Action Indice Composition. Livrable n° A III. Rapport final, 23 septembre 2014 ArchiMer

The European Union Water Framework Directive (WFD) refers to phytoplankton as one of the biological quality elements that should be regularly monitored and evaluated thanks to three ecological indices: biomass, abundance and composition. The first two have already been defined, but the third one has not yet been proposed in the English Channel-Atlantic zone. The construction of a composition indice must consider the diversity of all the phytoplankton. As the microscopic counts concern only the micro-phytoplankton, it is necessary to use additional information which concerns the nano- and pico-phytoplankton, of great importance when estimating the phytoplankton diversity. These components of the phytoplankton can be measured by different complementary...

Action 3. Indice Composition. Livrable n° A II. Rapport final, 13 novembre 2014. ArchiMer

In this paper we review the technologies available to make globally quantitative observations of particles in general—and plankton in particular—in the world oceans, and for sizes varying from sub-microns to centimeters. Some of these technologies have been available for years while others have only recently emerged. Use of these technologies is critical to improve understanding of the processes that control abundances, distributions and composition of plankton, provide data necessary to constrain and improve ecosystem and biogeochemical models, and forecast changes in marine ecosystems in light of climate change. In this paper we begin by providing the motivation for plankton observations, quantification and diversity qualification on a global scale. We...

High spatial variability of phytoplankton assessed by flow cytometry, in a dynamic productive coastal area, in spring:the eastern English Channel ArchiMer
Bonato, Simon; Christaki, Urania; Lefebvre, Alain; Lizon, Fabrice; Thyssen, Melilotus; Artigas, Luis Felipe.
The distribution of phytoplankton (from pico-to microphytoplankton) was investigated, at single-cell level and at high spatial resolution, during an oceanographic cruise across the eastern English Channel (EEC) between April 27 and 29, 2012. Seawater was continuously collected from surface waters and analysed on board at high frequency (one sample every 10 min), by using a new generation of pulse-shape recording scanning flow cytometer (CytoSense, Cytobuoy©). A Bray-Curtis matrix analysis based on phytoplankton composition allowed the discrimination of 4 communities. Within these communities, abundance, cell size as well as single cell and total red fluorescence of 8 phytoplankton groups were measured. Picoeukaryotes and Synechococcus spp. cells dominated...

From 1999 to 2005, studies carried out in the frame of regional and national French programs aimed to determine whether the Phaeocystis globosa bloom affected the intertidal benthic communities of the French coast of the eastern English Channel in terms of composition and/or functioning. Study sites were chosen to cover most of the typical shore types encountered on this coast (a rocky shore, an exposed sandy beach and a small estuary). Both the presence of active Phaeocystis cells and their degradation product (foam) did have a significant impact on the studied shores. The primary production and growth rates of the kelp Saccharina latissima decreased during the bloom because of a shortage of fight and nutrient for the macroalgae. On sandy sediments, the...

Puillat Felix, Ingrid; Karlson, Bengt; Artigas, Luis Felipe; Grémare, Antoine; Nizzetto, Luca; Rubio, Anna; Laakso, Lauri; Seppala, Jukka; Mourre, Baptiste.
The WP4 of the JERICO-Next is a synthesis of the project built upon activities in other WPs, gathering the consortium around applied Joint Research Activity Projects (JRAPs) selected to put forward the added value of JERICO-NEXT. In order to reach this objective, WP4 keeps on track the JERICO-NEXT course of actions via synthesis and application activities based on interactions with other WPs. WP4 helps establishing some topical approaches for the scientific strategy in WP1 (task 1.2) and will give inputs to establish the network strategy after JERICO-NEXT (task 1.6). Indeed, six JRAPs are being implemented to address different key environmental questions and/or policy requirements such as those considered by the MSFD, and according to the 6 JERICO...

Redefinition of the Dinoflagellate Genus Alexandrium Based on Centrodinium : Reinstatement of Gessnerium and Protogonyaulax , and Episemicolon gen. nov. (Gonyaulacales, Dinophyceae) ArchiMer
Gómez, Fernando; Artigas, Luis Felipe.
The genus Centrodinium contains oceanic and predominantly tropical species that have received little attention. Three species of Centrodinium were examined using thecal plate dissociation, scanning electron microscopy, and molecular sequences. The apical horn of Centrodinium intermedium and C. eminens is formed by the elongation of the fourth apical plate, and a second apical split into two plates. In C. punctatum two apical plates (2′ and 4′) almost completely encircle the apical pore plate (Po), while the contact with the plate 1′ in the ventral side is much reduced, and the plate 3′ does not reach the Po. Moreover, its left posterior lateral sulcal plate is longer than its right pair, while reversed in the typical Centrodinium spp. The sulcal posterior...

Action 9. FlowCam ZooPhytoImage. Livrable n° 4. Rapport final, 23 septembre 2014 ArchiMer
Ali, Nour; Wacquet, Guillaume; Didry, Morgane; Hamad, Denis; Artigas, Luis Felipe; Grosjean, Philippe.
The goal of this study is to investigate about the possibility of coupling measurements made by image analysis from the FlowCAM with Zoo/PhytoImage with data obtained with a flux cytometer (pulse-shape-recording Scanning Flow Cytometry) on the same samples gathered in current monitoring networks in the eastern Channel and southern North Sea. In this preliminary study, we collected a series of samples off Boulogne-sur-Mer (SRN-REPHY monitoring system run by IFREMER) and along a transect in the Baie St-Jean (Wimereux-Slack) run by LOG. All these samples were digitized with a FlowCAM and measured with a scanning flow cytometer (CytoSense). The complete analysis with the FlowCAM and Zoo/PhytoImage is detailed in the present report. In order to get a better...

Nouvelles perspectives ArchiMer
Wacquet, Guillaume; Grosjean, Philippe; Colas, Florent; Hamad, Denis; Artigas, Luis Felipe.
The coupled system FlowCAM/ZooPhytoImage has become a real operational tool in 2014. However, to be fully adapted to the observations of phytoplankton performed in the context of the REPHY observation network and in order to better respond to present and future requests concerning the evaluation of quality of coastal and marine waters within the European requirements, such as the WFD and MSFD, new functionalities must be integrated into existing tools. Therefore, different axis of development have been proposed by UMONS and Ifremer to adapt both the digitization device and the Zoo/PhytoImage software to the constraints defined by the REPHY. First, version 5 of Zoo/PhytoImage contains recent innovations such as the development of routines to automatically...
