Sequences potentially associated with coffee resistance to diseases were identified by in silico analyses using the database of the Brazilian Coffee Genome Project (BCGP). Keywords corresponding to plant resistance mechanisms to pathogens identified in the literature were used as baits for data mining. Expressed sequence tags (ESTs) related to each of these keywords were identified with tools available in the BCGP bioinformatics platform. A total of 11,300 ESTs were mined. Coffee beans contain two types of alkaloids, caffeine and trigonelline, as major components. This review describes the distribution and metabolism of these compounds. Caffeine is synthesised from xanthosine derived from purine nucleotides. The major biosynthetic route is xanthosine -> 7-methylxanthosine -> 7-methylxanthine -> theobromine -> caffeine. Degradation activity of caffeine in coffee plants is very low, but catabolism of theophylline is always present. Theophylline is converted to xanthine, and then enters the conventional purine degradation pathway. It has been reported that Coffea arabica seeds contain as the main reserve protein, a legumin-like protein, constituted of two subunits, alpha and beta, of approximately 35 and 20 kDa. In this work the seed proteins of several coffee species and varieties were investigated by SDS-PAGE and gel filtration. No differences were observed in the electrophoretic profiles among varieties of C. arabica, however, marked differences were observed among species, or even among individuals of some species. In general, the molecular weight of the subunits alpha and beta accounted for a monomer of 48 to 62 kDa. A propagação do cafeeiro é feita de duas formas: sexuada, que se baseia no uso de sementes, e assexuada, baseada no uso de estruturas vegetativas das plantas, a exemplo do uso das estacas, ou pedaços de seus ramos. No Acre, o processo usual de propagação do cafeeiro, tanto da espécie arabica como canephora, é por semente. Embora a Embrapa já tenha difundido a produção vegetativa por meio de estaca para o café Conilon, da espécie canephora, ainda são poucos os produtores que utilizam esta técnica. Possible presence of vascular bundle sheath cells and its relation to photosynthetic pathway, leaf anatomy and carbon isotope composition (delta13C) were examined in six species of genus Coffea: C. arabica (cvs. Catuaí Vermelho, Mundo Novo, Bourbon Vermelho and Icatu Amarelo), C. canephora (cvs. Apoatã and Guarini), C. liberica, C. dewevrei, C. salvatrix and C. stenophylla. In all genotypes, the vascular bundle was surrounded by a layer of cells with numerous chloroplasts in a centrifugal position. Visually no differences could be seen between the spongy parenchyma cells and the bundle sheath cells, neither in size nor in chloroplast number. Em trabalho efetuado visando o levantamento de inimigos naturais da broca-do-café (Hypothenemus hampei, Ferrari) no estado de Rondônia, verificou-se a ocorrência do fungo Beauveria bassiana (Bals.) Vuill. conforme identificação da Embrapa Rondônia. Os frutos de café foram coletados em cafezais da cultivar Conilon (Coffea canephora), nos municípios de Rolim de Moura, Ouro Preto do Oeste e Machadinho do Oeste, Rondônia, nos meses de fevereiro a março de 2000. A percentagem média de infecção de B. bassiana em frutos broqueados variou de 0,23% a 0,47%, sendo menor que em outras regiões do Brasil. |

This study aimed the identification of cultivars and/or lines of Coffea arabica of commercial interest, using PCR-RAPD markers. The DNA of ground seeds lots of 12 cultivars and/or lines were evaluated with five primers (Operon OPA 01, OPA 04, OPG 11, OPY 16, and OPX 09) were obtained from a selection of 56 primers. The electrophoretic profiles allowed distinction among eight cultivars and/or lines as well as heterogeneity between and within lots of IAPAR59. |

The objective of the study was to evaluate the resistance to rust in coffee cultivars developed by research institutes of Brazil in Paraná state. Resistance to the local leaf rust races was assessed in high disease intensity field conditions at Londrina and Congonhinhas in 2009 and 2010.The cultivars were developed by the EPAMIG/UFV, IAPAR, IAC and MAPA/Procafé. In coffee, bioreactors are the most promising way for scaling-up micropropagation processes, particularly somatic embryogenesis. The availability of an efficient somatic embryogenesis process would allow the rapid mass production of heterozygous materials such as selected Coffea canephora clones and F1 Arabica hybrid varieties. Phenolic compounds are secondary metabolites generally involved in plant adaptation to environmental stress conditions. Chlorogenic acids (CGA) and related compounds are the main components of the phenolic fraction of green coffee beans, reaching levels up to 14 % (dry matter basis). These compounds have a number of beneficial health properties related to their potent antioxidant activity as well as hepatoprotective, hypoglycemic and antiviral activities. Conduziu-se este trabalho com o objetivo de quantificar e caracterizar a composição química de cafés arábica e robusta de safras diferentes e seus efeitos na qualidade do café torrado. Utilizaram-se grãos de café arábica (Coffea arabica L.) da safra 88/89 e safra 2000, proveniente da região sul de Minas gerais, e o café conilon (Coffea canephora Pierre) safra 2000, proveniente do Estado do Espírito Santo. Preparou-se um "blend" na proporção 70% arábica e 30% conilon.
