabaci (Genn.) B biotype. Resistant germoplasm plants can be an important method for B. tabaci control in integrated pest management approaches. The attractiveness and ovipositional preference of B. tabaci for 17 tomato genotypes were evaluated in a free-choice test. Trials were set up in a randomized block design with ten replicates. Each replicate (one tomato plant per pot) was placed in a cage (80 x 50 x 50 cm) and infested with 1,000 adults during four days. Linear correlation tests were applied between the number of insects and eggs and number of trichomes in each tomato genotype. LA716, LA444-1 and PI134418 genotypes were...

Biological parameters of Bemisia tabaci (Gennadius) biotype B (Hemiptera: Aleyrodidae) on Jatropha gossypiifolia, commercial (Manihot esculenta) and wild cassava (Manihot flabellifolia and M. carthaginensis) (Euphorbiaceae) Neotropical Entomology
Carabalí,Arturo; Belloti,Anthony C; Montoya-Lerma,James.
Bemisia tabaci (Gennadius) is one of the most important pests of cassava in Africa and several countries of Asia due to the damage caused by direct feeding, the excretion of honeydew, and its capacity as a vector of cassava mosaic geminivirus. There is a general consensus that B. tabaci is a complex of morphologically indistinguishable populations with different biotypes. In the Americas, the polyphagous biotype B does not appear to feed on cassava. Recent studies indicate that it is possible, however, for biotype B to gradually adapt to cassava using phylogenetically related hosts. Therefore, the possibility that some wild species of cassava constitute intermediate hosts in the adaptation process may lead to the establishment of biotype B on commercial...

Assessment of physiological aspects of three tomato genotypes infested by Bemisia tabaci (Gennadius) biotype B (Hemiptera: Aleyrodidae) Neotropical Entomology
Toscano,Luciana C.; Boiça Júnior,Arlindo L.; Maruyama,Wilson I..
The performance of three tomato genotypes (PI 1134417, LA 716 and of Bruna VFN hybrid) was assessed during infestation by Bemisia tabaci (Gennadius) biotype B, in relation to the physiological aspects of plants attacked and not attacked by the insect. The experiment was carried out in a greenhouse at environmental temperature. Thirty-day-old plants were artificially infested with 100 insects/plant for ten days. The Bruna VFN hybrid was damaged physiologically with lower height and mean number of leaves per plant than in the non attacked plants, ten days after infestation. Also, it presented shorter root length and decrease in canopy and root dry matter when compared to the non attacked plants. The wild genotypes (PI 1344417 and LA 716) did not present...

Estrutura genética populacional de Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) utilizando marcadores microssatélites Neotropical Entomology
Valle,GE do; Zucchi,MI; Stabellini,NS; Lourenção,AL; Pinheiro,JB.
We aimed to characterize the population genetic structure within and among five Bemisia tabaci (Gennadius) populations collected from different host plants and geographic regions by using microssatelites as a molecular marker. Each population was represented by 19 specimens. The host plants and geographic origins of these populations were described as follows: Pop 1: Squash Barreiras (BA); Pop 2: Cotton Barreiras (BA); Pop 3: Soybean Campinas (SP); Pop 4: Tomato Cruz das Almas (BA); and Pop 5: Soybean Rondonópolis (MT). Six polymorphic loci were observed, which discriminated 31 different alleles in the studied populations, with a mean number of alleles per population of 3.30 (2.67 - 4.00). Using Fisher's Exact test, it was observed that at least three...

Does the presence of weeds affect Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) infestation on tomato plants in a semi-arid agro-ecosystem? Neotropical Entomology
Bezerra,Mary-Ann S.; Oliveira,Maria R.V. de; Vasconcelos,Simão D..
Integrated Bemisia tabaci (Gennadius) management requires quantitative knowledge on the mechanisms of persistence of pest populations in non-commercial host plants. We investigated B. tabaci oviposition preference among tomato and four weed species, and the insect population fluctuation under field conditions in the semi-arid region. At weekly intervals, we determined the level (number of eggs and nymphs per leaf) and the density of infestation (number of eggs and nymphs per leaf area) in all species. We observed an oviposition preference gradient among the species, with different degrees of attractiveness; Acanthospermum hispidum was the most infested species whereas Amaranthus deflexus was the least infested one. We also detected a significantly higher...

Characterization of Tomato yellow spot virus, a novel tomato-infecting begomovirus in Brazil PAB
Calegario,Renata Faier; Ferreira,Sávio de Siqueira; Andrade,Eduardo Chumbinho de; Zerbini,Francisco Murilo.
The objective of this work was the biological and molecular characterization of a begomovirus detected in São Joaquim de Bicas, Minas Gerais, Brazil, named TGV-[Bi2], by determining its host range, complete nucleotide sequence and phylogenetic relationships with other begomoviruses. Biological characterization consisted of a host range study using either sap inoculation or particle bombardment as inoculation methods. The yellow spot virus can infect plants in Solanaceae and Amaranthaceae, including economically importat crops as sweet pepper, and weeds as Datura stramonium and Nicotiana silvestris. For the molecular characterization, the full-length genome (DNA-A and DNA-B) was amplified, cloned and completely sequenced. Sequence comparisons and...

Toxicity of neem oil to Bemisia tabaci biotype B nymphs reared on dry bean PAB
Pinheiro,Patricia Valle; Quintela,Eliane Dias; Oliveira,Jaison Pereira de; Seraphin,José Carlos.
The objective of this work was to determine the most susceptible nymphal stage of Bemisia tabaci biotype B to neem (Azadirachta indica A. Juss.) oil applied to dry bean (Phaseolus vulgaris L.) in a screenhouse. A solution of commercial oil (Dalneem) extracted from neem seeds was sprayed directly on each nymphal instar at 0, 0.1, 0.25, 0.5, 1 and 2% concentrations for lethal concentration (LC) determination, and at 0, 0.5 and 1% concentrations for lethal time (LT) determination. The number of living and dead nymphs was recorded five days after spraying for LC determination, and daily during six days for LT determination. The LC50 estimated for fourth instar nymphs occurred at 0.56% concentration. For all instars, LC50 and LC95 were estimated at 0.32 and...

Factors affecting the attack rate of Bemisia tabaci on cucumber PAB
Leite,Germano Leão Demolin; Picanço,Marcelo; Guedes,Raul Narciso Carvalho; Ecole,Carvalho Carlos.
The objective of this work was to determine the effects of rainfall, temperature, predators, parasitoids, plant age, leaf chemical composition, levels of leaf nitrogen and potassium, besides density of leaf trichomes, on attack intensity of Bemisia tabaci biotype B on the Cucumis sativus. An increase in the number of whitefly adults and nymphs per leaf was observed with plant aging. A higher number of whitefly adults per leaf and eggs cm-2 was verified in the apical part than in the middle and bottom part of the plants canopy. However, the higher number of whitefly nymphs was observed in the mid-part than in the apical and bottom part of the plant canopy. The incidence of whitefly nymphs was negatively affected with foliar nitrogen. Pentacosane and...

Economic impact analysis of marker-assisted breeding for resistance to pests and post harvest deterioration in cassava AgEcon
Rudi, Nderim; Norton, George W.; Alwang, Jeffrey Roger; Asumugha, Godwin N..
Marker-assisted breeding could have a major impact in relieving productivity constraints that cannot as easily or rapidly be relieved by conventional breeding alone. This paper estimates the benefits of using marker-assisted breeding, as compared to conventional breeding alone, in developing cassava varieties resistant to cassava mosaic disease, green mite, whitefly and post-harvest physiological deterioration in Nigeria, Ghana and Uganda. Marker-assisted breeding is estimated to save at least four years in the breeding cycle for varieties resistant to the pests and to result in incremental net benefits over 25 years in the range of $34 to $800 million depending on the country, the particular constraint and various assumptions. Benefits may reach as high...
