Demonstrating the need for chemical exposure characterisation in a microplate test system: Toxicity screening of sixteen pesticides on two marine microalgae ArchiMer

Pesticides used in viticulture create a potential risk for the aquatic environment due to drift during application, runoff and soil leaching. The toxicity of sixteen pesticides and one metabolite were evaluated on the growth of two marine microalgae, Tisochrysis lutea and Skeletonema marinoi, in 96-h exposure assays conducted in microplates. For each substance, concentrations of stock solutions were analytically measured and abiotic assays were performed to evaluate the chemical stability of pesticides in microplates. For two chemicals, microalgae exposures were run simultaneously in microplates and culture flasks to compare EC50 calculated from the two exposure systems. Environmental fate of chlordecone in coastal habitats: recent studies conducted in Guadeloupe and Martinique (Lesser Antilles) ArchiMer

The organochlorine pollution by chlordecone, an insecticide spread in the past in banana plantations, is now recognized as a major ecological, economic, and social crisis in Guadeloupe and Martinique Islands. Due to its physical and chemical properties, this molecule is particularly persistent in the natural environment. Volcanic soil of Guadeloupe and Martinique contain allophanes (amorphous clays), which favor chlordecone trapping due to their structure and physical properties. Evaluating the ecological impact of an oil spill is a complex issue requiring coherently articulated examination of the sequence of interactions that link the cell, where contaminants exert their effects, to the ecosystem, where interactions with human activities arise. This sequence of interactions traverses the frontiers between scientific disciplines (chemistry, toxicology, physiology, and fisheries ecology). Link between exposure of fish (Solea solea) to PAHs and metabolites: Application to the "Erika" oil spill ArchiMer

An analytical method consisting in enzymatic deconjugation, solid phase extraction and purification, and gas chromatography/mass spectrometry analysis after derivatization was used in this study to quantify Polycyclic Aromatic Hydrocarbon (PAH) metabolites in the bile of fish. The method has been applied in a laboratory experiment studying the fate of pyrene in basin containing soles. This study has allowed the identification of 1-hydroxypyrene as the single metabolite in bile after enzymatic deconjugation. In a second time, 1-hydroxypyrene has been used as a biomarker of exposure in the case of the "Erika" oil spill. This biomonitoring was successful in demonstrating the exposure of juvenile soles to PAHs present in the "Erika" fuel oil.

Mise en place d'un indicateur d'évaluation du risque lié à la présence de pesticides en milieu lagunaire méditerranéen ArchiMer

Ce travail a permis de construire et de valider un indicateur d'évaluation du risque lié à la présence de pesticides dans les lagunes méditerranéennes françaises. Cet indicateur, le Quotient de Risque (QR) s'appuie sur une stratégie de suivi globale, adaptée au contexte des lagunes, de leurs fortes dilutions et de leur dynamique complexe, tout en prenant en compte l'effet des mélanges de pesticides dans les eaux. This study screened binary mixtures of pesticides for potential synergistic interaction effects on growth of the marine microalgae Tisochrysis lutea and Skeletonema marinoi. It also examined the single and combined effects of three of the most toxic substances on microalgal physiology. Single substances were first tested on each microalgal species to determine their respective EC50 and concentration-response relationships.
