Xylanase and β-xylosidase from Penicillium janczewskii: Purification, characterization and hydrolysis of substrates Electron. J. Biotechnol.

Background: Xylanases and β-D-xylosidases are the most important enzymes responsible for the degradation of xylan, the second main constituent of plant cell walls. Results: In this study, the main extracellular xylanase (XYL I) and p-xylosidase (BXYL I) from the fungus Penicillium janczewskii were purified, characterized and applied for the hydrolysis of different substrates. Their molecular weights under denaturing and non-denaturing conditions were, respectively, 30.4 and 23.6 kDa for XYL I, and 100 and 200 kDa for BXYL I, indicating that the latter is homodimeric. XYL I is highly glycosylated (78%) with optimal activity in pH 6.0 at 65°C, while BXYL I presented lower sugar content (10.5%) and optimal activity in pH 5.0 at 75°C. The half-lives...

Hemicellulosic agricultural by-products such as corn stover (CS) are highly available materials which represent an opportunity to develop value added products. Native Aspergillus niger GS1 was used for solid-state fermentation (SSF) on alkali pre-treated CS (ACS) aimed to optimize xylanolytic enzymes production, and their effect on in vitro ruminal and true digestibility of ACS. In conifers and diffuse-porous angiosperms, tracheary elements have an important role in structural support and water transport. In these plants it is expected that the conflict between the mechanic and hydraulic function will result in less efficient water transport at the whole plant level than the expected for plants whose conduits do not fulfil a mayor structural role (vines and ring-porous trees). Background: Xylitol is a five carbons polyol with promising medical applications. It can be obtained from chemical D-xylose reduction or by microbial fermentation of Sugarcane Bagasse Hemicellulosic Hydrolysate. For this last process, some microbial inhibitors, as furfural, constitute severe bottleneck. In this case, the use of strains able to produce xylitol simultaneously to furfural neutralization is an interesting alternative. A wild-type strain of Geotrichum sp. was detected with this ability, and its performance in xylitol production and furfural consumption was evaluated. Y chromosomal STRs show sufficient variability among individduals in a population and a high degree of geographical differentiation, such that their polymorphic character makes them especially suited for population genetic studies. To investígate the polymorphism of a set of 17 Y-STR loci in northern China, we genotyped the 17 Y chromosomal STR loci in a population sample of 377 unrelated males from eight ethnic populations in northern China. Lipases are placed only after proteases and carbohydrases in world enzyme market and share about 5% of enzyme market. They occur in plants, animals and microorganisms and are accordingly classified as plant, animal and microbial lipases. Wherever they exist, they function to catalyze hydrolysis of triglycerides to glycerol and fatty acid. Like carbohydrases and proteases, lipases of microbial origin enjoy greater industrial importance as they are more stable (compared to plant and animal lipases) and can be obtained in bulk at low cost. In the present study we characterized the capacity of zinc to protect lipids and proteins from Fe2+-initiated oxidative damage. The effects of zinc on lipid oxidation were investigated in liposomes composed of brain phosphatidylcholine (PC) and phosphatidylserine (PS) at a molar relationship of 60:40 (PC:PS, 60:40). Lipid oxidation was evaluated as the oxidation of cis-parinaric acid or as the formation of 2-thiobarbituric acid-reactive substances (TBARS). Zinc protected liposomes from Fe2+ (2.5-50 muM)-supported lipid oxidation. The cisternae of the Golgi apparatus of dog epididymal principal cells were labeled by the zinc iodide-osmium tetroxide method (ZIO). These cisternae were observed in the supranuclear region of the cytoplasm of the epididymal principal cells. Abundant endoplasmic reticulum cisternae, multivesicular bodies, mitochondria, lysosomes and vesicular elements of variable size were also found in this region, all associated with the sacks of the well-developed Golgi apparatus. The use of the ZIO method facilitates the observation and identification of the cisternae of the Golgi apparatus, thus permitting a correlation between structure and function in the so-called Golgi area. La diversidad de macroinvertebrados marinos ha recibido una atención creciente, no obstante, con un escaso tratamiento en el contexto biogeográfico. Este estudio analiza los registros de 1.601 especies de macroinvertebrados bentónicos pertenecientes a: Demospongiae, Anthozoa, Polychaeta, Mollusca, Crustacea, Echinodermata y Ascideacea, agrupados en 10 zonas y tratados desde una perspectiva zoogeográfica. Zooplankton interactions with the oxygen minimum zone in the eastern south pacific Gayana

Zooplankton in the coastal upwelling zone off the eastern South Pacific must cope with a shallow oxygen minimum zone. In this work, I summarize available information on the zooplankton species that interact with the OMZ in the upwelling region off northern Chile. Data are available from the MinOx cruise carried out in March 2000 and the ZOMEI cruise perfomed in October 2005, both onboard the R/V Vidal Gormaz of the Chilean navy. Nineteen species of copepods and 10 of euphausiids were analyzed in their vertical distribution and migrating behavior.
