In vivo assessment of possible probiotic properties of Zymomonas mobilis in a Wistar rat model Electron. J. Biotechnol.

In recent years the incorporation of probiotic bacteria into foods has received increasing scientific interest for health promotion and disease prevention. The safety and probiotic properties of Zymomonas mobilis CP4 (UFPEDA-202) was studied in a Wistar rat model fed the 10(9) colony forming units (cfu)/mL-1 of the assayed strain for 30 days. No abnormal clinical signs were noted in the group receiving viable cells of Z. mobilis and water (control) during the period of the experiment. There were no significant difference (p > 0.05) in feed intake and weight gain among mice fed the Z. mobilis in comparison to the control group. Background: Zymomonas mobilis, as a novel platform for bio-ethanol production, has been attracted more attention and it is very important to construct vectors for the efficient expression of foreign genes in this bacterium. Results: Three shuttle vectors ( pSUZM 1, pSUZM2 and pSUZM3 ) were first constructed with the origins of replication from the chromosome and two native plasmids (pZZM401 and pZZM402) of Z. mobilis ZM4, respectively. Background The major challenges associated with the fermentation of lignocellulosic hydrolysates are the reduction in the operating cost and minimizing the complexity of the process. Zymomonas mobilis biofilm has been emerged to resolve these complexities. Biofilm has been reported to tolerate to the toxic inhibitors and easily manipulated toward the cell recycle through the cell immobilization. Results Z. mobilis ZM4 and TISTR 551 were able to develop biofilms on DEAE cellulose under the differences in the morphologies. This study aimed to assess the use of mesquite pods hydrated mash as biomass for the growth of Saccharomyces cerevisiae UFEPEDA-1012 and Zymomonas mobilis UFEPEDA-205 and for ethanol production using a submerged fermentation. A 2³ factorial design was used to analyze the effects of the type of microorganism, time of fermentation and condition of cultivation on the ethanol production in mesquite pods mash (30 g 100 mL-1). From the obtained results the hydrated mesquite pods mash presented as a good substrate for the growth of S. cerevisiae and Z. mobilis in comparison to the standard media. Influence of high osmotic pressure on sorbitol production by Zymomonas mobilis BABT

The objective of the present work was to study the variation on the sorbitol production in relation to the concentration of sugars, (metabolizable or not) and the cultivation time. A full factorial design was used considering the factors such as sucrose and maltose concentration and cultivation time. The addition of sugars caused increases on the sorbitol production up to the concentration of 300g/L however, decreases on the sorbitol production were observed when the concentration reached values above this. Increasing the time of fermentation was statistically significant to sorbitol production, however, little increase the production was noticed after 36h.

Nutritional requirements of Zymomonas mobilis CCT 4494 for levan production BABT

The aim of this work was to study the nutritional requirements of Zymomonas mobilis CCT 4494 for levan production in three chemically defined media. During the optimization of the fermentative process for the production of the exopolysaccharide, different concentrations of glucose, fructose and sucrose as carbon source and yeast extract as vitamin source were tested. Variations of incubation temperature and initial pH of the medium were observed. The results showed that medium containing 20.0% sucrose and 0.5% yeast extract, with initial pH of 7.0, incubated at 30°C gave a 43.0% yield of the biopolymer.

Sugar cane juice fermentation by Zymomonas mobilis CP4 subjected to inhibition by ethanol and high initial concentration of substrate BABT

Sugar cane juice fermentation by Z. mobilis CP4 subject to stress by ethanol and high concentration of sucrose was investigated. Supplementation with saponifiable portion of soybean oil deodourising destillate (SOD) at 5; 10 and 15 mL/L.The ethanol production resulted values of 15.91; 16.99 and 15.63 g/L respectively. Values of Yp/s 0.35; 0.36 and 0.37 were achieved, which represented 20.69; 24.14 and 27.59% respectively higher when unsupplemented medium fermentation was carried out.

Optimization of levan production by Zymomonas mobilis BABT

Effect of different fermentation conditions on levan production by Zymomonas mobilis B-4286 was studied. Levan production increased from 5.7-g/l to 12.6-g/l with an increase in initial sucrose concentration (50-150 g/l). Above 15% (20 and 25%) sucrose concentration, there was no increase in the biomass. The sucrose hydrolysis and levan production occurred even in the absence of significant growth of cells. Maximum amount of levan was produced (14.5 g/l) at pH 5 and 15 g /l at 25(0)C temperature. At temperature between 35(0)C and 40(0)C, levan production was not detected. Presence of glucose in the medium considerably reduced levan production (2.8 g/l) than fructose 6.7g/l.

Differential expression of Zymomonas mobilis sucrase genes (sacB and sacC) in Escherichia coli and sucrase mutants of Zymomonas mobilis BABT

The sacB and sacC genes encoding levansucrase and extracellular sucrase respectively were independently subcloned in pBluescript (high copy number) and in Z. mobilis-E. coli shuttle vector, pZA22 (low copy number). The expression of these genes were compared under identical background of E. coli and Z. mobilis host. The level of sacB gene expression in E. coli was almost ten fold less than the expression of sacC gene, irrespective of the growth medium or the host strain. In Z. mobilis the expression of sacB and sacC genes was shown to be subject to carbon source dependent regulation. Influence of ultrasound on sorbitol release by Zymomonas mobilis grown on high sucrose concentration BABT

This work investigated the effect of applying low intensity ultrasound on sorbitol release by Z.mobilis cultures grown on 200 g/L sucrose medium up to 48 h. The best sorbitol production was 36.09 g/L in 36 h culture. Ultrasound irradiation did not alter the sorbitol values detected after disrupting the cells with 20 minutes treatment.

Activity of glucose-fructose oxidoreductase in fresh and permeabilised cells of Zymomonas mobilis grown in different glucose concentrations BJM

Previously grown cells of the ethanologenic bacterium Zymomonas mobilis produce sorbitol and gluconic acid, in reactions catalysed by the periplasmic enzymes glucose-fructose oxidoreductase (GFOR) and glucono-delta-lactonase (GL). The GFOR/GL system activity, in cells to be used in this bioconversion, depends on growth conditions. In batch runs, with initial glucose concentrations (S0) from 42 to 230 g/L, the highest specific and total GFOR/GL activities were obtained with S0 = 153 g/L (12.6 U/g cells and 62 U/L). Higher S0 led to decreasing activities in fresh cells. With S0 = 209 g/L, the final specific activity was only 7.0 U/g. After disruption of cells, however, an activity over 15 U/g was revealed. Synthesis of sorbitol by Zymomonas mobilis under high osmotic pressure BJM

The bacterium Zymomonas mobilis presents potential for sorbitol production when grown in culture medium with high sugar concentration. Sorbitol is produced and accumulated in the periplasma of the bacterium to protect the cells from the harmful effects of high osmotic pressure that results from the action of invertase on sucrose. The conversion of sucrose into glucose and fructose increases the osmolarity of the medium. However, an excessive increase in the osmotic pressure may decrease the sorbitol production. In this work Saccharomyces cerevisiae invertase was added two media containing sucrose 200 and 300 g.L-1. Ethanolic fermentation of sucrose, sugarcane juice and molasses by Escherichia coli strain ko11 and Klebsiella oxytoca strain P2 BJM

Escherichia coli KO11 and Klebsiella oxytoca P2 recombinants fermented sucrose to ethanol. In minimal medium with 2% or 12% added sucrose KO11 produced 75% and 41%, respectively, of the maximum theoretical yield (0.54g ethanol/g sucrose). In Luria-Bertani (LB) broth with up to 8% sucrose, KO11 presented a 94-96% yield and with 12% sucrose, KO11 presented about 69% yield (44.5g ethanol/L). P2 presented 55% of the theoretical maximum yield in minimal medium supplemented with 2% sucrose and 47% of the maximum in 12% sucrose. Application of fractional factorial design to levan production by Zymomonas mobilis BJM

Levan is a non-toxic, biologically active, extra cellular polysaccharide composed solely by fructose units. Optimization of levan production by Zymomonas mobilis strain ZAG-12 employing a 2(4-1) fractional factorial design was performed to analyze the influence of the temperature (20, 25 e 30ºC) agitation (50, 75 e 100 rpm), and the initial concentrations of both sucrose (150, 200 e 250 g.L-1) and yeast extract (2.0, 3.5 e 5.0g.L-1) on final levan concentration.
