Burgeot, Thierry; Gagnaire, Beatrice; Renault, Tristan; Haure, Joel; Moraga, D.; David, E.; Boutet, L.; Sauriau, P.g.; Malet, Nathalie; Bouchet, V.; Lapegue, Sylvie; Bouilly, Karine; Le Moullac, Gilles; Arzul, Genevieve; Knoery, Joel; Quiniou, Francoise; Bacher, Cedric; Soletchnik, Patrick.

Chromosomal organization of simple sequence repeats in the Pacific oyster (Crassostrea gigas): (GGAT)(4), (GT)(7) and (TA)(10) chromosome patterns ArchiMer
Bouilly, Karine; Chaves, R.; Leitao, A.; Benabdelmouna, Abdellah; Guedes Pinto, H..
Chromosome identification is essential in oyster genomic research. Fluorescence in situ hybridization (FISH) offers new opportunities for the identification of oyster chromosomes. It has been used to locate satellite DNAs, telomeres or ribosomal DNA sequences. However, regarding chromosome identification, no study has been conducted with simple sequence repeats (SSRs). FISH was used to probe the physical organization of three particular SSRs, (GGAT)(4), (GT)(7) and (TA)(10) onto metaphase chromosomes of the Pacific oyster, Crassostrea gigas. Hybridization signals were observed in all the SSR probes, but the distribution and intensity of signals varied according to the oligonucleotide repeat. The intercalary, centromeric and telomeric bands were observed...

Effects of cadmium on aneuploidy and hemocyte parameters in the Pacific oyster, Crassostrea gigas ArchiMer
Bouilly, Karine; Gagnaire, Beatrice; Bonnard, Marc; Thomas-guyon, Helene; Renault, Tristan; Miramand, Pierre; Lapegue, Sylvie.
Pacific oysters, Crassostrea gigas, are commonly reared in estuaries where they are exposed to anthropogenic pollution. Much research has been made on the toxicity of cadmium to aquatic organisms because the compound recurrently contaminates their environment. Our study examined the influence of cadmium on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in C. gigas at different stages of life. Adults and juveniles were exposed to two different concentrations of cadmium. The first concentration applied was equivalent to a peak value found in Marennes-Oleron bay (Charente-Maritime, France; 50 ng L-1) and the second was 10 times higher (500 ng L-1). Exposure to 50 ng L-1 cadmium caused a significant...

Endonuclease banding reveals that atrazine-induced aneuploidy resembles spontaneous chromosome loss in Crassostrea gigas ArchiMer
Bouilly, Karine; Leitao, Alexandra; Chaves, Raquel; Guedes Pinto, Henrique; Boudry, Pierre; Lapegue, Sylvie.
Aneuploidy has previously been observed in the Pacific oyster, Crassostrea gigas, and shown to be negatively correlated with growth. Moreover, a significant impact of atrazine exposure has been described in C. gigas, and persistence of that effect has been observed between generations. Evidence of differential chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique. Pairs 1, 5, 9, and 10 are characterized by the loss of 1 chromosome. As restriction enzyme (RE) digestion chromosome banding allows a better identification of chromosome pairs, we used this technique to identify which chromosomes are affected when aneuploidy is increased by exposure to atrazine. The progeny of oysters contaminated by atrazine were...

Convention 2004 ArchiMer
Bouilly, Karine; Bonnard, Marc; Lapegue, Sylvie.

Bouilly, Karine.
The impact of environmental factors on aneuploidy level in the oyster Crassostrea gigas was studied with experiments in controlled conditions which exposed adult and juvenile oysters to different pollutants (atrazine, a herbicide, and cadmium, a heavy metal) frequently found in Marennes-Oléron Bay. A positive correlation between atrazine concentration and aneuploidy in C. gigas was observed. Moreover, the impact of atrazine on aneuploidy level persisted in time both within and between generations, demonstrating the influence of an environmental pollution on this phenomenon. On the other hand, the aneuploidy level of C. gigas was not influenced at the cadmium concentrations tested. Significant differences in aneuploidy levels were observed in oysters...

In the Pacific oyster, Crassostrea gigas hypodiploid aneuploid cells with 2n=19, 18 or 17 chromosomes have regularly been reported. There is a negative correlation between somatic aneuploidy and growth rate in oysters (Leitão et al., 2001a) as well as evidence for a genetic basis (Leitão et al., 2001b). Furthermore, non-random chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique (Leitão et al., 2001c). This poster summarises different studies used to investigate the effects of the herbicide atrazine on the level of aneuploidy in this species

Impact of diuron on aneuploidy and hemocyte parameters in Pacific oyster, Crassostrea gigas ArchiMer
Bouilly, Karine; Bonnard, Marc; Gagnaire, Beatrice; Renault, Tristan; Lapegue, Sylvie.
Diuron is a substituted urea herbicide used for agricultural and nonagricultural weed control. Its widespread use and relatively slow breakdown led us to analyze its influence on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in Pacific oysters, Crassostrea gigas. Adult oysters were subjected to two diuron concentrations (300 ng L-1 and 3 mu g L-1) for 11 weeks. Significantly higher aneuploidy level was observed in diuron-treated oysters compared with the control. Furthermore, the observed impact on aneuploidy persisted to the next generation as offspring exhibited significantly higher aneuploidy levels when their parents had been exposed to diuron. Significant increases in hemocyte parameters (cell...

Aneuploidy is the alteration of the normal diploid chromosome number. In the Pacifie oyster, Crassastrea gigas, hypodiploid cells have regularly been reported as have a negative correlation between this phenomenon and growth and evidence for a genetic basis. We previously demonstrated a positive relationship between a pollutant, atrazine, and aneuploidy in Crassastrea gigas adults and juveniles. To evaluate the persistence of this impact, the present study focused on a sample of the same juveniles previously exposed to different atrazine treatments (0.0 1 mg/l which represents a peak value found in a polluted environment and 0.1 mg/l) for three and a half months. Then, we evaluated them for aneuploidy after another two and a half months in non polluted...

Widespread use of the herbicide atrazine has incited much research on its toxicity in aquatic systems, where it is routinely detected due to runoff from cultivated fields. Moreover, the determination of the genotoxic effect of such pollutants in the marine environment has become a major requirement for ecosystem protection. In the Pacific oyster, Crassostrea gigas, hypodiploid aneuploid cells have regularly been reported. There is a negative correlation between this phenomenon and growth, as well as evidence for a genetic basis.
Bouilly, Karine.
Aneuploidy has previously been reported in wild and cultivated Pacific oysters, Crassostrea gigas, and it has been shown to be negatively correlated with growth. This is especially important since high variability of growth rate is one of the major problems in the aquacultural production of this species. The existence of an environmental factor (such as atrazine contamination) with an effect on aneuploidy level was first investigated through the study of a population of Crassostrea gigas subjected to different concentrations of atrazine. The treatments applied represent peak values found in the environment (0,01 mg/l) plus a value ten times higher (0,1 mg/l). Our results have shown that although atrazine did not have an effect on the mortality of adult...

In the last decades, shellfish farming and in particular ostreiculture have developed in a significant way in France. Marennes-Oleron (Charente-Maritime) basin is the first French producer of oysters. But this area is also subjected to many recurring pollution, brought mainly by the Charente river. The recrudescence using of herbicides in agriculture including atrazine implies the transfer towards the aquatic environment of new pollutants in estuarine areas. Moreover industrial rejections bring many heavy metals in these same estuarine areas. It is appropriate of wondering on harmful effects of all these pollutants on the animals living in these areas, particularly oysters. Bivalve molluscs have been postulated as ideal indicator organisms because of...

Shellfish farming is an important economic activity around the world. This activity often takes place in areas subjected to various recurring pollutions. The recrudescent use of herbicides in agriculture including atrazine implies pollutant transfer towards aquatic environment in estuarine areas. Harmful effects of such substances on animals in marine environment, particularly on cultured bivalves, are poorly documented. Bivalve molluscs such as mussels and oysters have been postulated as ideal indicator organisms because of their way of life. They filter large volumes of seawater and may therefore accumulate and concentrate eontaJ.11inants within their tissues. Moreover, development of techniques allowing effect analysis of such compounds on bivalve...
